A hősokkfehérjék (Hsp) legfontosabb feladata, hogy segítenek a hibás fehérjék helyreállításában, valamint a sejtvázhoz és a plazmamembránhoz kapcsolódva megóvják azokat stressz körülmények között. A Hsp-k szintje csökkent az agyban különböző neurodegeneratív elváltozások során, ezért fontos terápiás célpontok lehetnek e betegségekben. Az Alzheimer kór legfontosabb tünetei az amiloid plakkok és neurofibrilláris kötegek kialakulása, de a vér-agy gát szerkezete és funkciója is megváltozik. Ezen kívül már a betegség korai szakaszaiban megfigyelhető a membrán raftok károsodása és különböző lipid molekulák megjelenése, ezek a változások a kór korai, ám eddig kevéssé vizsgált tünetei közé tartoznak. Újabb eredmények szerint a stressz hatására felszabaduló specifikus lipidek másodlagos hírvivő szerepet töltenek be, és részt vesznek a Hsp-k szabályozásában a HSF1 transzkripciós faktoron keresztül. A „kalpain- hydroxynonenal-Hsp70-katepszin kaszkád” elmélet szerint a HNE-karbonilált Hsp70 fokozott lebontása a lizoszómális membrán destabilizációjához, majd a katepszin enzim felszabadulásához vezet. A Hsp70 diszfunkciója a foszforilált tau aggregációját, és az axonális transzport működési zavarait okozhatja. Korábbi eredményeink szerint a dokozahexaénsav (DHA) kezelés hatására csökken az Aβ fehérje termelődése, amely folyamatban a lipid raftok újrarendeződésének is fontos szerepe van. Azt is kimutattuk, hogy a Hsp27 védő hatással rendelkezik az Alzheimer kór egyes tüneteivel szemben. Kísérleteink célja, hogy hogy jobban megismerjük a Hsp27 által kifejtett védőhatás molekuláris mechanizmusát, különösen a membránnal, membrán lipidekkel való kölcsönhatás tekintetében.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.
Az Alzheimer kór (AD) kialakulásának és a stresszfehérjék által kifejtett védő hatásnak a pontos molekuláris mechanizmusa máig sem teljesen tisztázott. Kísérleteink során szeretnénk megvizsgálni a vér-agy gát károsodásának mértékét az AD egérmodelljében, valamint arra is kíváncsiak vagyunk, hogy a Hsp27 túltermelése képes-e enyhíteni a vér-agy gát szerkezetében és funkciójában bekövetkező változásokat. Részletesebben tanulmányozzuk a Hsp27 és az β-amiloid (Aβ) fehérje kölcsönhatását, megvizsgáljuk milyen szerepet játszik a Hsp27 az Aβ keletkezése és lebontása során. A membránhoz köthető változásokat lipidomikai analízissel tanulmányozzuk az AD egérmodelljében, a hiperlipidémiás (ApoB-100 transzgenikus) egérmodellben valamint a Hsp27 túltermelő egértörzsben, hogy jobban megismerjük a betegségek kialakulásában és a Hsp27 által kifejtett védő hatásban szerepet játszó, membránhoz kapcsolódó folyamatokat. Ezen kívül a Hsp27 és a sejtmembrán között kialakuló kölcsönhatásokat is tanulmányozzuk. DHA kezelés után szeretnénk vizsgálni az Aβ termelődés és felhalmozódás mértékét, valamint a membránban bekövetkező változásokat az AD egérmodelljében és a Hsp27 fehérjét túltermelő AD modellben. Mivel a dembrán lipid összetétele és szervezettsége befolyásolja a stresszfehérjék termelődését, terveink között szerepel még a HSF1 vizsgálata az AD egérmodelljében (DHA kezelt illetve nem kezelt állatokban), ami más stresszfehérjék, köztük a Hsp70 szabályozását is befolyásolja.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!
A tervezett kísérletek eredményei várakozásunk szerint közelebb vihetnek az Alzheimer kór valamint a hipelipidémia által kiváltott neurodegeneráció kialakulásának pontosabb megértéséhez. Ezen kívül jobban megismerhetjük a stresszfehérjék idegrendszerben kifejtett védő hatásának mechanizmusát. Az eredményeket felhasználva a jövőben lehetőség lesz új gyógyszerek és terápiák kifejlesztésére, amelyek segítségével megelőzhetőek vagy akár kezelhetőek lesznek egyes neurodegeneratív betegségek.

Jelenleg közel 30 millió ember szenved Alzheimer kórban világszerte, évente körülbelül 5 millió új esetet diagnosztizálnak. Ahogy az emberek várható élettartama növekszik, az időskori demencia egyre súlyosabb problémát jelent társadalmunkban. Az Alzheimer kór jelenleg gyógyíthatatlan betegség, kialakulásának oka és a betegség lefolyásának pontos mechanizmusa máig sem teljesen tisztázott. A vizsgálatainkból származó eredmények reményeink szerint hozzájárulnak a betegség alaposabb megismeréséhez. A kutatások nem csak elméleti jelentőségűek, de lehetővé teszik, hogy a jövőben kifejleszthessünk és tesztelhessünk új gyógyszereket és terápiákat, melyek a betegség megelőzését, gyógyítását célozzák.

Heat shock proteins (Hsps) are molecular chaperones, they can refold damaged proteins and stabilize cytoskeletal elements, cell membranes during stress conditions. Hsps are downregulated in the brain in many neurological diseases therefore they are promising therapeutic targets in these pathological conditions. The major hallmarks of Alzheimer’s disease (AD) are amyloid plaque deposition and neurofibrillary tangle formation, but the structure and function of BBB is also damaged. Moreover, disruption of lipid „rafts” and appearance of distinct lipid molecular species were observed in the first stages of AD suggesting that these alterations represent hitherto unexplored, early events in the neuropathological development. The stress- activated release of specific lipids have emerged as second messengers which play a key role in the regulation of Hsp expression, via HSF1. As described in the novel “calpain-hydroxynonenal-Hsp70-cathepsin cascade” hypothesis the enhanced cleavage of HNE-carbonylated Hsp70 leads to destabilization of the lysosomal membranes and the release of cathepsin enzymes into the cytoplasm. The dysfunction of Hsp70 is causing aggregation of phosphorylated tau and disturbance of the axonal transport. Earlier we have shown, that docosahexaenoic acid (DHA) intake strongly reduces Aβ production via multiple mechanisms including remodeling the lipid rafts. Our previous results also revealed that Hsp27 has a significant neuroprotective function in AD. The major aim of the present study is to reveal the molecular nature of the neuroprotective mechanism of Hsp27 with regard to its specific interaction with, and regulation by membrane lipids.

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.
The exact molecular mechanism of the development of AD and the method of neuroprotection is still not completely understood. In this study we will investigate the BBB permeability in AD model mice, and also test whether Hsp27 can help to restore the stucture and function of BBB in Hsp27/AD model mice. Then the role of Hsp27 in Aβ accumulation and clearance and the interaction between Hsp27 and Aβ will be studied. With lipidomic analysis, pathophysiological alterations in AD model, in hyperlipidemic (ApoB-100 transgenic) and also in Hsp27 overexpressing mice will be studied to reveal the early membrane related events in disease development. We will also investigate the interaction of Hsp27 with plasma and cellular membranes. Following DHA supplementation the Aβ production and lipid alterations will be tested simultaneously both in the AD and AD/Hsp27 mice models. Since the lipid composition and microdomain organization are known to refine Hsp expression, finally we will determine the HSF1 modulations which may result in deregulation of other Hsps, especially Hsp70, using the different AD mice models, treated or untreated by DHA diet.

The expected results of the proposed experiments will contribute to the better undestanding of the molecular mechanism of the development of AD and hyperlipidemia induced neurodegeneration. The study will also help to reveal the mechanism of the known neuroprotective effects of Hsp27. In the future, using these results we can develop drugs and methods to treat or prevent neurodegenerative diseases.

Currently almost ~30 million people are diagnosed with AD worldwide, with ~5 million new cases annually. As life expectancy increases age-associated dementias constitute a serious health problem for our societies. Currently there is no cure for AD and the origin and development of the disease are not completely understood. The expected results will contribute to the better understanding of the disease. These investigations have not merely of theoretical importance, but they will contribute to the development of drugs and successful therapies against neurodegenerative diseases, such as Alzheimer’s disease.

Kísérleteink során az időskorban előforduló idegrendszeri betegségek (vaszkuláris demencia, Alzheimer-kór) kialakulásának valamint a stresszfehérjék által kifejtett idegrendszeri védő hatás mechanizmusának pontosabb megismerését tűztük ki célul. Ennek érdekében megvizsgáltuk az Apob100 túltermelő hiperlipidémiás egerekben kialakuló idegrendszeri elváltozásokat. Megfigyeltük, hogy a törzsre jellemző magas szérum triglicerid szint hatására megváltozik a neurovaszkuláris egységet alkotó sejtek morfológiája és funkciója, ami a vér-agy gát áteresztőképességének növekedéséhez vezet. Ezzel a kezünkbe került egy olyan állatmodell, amely kulcsfontosságú a keringési eredetű neurodegeneratív betegségek kialakulásának jobb megértéséhez. A Hsp27 túltermelő egértörzsben végzett vizsgálataink igazolták, hogy a stresszfehérje hatással van a vér-agy gátat alkotó endotélsejtek génexpressziójára, ezen kívül szerepe lehet a gyulladásos folyamatok szabályozásában azáltal, hogy fokozza különböző citokinek expresszióját, valamint befolyásolja a glia aktivációt agysérülést követően. Az Alzheimer-kór transzgenikus egérmodelljén kimutattuk, hogy egy stresszfehérje koindukáló szerrel végzett kezelés csökkenti az amiloid plakkok számát és a neuronpusztulás mértékét, valamint javítja az állatok kognitív képességeit. Eredményeink alapján feltételezzük, hogy a stresszfehérjék terrmelődésének farmakológiai modulációja protektív hatású lehet különböző neurodegeneratív betegségekben.
Results in English
In this project our specific aims were to reveal the molecular mechanism of the neuroprotective role of Hsp27 and to understand the pathology of different neurodegenerative dysfunctions like Alzheimer’s disease and vascular dementia. First we analyzed the effects of hyperlipidemia on neurovascular function in the ApoB100 overexpressing mouse strain. The increased serum triglyceride level resulted in the morphological and functional changes of the neurovascular unit leading to increased permeability of the blood-brain barrier. Based on these results, we can suggest that this mouse strain is a suitable model for hyperlipidemia-induced vascular neurodegeneration. Using the Hsp27 overexpressing mouse strain we found, that this heat shock protein alters the gene expression of brain endothelial cells, and can promote the synthesis of inflammatory cytokines influencing glia activation under acute brain injury, indicating a pivotal role in the regulation of inflammatory responses. It was also demonstrated that chronic administration of a stress protein co-inducer preserves the number of neurons, reduces amyloid plaque formation and improves cognitive function in a mouse model of Alzheimer’s disease. According these data, we propose that pharmacological modulation of heat shock protein production could be a potential therapeutic strategy in different neurodegenerative diseases.
