Növekvő szükséglet mutatkozik olyan új biokompatibilis anyagok iránt, melyek biomimetikus karakterük mellett kis költségű, de nagy hatékonyságú anyagok amelyek kis mellékhatást hatást mutatnak. A polimer gélek várhatóan jól megfelelnek ezeknek a feltételeknek, hiszen képesek ötvözni a kívánt funkcionalitást és szenzor mechanizmust.
Jelen kutatás egyik legfontosabb célkitűzése új biokompatibilis és biodegradábilis makromolekulák és gélek kifejlesztése, melyek jó alapanyagul szolgálhatnak gyógyszer hatóanyagok szállítását, célba juttatását, vagy szövetek regenerálását célzó rendszerek kiépítésére. Hatóanyag szállításra olyan gélek jöhetnek számításba, amelyek gyors és intenzív térfogat növekedéssel reagálnak különböző stimulációkra (pl. hőmérséklet, pH, redoxi potenciál változásra). A jelentős térfogat változás révén képesek ezek a polimerek kontrollált mértékben kiengedni a bezárt aktív anyagokat egy nyitó-záró mechanizmusnak megfelelően. Ha lejárt szabadalmú generikus hatóanyagokat új biokompatibilis gélkompozitokba zárjuk, akkor lehetőség nyílhat újbóli szabadalmaztatásukra, mely jelentős gazdasági haszonnal járhat. Egyes funkcionális biodegradábilis polimerek alkalmasak környezetre káros anyagok hatékony megkötésére is.
A projekt másik tervezett célkitűzése kalcium-szilikát és polimertartalmú hibrid rendszerek szintézise, mely rendszerek bioaktívak, alkalmasak orvos-biológiai felhasználásra. A hibrid rendszerek előállítására alkalmazott technika egy új, kis energia-felhasználású szol-gél módszer, mely különböző katalizátorok és szerves molekulák, biokompatibilis polimerek alkalmazását vizsgálják. A biohibridek legfontosabb jellemzése azon alapul, hogy milyen erős a hajlamuk apatit képzésre. A kutatás főbb célkitűzései szorosan kapcsolódnak a 2006-ban alapított Nanogyógyszerek Európai Technológiai Platformjához.
Summary
There is an increasing need for new smart biocompatible materials in many medical applications, with biomimetic functionalities that combine low cost, high efficiency and minor side effects. There is an increasing need for new smart biocompatible materials in many medical applications, with biomimetic functionalities that combine low cost, high efficiency and minor side effects. Polymer gels are good candidates, since they combine a more efficient actuation or sensory mechanism along with minimum investment on expensive raw materials.
One of the main purposes of the present research is to develop novel biocompatible and biodegradable macromolecules and gels that make possible to prepare promising drug carriers as well as novel tissue engineering scaffolds. The prompt and pronounced alteration of volume of gels by different stimuli (temperature, pH, redox potential etc.) can be used to tune the release of enclosed active ingredients according to an open-close mechanism that are able to accomplish the targeted and controlled delivery of active ingredients. Furthermore smart biodegradable polymer gels can be used for the controlled binding of environment damaging compounds. A special economic advantage can be realised by the proposed drug carrier gels, since the combination of expired patents of generic drug molecules with novel biocompatible drug carriers makes possible to file new patents.
The other aim of this project is to combine hard inorganic (calcium silicate and phosphate) and soft bioactive macromolecules in form of biohybrid systems. The synthesis used for the preparation of hybrid systems is a new low energy-consuming sol-gel method applying various catalysts and organic components. The most important characterization of the biohybrids is the comparison of their apatite-forming ability. The outlined plans are tightly connected to the European Technology Platform of Nanomedicine which was founded in 2006.

A poli(szukcinimid)-et termikus polikondenzációval állítottunk elő L-aszparaginsavból o-foszforsav katalízis mellett. Poli (glicerin-szebacát)-ot szebacinsav és glicerin termikus polikondenzációjával állítottuk elő. Ezeket a polimereket különböző biokompatibilis molekulákkal térhálósítottuk. Vizsgáltuk a gélek mechanikai és duzzadási tulajdonságait. Kidolgoztuk a reaktív elektrosztatikus szálhúzás technikáját, amellyel kémiai kötésekkel térhálósított gélszálakat állítottunk elő. Model hatóanyagok szabályozott leadásához gél diszperziókat, és nano-, mikro-gélszálakat állítottunk elő. A mágneses nanorészecskéket elegyítve a PSI-oldattal speciális szálstruktúrákat sikerült felépíteni. Eljárást dolgoztunk ki dopaminnal módosított PSI és poli(aszparaginsav) gyógyszerkonjugátumok szintetizálására. Meghatároztuk a minták vízbeli oldhatóságát és lipofilitását. Vizsgáltuk a dopaminnal módosított poli(aszparaginsav) enzimatikus degradációját. Biokerámia (foszfortartalmú kalcium-szilikátok) és biokompozitok (Al2O3 részecske erősítésű, cellulózalapú biopolimerek) szol-gél szintézisét sikerült megoldani. A biokerámia csont implantátumok fejeként alkalmazhatók, a biokompozitok már más implantátumként is számba vehetők elasztikus karakterük miatt. Legfontosabb eredményeinkről két angol nyelvű könyvfejezetben és 12 nemzetközi, valamint három magyar nyelvű közleményben számoltunk be. További 5 folyóirat cikkben összegeztük ehhez a témához részben kapcsolódó kutatásainkat.

Poly(succinimide) have been synthesized by solvent free thermal polycondensation of L-aspartic acid in the presence of o-phosphoric acid catalyst. Poly(glycerol-sebacate) have been synthesized by thermal polycondensation of glycerol and sebacic acid. Different biocompatible molecules have been used for crosslinking these polymers. The mechanical and swelling properties of the gels have been investigated. We have developed a reactive electrospinning method for creating chemically crosslinked gel fibers. For controlled drug release of model drugs we have prepared nano and micro gel fibers and gel suspensions. By incorporating magnetic nanoparticles into the PSI solution we were able to prepare a special fiber structure. We have developed a method for preparing dopamine modified PSI and poly(aspartic acid) drugconjugates. The solubility in water and lipofility of sampels have been measured. The enzymatic degradation of dopamine modified poly(succinimide) have been investigated. Sol-gel synthesis of bioceramic (P-containing Ca silicate) and biocomposites (Al2O3 particle reinforced, cellulose-based biopolymers) has successfully been developed. The bioceramic can be utilized as a head of bone implants, the biocomposites may also applied for other implants owing to their elasticity. The important results have been reported in two chapters of books, 12 papers of international journals, and 3 papers of Hungarian journals. The research work has been summarized in further five papers.
