A tervezett kutatások fő célja olyan általános törvényszerűségek és molekulaszerkezet-tulajdonság összefüggések felismerése, melyek elősegítik különféle igényeknek megfelelő fluoreszcenciás jelzőanyagok, fénnyel szabályozható tulajdonságú rendszerek, nanoszerkezetek és gyógyszerhordozók kifejlesztését. Biológiai fontosságú alkaloidok fluoreszcenciás viselkedését és nyomnyi mennyiségű kationok hatását tervezzük vizsgálni, hogy feltárjuk az analítikai alkalmazás lehetőségét és a kettős fluoreszcencia kibocsájtás okát. Különös figyelmet fordítunk kukurbiturilok és szulfonátokalixarének üregében lejátszódó fotoiniciált folyamatok tanulmányozására, mert e nem-mérgező, vízoldható makrociklusokkal szabályozhatóvá válhat a fotokróm és energiavesztési folyamatok kinetikája. Választ adunk arra a kérdésre, hogy a makrociklus mérete és flexibilitása valamint két molekula beékelődése miként befolyásolja a fényelnyelést követő folyamatokat. Ígéretes feltáró jellegű munkák folytatásaként szulfonátokalixarénekkel előidézett aggregációt és szerves nanorészecske képződést vizsgálunk. Fluoreszcenciás jelzőanyagok és alkaloidok segítségével jellemezzük természetes polielektrolitok és francia kutatók által előállított új polimerek pH változtatással elérhető anyagleadási tulajdonságait. A tervezett projekt részben kapcsolódik egy elfogadott CNRS-MTA kétoldalú együttműködési programhoz.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.
A javasolt kutatások annak az alapvető kérdésnek a megválaszolására összpontosulnak, hogy elektrolitok és makrociklusokba kötődés miként változtatja a gerjesztett molekulák sajátságait. Szisztematikus vizsgálatokkal feltárjuk kationok hatását biológiai fontosságú, ismeretlen fotofizikai sajátságokkal rendelkező alkaloidok fluoreszcenciás viselkedésére. A kukurbiturilok és szulfonátokalixarének iránt tapasztalható egyre nagyobb nemzetközi érdeklődés inspirált bennünket arra, hogy tanulmányozzuk, miként befolyásolja e vízoldható, biokompatibilis makrociklusokba ékelődés a gerjesztett állapotból kiinduló folyamatok sebességét. Széleskörű kutatásokat tervezünk annak a megismerésére, hogy makrociklusos vegyületek üregében miként módosul a fotokróm vegyületek viselkedése. Feltárjuk a makrociklus elektrosztatikus töltésének, méretének, flexibilitásának és redox potenciáljának a hatását a fotokróm átalakulások kinetikájára és a fotokróm anyagok stabilitására. A szulfonátokalix[8]arénnel előidézett szerves nanorészecske képződésre vonatkozó megfigyeléseink szolgálnak kiinduló pontul azokhoz a munkákhoz, melyek célja a makrociklusokkal elősegített aggregáció molekulaszerkezeti feltételeinek megismerése. Ezek az eredmények hozzájárulhatnak a gyakran alkalmazott H- és J-aggregátumok fluoreszcenciás sajátságainak változtatásához. Tanulmányozni kívánjuk azt is, hogy gyenge sav szubsztituenst tartalmazó polielektrolitok miként módosítják alkaloidok és jelzőanyagok fluoreszcenciáját.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Növekvő igény mutatkozik a mikrokörnyezet változását érzékelni képes jelzőanyagok és fénnyel szabályozható tulajdonságú önszerveződő rendszerek iránt. Az ilyen gyakorlati igények kielégítése érdekében szükség van a gerjesztett molekulák energiavesztési folyamatainak sebességét meghatározó tényezők részletes ismeretére. E projekttel jelentősen hozzájárulhatunk annak a jobb megértéséhez, hogy a mikrokörnyezet sajátságainak változtatása miként befolyásolja a fény hatására végbemenő folyamatokat. Eredményeink elősegítik az alkaloidok fluorescenciás jelzőanyagként történő alkalmazását és új távlatokat nyitnak a fotokróm vegyületek vizes közegű alkalmazása terén. A spiro forma elhanyagolható vízoldékonysága és a merocianin tautomer gyors bomlása megakadályozta a spiropirán típusú fotokróm anyagok felhasználását vizes közegben. Beékelődési komplex képzéssel ezek a problémák megoldhatók, és lehetőség nyílik a két forma közötti átmenet sebességének változtatására is. Alkaloidok makrociklusokhoz illetve polielektrolitokhoz kötődésének vizsgálata során kapott eredményeink jól hasznosíthatók új gyógyszerhordozó anyagok kifejlesztéséhez. A beékelődési komplexek tanulmányozásakor szerzett ismeretek alapjául szolgálnak új nanotechnológiai alkalmazások, szenzorok, analitikai módszerek és sztereoszelektív fotokémiai reakciók tervezéséhez. Lényegesen kedvezőbb komplexképzési sajátságaik miatt a kukurbiturilok sok területen előnyösebben használhatók, mint a ciklodextrinek. Kukurbit[8]urilnak az a képessége, hogy két különböző molekulát is üregébe tud fogadni lehetővé teszi felhasználását különféle önszerveződött szerkezetekben és szupramolekuláris polimerekben. Így a terner kukurbit[8]uril komplexek tanulmányozásakor elért eredmények gyakorlati szempontból is fontosak.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.
Mivel a fény a mindennapi életünk egyik legfontosabb kísérője, alapvető fontosságú, hogy megértsük a fényelnyelődést követő folyamatokat. Az anyagok megvilágításának hatására fluoreszcencia kibocsájtás illetve kémiai változások mehetnek végbe. Célunk annak a megismerése, hogy ezek a fénnyel előidézett folyamatok miként befolyásolhatók fémsók, makrociklusos vegyületek vagy polimerek segítségével. A fluoreszcenciás sajátságok változása alapján következtetünk molekulák illetve ionok közötti kölcsönhatásra. Gyógyászati szempontból fontos alkaloidok fluoreszcenciája alkalmas lehet nyomnyi mennyiségű fémionok kimutatására. Jelentős figyelmet fordítunk szerves molekulák makrociklusokba ékelődésének tanulmányozására. Az ilyen gazdavegyületek védelmet nyújthatnak nemkívánatos kémiai átalakulások ellen. Két molekulának a makrociklusba ékelődését felhasználjuk a fluoreszcenciás tulajdonságok változtatására, illetve a fotokémiai reakciók sebességének és szelektivitásának módosítására. Fény hatására színt és molekulaszerkezetet változtató fotokróm vegyületek különösen fontosak fénnyel szabályozható tulajdonságú anyagok, sötétedésre képes szemüveg-lencsék tervezéséhez és információ tárolási feladatok megvalósításához. A fotokróm vegyületeket makrociklusokba tervezzük beépíteni, hogy szabályozzuk a színváltozást okozó folyamatok sebességét és növeljük az oldhatóságot továbbá a kémiai stabilitást vízben. Tanulmányozni fogjuk polimerek és alkaloidok kölcsönhatását is. E téren elért eredmények hozzájárulnak a biológiailag aktív komponens szabályozott felszabadulását biztosító új hordozóanyagok kifejlesztéséhez.

The main goal of the project is to find general principles and structure-property relationships which facilitate the rational design of new tailor-made fluorescent probes, photoresponsive systems, nanoobjects and drug delivery vehicles. The fluorescent behaviour of pharmaceutically important alkaloids will be examined and the effect of the addition of trace amount of cations will be tested to unravel the possibility of analytical applications and the origin of the dual fluorescence. We devote particular attention to the photoinitiated processes in the cavity of cucurbiturils and sulfonatocalixarenes because the embedment in these water-soluble, nontoxic molecular containers is anticipated to allow the control of the kinetics of the photochromic and energy dissipation processes. The size and the flexibility of the cavitands as well as the molecular structure of the guest compounds will be varied. We also intend to reveal how the inclusion of two guests influences the processes following light absorption. On the basis of the promising preliminary results sulfonatocalix[8]arene is expected to promote organic nanoparticle formation and aggregation. The binding of environment-sensitive fluorescent dyes and alkaloids to natural polyelectrolytes and to new substances synthesized by French scientists will be investigated to get deeper insight into the factors determining the pH-controlled release process. A part of the proposed research is related to a CNRS-MTA French-Hungarian bilateral cooperation program.

What is the major research question?

The proposed research focuses on the fundamental question how the addition of electrolytes and the confinement in the cavity of macrocyclic compounds alter the properties of the excited molecules. Systematic studies are planned to understand the effect of cations on the fluorescent behaviour of selected alkaloids, whose photophysical processes have not been studied despite their biological importance. Inspired by the rapidly growing international interest in cucurbiturils and sulfonatocalixarenes, we intend to reveal how the competition among the various excited-state deactivation processes of the excited guest molecules alters upon interaction with these water-soluble, biocompatible cavitands. As an extension of the results reported in our recent proof-of-concept paper, we start a comprehensive study on the photochromism in the cavity of macrocycles. We intend to unravel how the charge, size, flexibility and redox potential of the macrocycle influence the kinetics of the photochromic transformations and the long-term stability of various dyes. Our preliminary observation that sulfonatocalix[8]arene is able to promote organic nanoparticle formation will serve as a starting point for the investigation of the chemical structure requirements of the macrocycle-induced aggregation. Such process may facilitate the tuning of the fluorescence properties of widely used H- and J-aggregates of cyanine dyes. We will also study how the interaction with weakly acidic polyelectrolytes modifies the fluorescent behaviour of alkaloids and environment-sensitive probe molecules.

What is the significance of the research?

There is an increasing demand for fluorescent probes that are capable of the indication of the local changes in their environment as well as for self-assembled systems whose properties can be regulated by light. To comply with such demands, detailed knowledge of the major factors determining the rate of the various deactivation pathways of the excited molecules is needed. This project will significantly contribute to the better understanding of the effect of the microenvironment change on the light-initiated processes. The results are expected to promote the utilization of alkaloids as fluorescent probes and open up new possibilities in a wide variety of applications of photochromic compounds in aqueous solution. The negligible solubility of the spiro form and the rapid decomposition of the merocyanine form precluded the utilization of the spiropyran type of photochromes in water. With inclusion complex formation not only these problems can be solved but also the rate of the switching between the two forms becomes controllable. Our results on the alkaloid binding to cavitands and polyelectrolytes will foster the development of new drug carrier systems. The investigation of the inclusion complex formation may lay the foundation for the design of novel molecular-scale devices, sensors, analytical assays and stereoselective photochemical reactions. Due to their much stronger and more selective binding capability, cucurbiturils can replace cyclodextrins in many applications. The ability of cucurbit[8]uril to accommodate two different guests can lead to a wide variety of self-assembled architectures and supramolecular polymers. Therefore, the scientific knowledge obtained from basic studies of ternary cucurbit[8]uril complexes is of significant practical value.

Because of the ubiquitous nature of light, it is of primary importance to understand the sequence of events following light absorption. The illumination of materials can lead to fluorescence emission or chemical changes. The aim of this project is to reveal how these photoinitiated effects can be modified by the addition of metal salts, macrocyclic compounds and polymers. We use the alteration of the fluorescent behaviour to get information on the interactions among molecules and ions. The fluorescence of the pharmaceutically important alkaloids will be studied to reveal whether it can be utilised for the detection of trace amount of metal ions. Considerable attention will be devoted to the confinement of molecules in the cavity of macrocyclic compounds that can serve as molecular containers providing protection and different local environment for the encapsulated species. We intend to use the inclusion of two different molecules in the molecular containers to tune the fluorescent properties and to modify the rate and selectivity of chemical reactions. The photochromic compounds, which change colour and molecular structure upon irradiation with light, are particularly important for the design of photoresponsive materials, ophthalmic lenses and for information recording. The encapsulation of these substances in various molecular containers is planned not only to regulate the rate of the colour variation but also to enhance the solubility and stability in water. We will also study the interactions between polymers and alkaloids. The results will contribute to the development of new drug delivery vehicles allowing the controlled release of the biologically active component.

Biológiailag aktív alkaloidok, intenzíven fluoreszkáló és fotokróm vegyületek fényelnyelést követő folyamatait tanulmányoztuk vízben és makrociklusok üregében. Az asszociációs folyamatok által okozott jelentős fluoreszcenciás sajátság változás alapján dimerizációt, illetve kukurbituril és 4-szulfonátokalixarén makrociklusos vegyületekkel beékelődési komplex képződést tudtunk kimutatni. Különböző kísérleti technikák alkalmazásával és a kapott eredmények együttes kiértékelésével feltártuk az egyes kötődési folyamatok termodinamikai és kinetikai paramétereit. Nagypontosságú módszert fejlesztettünk ki kukurbit[7]uril üregéből kilépés sebességi együtthatójának szelektív meghatározására. Megállapítottuk, hogy a makrociklus mérete és a pH miként befolyásolja a 4-szulfonátokalixanénekhez kötött fotokróm vegyület átalakulásait. Kimutattuk, hogy a 6-metoxi-1-metilkinolinium fluoreszcencia összetett lecsengési kinetikája két izomer jelenlétére utal, melyek a metoxicsoport irányában különböznek egymástól. Meghatároztuk e vegyület kukurbit[7]urillal és 4-szulfonátokalix[4]arénnel képzett komplexének sajátságait. 1-Alkil-3-metil-imidazolium kation és 4-szulfonátokalixarének önszerveződésével nanorészecskéket állítottunk elő, melyek meghatározott körülmények között reverzibilis folyamatban szupramolekuláris micellává alakultak át. Protonált kitozánból 4-szulfonátokalixarénnel olyan nanorészecskéket tudtunk képezni, melyek alkalmazhatók farmakológiai fontosságú alkaloidok hordozására.
Results in English
Photoinduced processes of biologically active alkaloids, highly fluorescent dye, and photochromic compound were studied in water and in the cavity of macrocycles. The substantial change of the fluorescent properties upon association was exploited to monitor dimerization and inclusion in the biocompatible cucurbituril and 4-sulfonatocalixarene cavitands. Both the thermodynamics and the kinetics of the individual binding processes were revealed by the combination of various experimental techniques and the global analysis of their results. A highly accurate method was developed for the selective determination of the rate constant of the exit from the apolar interior of cucurbit[7]uril. We revealed how the macrocycle size and pH influence the photochromic transformations in the presence of 4-sulfonatocalixarenes. The dual-exponential fluorescence decay kinetics of the free 6-methoxy-1-methylquinolinium was assigned to two torsional isomers differing in the orientation of the methoxy moiety. Its binding to cucurbit[7]uril or 4-sulfonatocalix[4]arene had markedly different features. Multiple binding of 1-alkyl-3-methylimidazolium to 4-sulfonatocalixarenes promoted the formation of nanoparticles, which could reversibly reorganize to supramolecular micelles under specific conditions. The nanoparticles produced by the 4-sulfonatocalixarene-initiated self-assembly of protonated chitosan efficiently incorporated pharmaceutically important alkaloids.
