A peptaibolok a gombák peptid antibiotikumainak egyik családját alkotó másodlagos anyagcseretermékek. A legtöbb peptaibol-termelő szervezet az Ascomycota törzs Hypocreales rendjébe sorolt gombafaj, melyek közül a Trichoderma nemzetség képviselői a legjelentősebb, az ismert peptaibol vegyületek mintegy kétharmadának termeléséért felelős fajok. Ismert, hogy a peptaibolok a biológiai aktivitások széles spektrumával (antibakteriális, antifungális, antivirális hatások, immunszupresszív és neuroleptikus sajátságok, toxicitás emlőssejtekre, szisztémás rezisztencia indukciója növényekben) jellemezhetők, a szakirodalomban mégis kevés, széles skálájú bioaktivitási felmérésből származó adatot közöltek.
Kutatásunk céljai között szerepel:
- peptaibolok izolálása a Hypocreales rendbe tartozó gombákból,
- a molekulák kémiai összetételének és háromdimenziós térszerkezetének meghatározása,
- a peptaibolok baktériumokkal, gombákkal, emlőssejtekkel és növényekkel szembeni bioaktivitásának jellemzése,
- a peptaibolok szerkezeti sajátságai és bioaktivitása közötti szerkezet-aktivitás összefüggések azonosítása,
- a peptaibolok micellákkal és membránokkal történő kölcsönhatásainak tanulmányozása.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.
Az izolálás, tisztítás, elválasztás és biológiai jellemzés eredményei megmutatják, mely taxonómiailag azonosított törzsek termelnek peptaibolokat; milyen típusú molekulákat termelnek a különböző fajok; és a peptaibolok milyen bioaktivitással rendelkeznek baktériumokkal, gombákkal, emlőssejtekkel és növényekkel szemben. A peptaibolok térszerkezetének vizsgálata a konformációs sajátságaik azonosítása miatt fontos, amelyek segíthetnek a hatásmechanizmusok megértésében. A szerkezet-aktivitás összefüggések vizsgálata információt adhat a peptaibolok szerkezeti tulajdonságai és bioaktivitása közötti korrelációkról.

Mi a kutatás jelentősége?

Az új peptaibolok azonosítását és szerkezeti értelmezését célzó komplex szűrőprogram a különböző organizmusokkal szembeni bioaktivitások tesztelésével együtt új antimikrobiális szereket azonosíthat. Az antimikrobiális kemoterápia számára egyre növekvő kihívást jelent az antibiotikumokkal szemben nagyfokú rezisztenciát mutató patogén mikroorganizmusok megjelenése, ezért különösen fontosak az új, élő szervezetek által termelt molekulák azonosítására irányuló kutatások. A peptaibolokkal kapcsolatos, mélyreható ismeretek fontos információkkal szolgálhatnak ezen vegyületek humán patogén mikroorganizmusok elleni alkalmazhatóságával kapcsolatosan. A klinikai szempontból jelentős baktériumokkal és gombákkal szemben aktivitást mutató, de emlőssejtekre nem toxikus peptaibolok antimikrobiális, immunszupresszív és neuroleptikus hatású gyógyszerjelölteket nyújthatnak a humán gyógyászat számára, a növénypatogén baktériumokkal és gombákkal szemben hatásos, de a hasznos mikroszervezeteket és a növényeket nem károsító peptaibolok pedig mezőgazdasági alkalmazásokra szánt biokontroll készítmények potenciális komponensei lehetnek.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.
A gombák által termelt peptid antibiotikumok csoportjába tartozó, ún. „peptaibol” vegyületek széleskörű biológiai hatásairól kevés információ áll rendelkezésre. Kutatásunk során célul tűztük ki peptaibol vegyületek gombákból történő tisztítását, kémiai összetételük és szerkezetük meghatározását, valamint az egyes peptaibol vegyületek baktériumokra, gombákra, emlőssejtekre és növényekre gyakorolt biológiai hatásainak feltérképezését. A tervezett szűrőprogram új, emberi betegségeket okozó mikrobák ellen hatásos gyógyszerjelölteket azonosíthat, és fontos információkkal szolgálhat a peptaibol vegyületek környezetbarát, biológiai növényvédelemben történő felhasználási lehetőségeivel kapcsolatban is.

Peptaibols are secondary metabolites constituting a family of fungal peptide antibiotics. Most of the peptaibol producers are fungal species from genera belonging to the Hypocreales order of the Ascomycota division, with members of the genus Trichoderma being the most important, responsible for the production of about two third of the known peptaibols. Although it has been revealed that peptaibols possess a wide spectrum of biological activities including antibacterial, antifungal and antiviral effects, immunosuppressive and neuroleptic properties, toxicity to mammalian cells and elicitation of systemic plant resistance, there is a lack of wide-scale bioactivity screening data in the literature.
The aims of our project are:
- to isolate peptaibols from Hypocrealean fungi
- to determine their chemical composition and three-dimensional structure
- to characterize their bioactivities against bacteria, fungi, mammalian cells and plants
- to identify structure-activity relationships between their structural features and bioactivities,
- to study the interactions of peptaibols with micelles and membranes.

What is the major research question?

The results of isolation, purification, separation and biological characterization will show, which taxonomically identified strains produce peptaibols; what type of these molecules are produced by different species; and what kind of bioactivities peptaibols possess against bacteria, fungi, mammalian cells and plants. The structural investigation of peptaibols is important to identify their conformational features, which can help to understand the mode of action of these molecules. The structure-activity relationship studies can supply information about the correlations between the structural properties and bioactivities of peptaibols.

What is the significance of the research?
Describe the new perspectives opened by the results achieved, including the scientific basics of potential societal applications. Please describe the unique strengths of your proposal in comparison to your domestic and international competitors in the given field.
Complex screening programs aimed at the identification and structural elucidation of new peptaibols along with testing their bioactivities towards various organisms could provide novel antimicrobial agents. The efficiency of antimicrobial chemotherapy is increasingly challenged by the emergence of pathogenic strains exhibiting high levels of antibiotic resistance, therefore it is very important to search for novel compounds produced by living organisms. Detailed knowledge of the properties of peptaibols may provide important information about their potential applicability as drugs against human pathogenic microorganisms. Peptaibols active against clinically relevant bacteria and fungi but non-toxic to mammalian cells may reveal new antimicrobial, immunosuppressive and neuroleptic drug candidates for human medicine, while those ones that are active against plant pathogenic bacteria and fungi but less affecting beneficial microorganisms and plants could be potential components of biocontrol preparations intended for agricultural applications.

Only limited information is available about the wide-scale biological activities of the so called "pepataibol" compounds belonging to the group of peptide antibiotics produced by fungi. The aim of our research is the purification of peptaibol compounds from fungi, the determination of their chemical composition and structure as well as the mapping of their biological activities towards bacteria, fungi, mammalian cells and plants. The planned screening programme may reveal new drug candidates that are effective against microbes causing human diseases, and provide important information about the application possibilities of peptaibol compounds in the environment-friendly, biological control of plant diseases.

Trichoderma törzseket izoláltunk különböző élőhelyekről, illetve szereztünk be törzsgyűjteményekből. Kidolgoztunk egy Micrococcus luteus gátlásán alapuló, gyors és érzékeny biológiai tesztet a Trichoderma törzsek peptaibol vegyületeinek detektálására. A peptaibolok termelésére a malátakivonatos agar bizonyult az optimális tápközegnek. Optimalizáltuk a peptaibolok tisztítását és tömegspektrometriai elemzését. Összesen 51 Trichoderma törzs peptaibol-termelését vizsgáltuk. A detektált peptaibolok között trichorzianinokat, trichotoxinokat, trichostrigocinokat, illetve trichobrachin- és hypomurocin-szerű peptideket azonosítottunk. A korábban leírt peptaibol-családokon belül számos új szekvenciát azonosítottunk, leírtunk továbbá a T. pleuroti-ból egy teljesen új peptaibol-családot, melyet tripleurin-oknak neveztünk el. Minden vizsgált peptaibol-csoport membránkárosodást okozott emlőssejtekben, illetve gátolta számos, klinikai és mezőgazdasági szempontból releváns baktérium, élesztő és fonalasgomba növekedését, az Arabidopsis thaliana gyökérnövekedésére viszont koncentrációfüggő módon hatottak a vizsgált peptaibolok. Különböző rövid-szekvenciájú peptaibolok esetén meghatároztuk a jellegzetes térszerkezeti tulajdonságokat, illetve különféle hosszú-szekvenciájú peptaibolok esetében azonosítottuk a karakterisztikus konformációs és folding sajátságokat. Jellemeztük továbbá a rövid- és hosszú-szekvenciájú peptaibolok és micellák/membránok között kialakuló kölcsönhatásokat.
Results in English
Trichoderma strains were isolated from different habitats and procured from culture collections. A rapid and sensitive biological test based on the inhibition of Micrococcus luteus was established for the detection of peptaibols from Trichoderma strains. Malt extract agar was identified as the optimal medium for peptaibol production. Purification and mass spectrometric analysis of peptaibols were optimized. A total number of 51 Trichoderma strains were examined for peptaibol production. The detected peptaibols included trichorzianins, trichotoxins, trichostrigocins as well as trichobrachin- and hypomurocins-like peptides. A series of new sequences were identified within the previously described peptaibol families, furthermore, an entirely new family of peptaibols named tripleurins were also described from T. pleuroti. All peptaibol groups were found to cause membrane damage in mammalian cells, to inhibit the growth of various clinically and agriculturally relevant bacteria, yeasts and filamentous fungi, while the root development of Arabidopsis thaliana was affected by te examined peptaibols in a concentration-dependent manner. For different short-sequence peptaibols, the typical structural properties were determined, as well as in the case of various long-sequence peptaibols, the characteristic conformational and folding features were identified. Nevertheless, the interactions evolved between the short- and long-sequence peptaibols and micelles/membranes were characterized.
