Olyan új NMR módszereket dolgoztunk ki, amellyekkel molekulák térszerkezetét és mozgását a korábbiaknál pontosabban tudjuk leírni. A módszerek a spin-spin csatolási állandó mérésén és a relaxációs jelenségen alapulnak. Fehérjék és kis molekulák kölcsönhatásának vizsgálatára új, az izotópjelzésen alapuló módszereket fejlesztettünk. Kidolgoztuk a 15N csoport-szelektív telítés módszerét, amely akkor is lehetővé teszi a fontos STD módszer alkalmazását, amikor a gazdamolekula és a ligandum NMR jelei átfednek. Példaként a 15N jelzett glikopeptid antibiotikum / sejtfal - analóg peptid rendszerben igazoltuk a kötődést. Megállapítottuk, hogy a kalcium kötő Calretinin fehérje I-II és a III-VI moduljai független doménokat alkotnak, valamint azt, hogy a vizsgált pH tartományban nem változik az I-II domén másodlagos szerkezete. Helikális modell peptideket használva valószínűsítettük az aromás-főlánc kölcsönhatások fontosságát helikális fehérjékben. Az a - amilázt gátló szintetikus poliszacharidok és a spiro-hidantoidin tartalmú akarbóz analóg szerkezetét meghatároztuk. Diffúziós NMR vizsgálatokkal bizonyítottuk, hogy az izopropilidén-guanozin polietilén-glikollal (PEG) kialakított telekelikus dimerje K+ ion jelenlétében G4 kvadruplexeket alkot. A képződő gyöngyfüzér-szerű szupramolekuláris polimerek (20 kDa - 10 MDa) makromonomerje G4-(PEG)4-G4 szerkezetű, amely képes kisméretű hidrofób ligandum „becsomagolására”. A hasonló vegyületek potenciálisan telomeráz enzim gátlók lehetnek.

We have elaborated new NMR methods that are useful for accurate characterization of molecular structure and dynamics. These methods are based on the measurement of spin-spin coupling constants and relaxation phenomena. For studying protein small-molecule interactions we introduced new techniques utilizing labelled compounds. The 15N group-selective saturation method allows magnetization transfer from the isotope labelled host to the unlabelled guest, that is very important in those STD applications where the spectra overlap. For demonstration, we verified the binding of cell -wall analogue peptides to 15N labelled glycopeptide antibiotics. Studying the calcium binding protein Calretinin we proved that modules I-II and III-VI form independent domains. Furthermore, the secondary structure of CR I-II is independent of pH. Studying helical model peptides we suggest the importance of aromatics / main chain interactions. We have determined the structure of synthetic polysaccharides and an acarbose analogue with a spiro-hidantoidin tag, all inhibiting a-amylase. We proved by NMR diffusion studies that a lipophilic, telechelic polyethylene glycol dimer of guanosine, in the presence of potassium ion, assembles into a dynamic supramolecular polymer with the macromonomer structure as G4-(PEG)4-G4. The ''bead in the string'' structure extends to 20 kDa - 10 MDa mass, and is able to encapsulate small hydrophobic ligands. Similar structures may be telomerase inhibitors.
