A pályázat keretében Magyarországon és Európa más területein is azonosítottunk és jellemeztünk egy korábban alig ismert vírust, a Lloviu vírust. Ez az Ebola és Marburg vírusokat is magába foglaló csoportba, a filovírusok csoportjába tartozik. Vizsgálataink során felmértük a fokozottan védett hosszúszárnyú denevérekre gyakorolt hatását, a denevérek közti terjedési mechanizmusait és több módszerrel vizsgáltuk az emberre vagy más állatra vonatkozó fertőzési képességét, a megelőzés lehetőségeit. A munka során egy sor új technológiai eljárást dolgoztunk ki (terepi diagnosztikai és genomszekvenálási eljárások), illetve hazánkban új technológiaként értelmezhető vírusgenomikai eljárást alkalmaztunk. A pályázat legfontosabb eredménye, hogy a vírus természetes formáját, azaz vad típusát is sikerült laboratóriumi körülmények között fenntartani, azaz izolálni – ez a filovírusok hat évtizedes kutatása során csupán a Marburg vírus esetében sikerült eddig közvetlenül denevérből. Így világszerte megnyílt a lehetősége, hogy a rendkívül súlyos emberi járványokért felelős filovírusokat jobban megérthessük. Az eredményt még különlegesebbé teszi, hogy ez az első mérsékelt égövi filovírus melyet a pályázatnak köszönhetően immáron széleskörű vizsgálatok alá lehet vetni. További újdonság, hogy a denevérek saját parazitáiban, az úgynevezett denevérlegyekben és kullancsokban is kimutattuk a vírust. Ezt korábban több évtizednyi kutatás során sem sikerült elérni a Lloviu vírus afrikai rokonai kapcsán.
Results in English
Through this grant, we identified and characterized a previously less known virus, the Lloviu virus in Hungary and other parts of Europe. This virus belongs to the filovirus group, which includes the Ebola and Marburg viruses. Our studies assessed its impact on the highly protected bent-winged bats, its transmission mechanisms among these bats, and its infectious potential for humans and other animals, and related prevention strategies. We developed a range of new technological procedures, including field diagnostic and genome sequencing techniques, and introduced novel virus genomics technology in Hungary. A key achievement of the project was the isolation of the natural, wild-type form of the virus under laboratory conditions, directly from bats. It was only accomplished before for the Marburg virus in six decades of filovirus research. This opens up novel research opportunities worldwide to better understand filoviruses, responsible for severe human epidemics. Remarkably, this research marks the first extensive study of a temperate-climate filovirus, enabled by this grant. An additional breakthrough was detecting the virus in the bats' own parasites, specifically bat flies and ticks, an achievement not reached in decades of research on the African relatives of the Lloviu virus.
