Az endoplazmás retikulum (ER) számos környezeti és metabolikus hatás szenzora. Mindazon tényezők, melyek károsan befolyásolják a szekréciós fehérjék poszttranszlációs módosítását és foldingját az ER lumenében, a fehérjék felhalmozódásához vezetnek, és kiváltják az ER stressz jelenségét. A stresszben jelpályák indulnak be az ER és a sejtmag között, melyeknek hatására a fehérjeszintézis csökken, viszont szelektíven indukálódnak a poszttranszlációs módosításokért felelős enzimek és az intraluminális dajkafehérjék. Huzamosan fennálló stressz esetén beindulnak a programozott sejthalál mechanizmusai. Az experimentális ágensekkel kiváltható ER stressz és apoptózis mellett a folyamat számos humán kórkép (cisztikus fibrózis, Alzheimer kór, diabetes, vírusfertőzések stb.) patomechanizmusában is szerepel.
A pályázat célja (i) redox aktív vegyületek, antioxidánsok és hepatotoxinok szerepének vizsgálata az ER eredetű stresszben és apoptózisban, valamint (ii) egy új lehetséges stresszmechanizmus, a redox hatásokra bekövetkező ER membrán permeabilitásváltozás tanulmányozása. Az első kérdéskört három kísérleti rendszerben tanulmányoznánk:
• Előzetes eredményeink szerint az aszkorbinsav fontos szerepet játszik a diszulfid kötések kialakításához szükséges elektrontranszferben. In vivo aszkorbáthiányos állapot (skorbut) tehát feltehetően kiválthatja az ER stresszt.
• A neutrofil granulocita ER membrán glukóz-6-foszfát transzporterének genetikai hiánya, illetve a transzport gátlása apoptózishoz vezet; a folyamatban szerepet tulajdonítunk az intraluminális redox státusz változásának. Vizsgálni kívánjuk az apoptózis eredetét, az ER szerepét, illetve a folyamat patomechanizmusát.
• A hepatotoxinok nagy része az ER-ban aktiválódik, illetve metabolizálódik. A folyamat befolyásolhatja a lokális redoxpotenciált. Feltételezzük, hogy ezen vegyületek toxikus, illetve apoptotikus hatásárt – a korábbi hipotézisekkel szemben – az ER stressz a felelős.
A második kérdéskört ER és szarkoplazmás retikulum (SR) eredetű mikroszómákon, valamint sejtkultúrákon vizsgánánk. Megfigyeléseink szerint az extravezikuláris redoxpotenciál emelése vagy a transzmembrán redox grádiens megszűnése átjárhatóvá teszi a membránt nagyobb méretű, specifikus transzporterrel nem rendelkező molekulák számára is mind ER, mind SR vezikulákban. Célunk a fokozott permeabilitásért felelős membráncsatorna azonosítása és szerepének vizsgálata az ER eredetű apoptózisban.
Summary
The endoplasmic reticulum (ER) is a sensor for many environmental and internal signals. Factors harmfully influencing the posttranslational modification and folding of secretory proteins in the ER lumen will lead to the accumulation of un- or misfolded proteins and provoke ER stress. The stress activates signaling pathways, causing the inhibition of translation, and a selective expression of lumenal chhaperones and foldases. Long-lasting ER stress usually activates the apoptotic machinery. The mechanism can be provoked not only by experimental agents, but it is present in the pathomechanism of severe human diseases.
The aims of the project are the investigation of (i) the role of redox-active compounds, antioxidants and hepatotoxins in the ER stress and apoptosis, and (ii) the redox-dependent permeability transition of the ER membrane as follows:
According to our preliminary results, ascorbate plays an important role in the electron transfer connected to the oxidative folding of secretory and membrane proteins. Therefore, in vivo ascorbate deficiency (scurvy) may cause ER stress.
The genetic absence of glucose-6-phosphate transporter in the ER membrane of neutrophil granulocytes causes apoptosis. We suppose that the intraluminal redox changes contribute to this effect. We want to study the mechanism of the apoptosis and the intraluminal redox processes.
The majority of hepatotoxins is activated or metabolized in the ER. The process can influence the local redox potential. We suppose that the ER stress is responsible - at least partially - for the apoptotic or toxic effects of these compounds.
The elevation of the cytosolic redox potential, or the disappearance of the transmembrane redox gradient makes the ER and SR membrane permeable towards relatively bulky molecules, which do not have specific transporters. We want to identify the pore or channel responsible for the increased permeability, and to determine its role in the ER-dependent apoptosis.

Az endoplazmás retikulum számos környezeti és metabolikus hatás szenzora. A jelátvitel gyakran a luminális redoxon keresztül valósul meg. Munkánk során azonosítottuk a hexóz-6-foszfát dehidrogenázt, mint a luminális piridin nukleotidok redox státuszának meghatározóját több sejttípusban. A luminális redox fontos a tápláltság érzékelésében, valamint a sejt apotózis/autofágia szabályozásában. A luminális redoxot befolyásoló antioxidánsok, hepatotoxinok, hormonanalógok és más környezeti hatások alapvetően befolyásolhatják a sejt életképességét. A nemzetközi együttműködés keretében elsősorban a hexóz-6-foszfát dehidrogenázt tanulmányoztuk mint szenzort és mint a luminális redox meghatározóját.
Results in English
The endoplasmic reticulum is an important sensor and integrator of environmental and metabolic stimuli. The signaling often involves the changes in luminal redox. We have identified the hexose-6-phosphate dehydrogenase as the main determinant of the redox state of luminal pyridine nucleotides in several cell types. Luminal redox is important in nutrient sensing, and in the regulation of programmed cell death. Antioxidants, hepatotoxins, endocrine disruptors and other environmental agents affecting luminal redox can profoundly alter the viability of the cell. In the frame of the international cooperation hexose-6-phosphate dehydrogenase was studied as a nutrient sensor and as the main regulator of luminal redox.
