Az elmúlt két évtizedben számos új sejtciklus-gént írtunk le. A sep1 és az ace2 transzkripciós faktorokat kódolnak, amelyek számos M-fázis- és citokinezis-gén transzkripciójához kellenek. A sep10/MED31, amely inaktiválása citokinezis-defektussal jár, a Mediátor-komplex alegységét kódolja. A tervezett munka fő céljai (1) a Sep10 által kontrollált gének identifikálása a S. pombe genomjában, (2) a köztük előforduló, még ismeretlen citokinezis-gének megtalálása és (3) a Sep10 fehérje funkcionális kölcsönhatásainak vizsgálata a Sep1 és az Ace2 transzkripciós faktorokkal.
Summary
Over the last two decades we have described numerous novel cell cycle genes. sep1 and ace2 turned out to act as transcription factors of many M-phase and cytokinesis genes. sep10/MED31, whose inactivation causes defective cytokinesis, encodes a subunit of the Mediator complex, a major regulator of transcription. The aims of the proposed project are (i) to identify all Sep10-dependent genes in the Sch. pombe genome, (ii) to find novel cytokinesis genes among them and (iii) to investigate the interactions between the Sep10 protein and the transcription factors Sep1 and Ace2.

Az elmúlt két évtizedben számos új sejtciklus-gént írtunk le. A sep1 és az ace2 transzkripciós faktorokat kódolnak, amelyek számos M-fázis- és citokinezis-gén transzkripciójához kellenek a Schizosaccharomyces pombe-ben. A sep10/MED31, amely inaktiválása citokinezis-defektussal jár, a Mediátor-komplex alegységét kódolja. A Mediátor egy általános transzkripciós faktor. Ebben a projektben azokat a géneket identifikáltuk a S. pombe genomjában, amelyek transzkripciójához szükséges a Sep10 protein aktivitása. A gének között találtunk újabb citokinezis géneket is, de sem a sep1 sem az ace2 nem bizonyult Sep10-függőnek. Az utóbbi eredményből arra következtettünk, hogy a Sep10 koaktivátorként működhet együtt az Ace2-vel a citokinezis-gének promóterein. Kiderítettük, hogy a SAGA komplex egyik alegységét képviselő Sep9 fehérje a citokinezist két szinten is regulálja: mint az ace2 transzkripciójának szabályzója és mint az Ace2 fehérjével együttműködő koaktivátor a citokinezis-gének promóterein. Észrevettük, hogy a Kin1 kináz részt vesz a citokinezis végén lejátszódó sejtszeparáció finom-hangolásában. Végül a citokinezisben szereplő egyes fehérjékkel filogenetikai analízist is végeztünk, amiből kiderült, hogy a Schizosaccharomyces fehérjék egy különálló evolúciós ágat alkotnak a gombák birodalmában.

Over the last two decades we have described numerous novel cell cycle genes. The products of sep1 and ace2 turned out to act as transcription factors of many M-phase and cytokinesis genes in Schizosaccharomyces pombe. sep10/MED31, whose inactivation causes defective cytokinesis, encodes a subunit of the Mediator complex, a major regulator of transcription. In this project we identified genes in the S. pombe genome that require active Sep10 protein for being transcribed and found novel cytokinesis genes among them. Neither sep1 nor ace2 are among the controlled genes, indicating that the Sep10 protein may act as a coactivator of ace2 at the promoters of the cytokinesis genes. We revealed that the SAGA-complex subunit Sep9 regulates the cytokinesis genes at two levels: as the regulator of ace2 and as a coactivator cooperating with Ace2 at the promoters of the cytokinesis genes. We noticed that the Kin1 kinase is involved in the fine-tuning of the cell separation machinery at the end of cytokinesis. Finally we performed a phylogenetic analysis of certain components of the cytokinetic machinery and found that the Schizosaccharomyces proteins represent a unique evolutionary lineage in the Kingdom of Fungi.
