A reaktív oxigén részecskék, bár citotoxikusak, olyan komplex és finoman szabályozott élettani folyamatokat váltanak ki gombákban, mint az oxidatív stresszválasz, a gombák differenciálódása és programozott sejthalála. A fonalas gomba modellszervezet Aspergillus nidulans antioxidáns védelmi rendszere végrehajtó és szabályozó elemeinek a felderítése és funkcionális jellemzése olyan eszközöket biztosíthat a számunkra, amelyekkel kontrollálhatjuk az iparilag és klinikailag fontos gombák stresszválaszát és sejthalálát. Ez a projekt néhány olyan stresszválasz fehérje funkcióanalízisét tűzi ki célul, melyek sarkalatos szerepet játszhatnak az oxidatív stressznek kitett micéliumokban a mitokondriumok integritásának és működésének a megőrzésében. Egy másik fő célkitűzés az AtfA target génjeinek az azonosítása és funkcionális jellemzése. Az AtfA egy olyan bZIP-típusú transzkripciós faktor, amely levezényli a különféle környezeti stresszhatásokra adott válaszokat. Az AtfA potenciális kölcsönható partnereinek és/vagy helyettesítőinek a felderítése és funkcióanalízise szintén a fő célkitűzéseink között szerepel. A kutatók egy kiterjedt tudományos együttműködési hálózatban tevékenykednek, ami a javaslat újszerűsége ellenére a projekt kockázatát elfogadható szinten tartja.
Summary
Reactive oxygen species, albeit being cytotoxic, elicit complex and delicately regulated physiological processes in fungi like oxidative stress response, fungal differentiation and programmed cell death. Screening for and functional characterization of executive and regulatory elements of the antioxidant defense system of the filamentous model fungus Aspergillus nidulans may provide us with suitable tools to control stress response and cell death in industrially and clinically important fungi. This project aims at the functional analysis of some stress response proteins, which can play a pivotal role in the preservation of the integrity and function of mitochondria in oxidative stress exposed mycelia. Another major goal is the identification and functional characterization of AtfA-target genes. AtfA is a bZIP-type transcription factor and orchestrates responses to different types of environmental stress. Screening for and functional analysis of potential interacting partner(s) and/or substitute(s) for AtfA are also among our major goals. The investigators work within a wide-spread scientific collaborative network, which keeps the risk of the project at an acceptable level despite the novelty of the proposal.

A projekt keretében jellemeztük a fonalas gomba modellszervezet Aspergillus nidulans néhány nagy fontosságú oxidatív stresszválasz elemét. A kutatásainkban olyan komplex megközelítési módokat alkalmaztunk, amelyek összehasonlító genomikai, transzkriptomikai, molekuláris genetikai, mikrobiális élettani, bioinformatikai és mikroszkópi módszereket foglaltak magukba. Az elért fő kutatási eredmények a következők: (i) Különbözőféle oxidatív stresszhatásoknak kitett tenyészetekben bizonyos stresszválasz gének transzkripciója hasonlóképpen változott, és ezek a gének alkotják a központi oxidatív stresszválasz („COSR”) gének csoportját. (ii) Megállapítottuk, hogy az élesztőkben leírt központi környezeti stresszválasz („CESR”) A. nidulansban nem volt kimutatható. (iii) Megfigyeltük, hogy a stresszválasz fehérjéket kódoló gének deléciója gyakran nem eredményezett stresszérzékeny fenotípust. (iv) A gomba glutation (GSH) metabolizmusának az enzimei közül a γ-L-glutamil-L-cisztein szintetáz és glutation reduktáz szükségesek voltak a konidiospórák csírázásához. (v) A GSH lebontásában szerepet játszó γ-glutamil transzpeptidáz szintén részt vehet az extracelluláris aminosavak és peptidek γ-glutamilizációjában szénéhezés esetén. (vi) Az A. nidulans fontos mitokondriális fehérjéit kódoló gének deléciója, illetve túltermeltetése általában nem befolyásolta jelentősen a sejtek élettani és öregedési folyamatait. (vii) Létrehoztuk a gomba stressz adatbázist („FSD”).
Results in English
We characterized some highly important stress response elements of the filamentous fungus model organism Aspergillus nidulans in the frame of the project. We employed complex approaches in our research, which included comparative genomic, transcriptomic, molecular genetic, microbial physiological, bioinformatic and microscopic methods. Our major research results are as follows: (i) The transcriptions of certain stress response genes changed similarly in cultures exposed to various types of oxidative stress, and these genes establish the group of core oxidative stress response („COSR”) genes. (ii) We found that the core environmental stress response („CESR”) described in yeasts could not be detected in A. nidulans. (iii) We observed that the deletion of genes encoding stress response proteins often did not result in any stress sensitive phenotype. (iv) Out of the enzymes of the glutathione (GSH) metabolism of the fungus, γ-L-glutamyl-L-cysteine synthetase and glutathione reductase were necessary for the germination of conidiospores. (v) γ-Glutamyl transpeptidase, which plays a role in the degradation of GSH, may also take part in the γ-glutamylization of extracellular amino acids and peptides under carbon starvation. (vi) The deletion or overexpression of genes coding for important mitochondrial proteins in A. nidulans generally did not influence significantly the physiological and ageing processes of the cells. (vii) We set up the fungal stress database („FSD”).
