Bár jelenleg nagy igény mutatkozik miniatürizált kromatográfiás technikák kifejlesztésére, a szakirodalomban alig találhatunk példát chip-alapú kromatográfiás rendszerekre, különösen az elektroforetikus chipekhez képest. Nemrégiben egy olyan műanyagból (PDMS) készült mikrofluidikai chip kifejlesztéséről számoltunk be, melybe fordított fázisú kromatográfiás (C18) töltetet integráltunk frit alkalmazása nélkül. A töltet egyszerű integrálására a műanyag nagyfokú rugalmassága és hidrofób jellege adott lehetőséget. A chip csatornájában és a töltetben több új hatást és jelenséget figyeltünk meg. Az előkísérletek alapján a kromatográfiás töltet és/vagy feszültség alkalmazása az elválasztás során sokféle típusú vegyület meghatározására lehet alkalmas. Az előállított chipek további tanulmányozását és alkalmazását tervezzük, illetve más, különböző típusú tölteteket tartalmazó és újfajta csatornamintázatú chipek kifejlesztésére és tesztelésére készülünk.
A tervezett kutatás korábbi elektroforetikus és kromatográfiás, illetve az elmúlt időszakban külföldön megszerzett mikrofluidikai, mikrofabrikációs tapasztalatainkon alapszik:
1, új típusú mikrofluidikai eszközök tervezése és előállítása,
2, elválasztások (CE, LC, CEC) tanulmányozása üres és töltött csatornákban, új állófázisok integrálása
3, a kifejlesztett mikrofluidikai eszközök orvosdiagnosztikai, gyógyszer- és környezetanalitikai alkalmazásai.
Munkánk során szeretnénk kihasználni a mikrochipekkel történő munka olyan előnyeit, mint a rendkívül kis mennyiségű minta- (10-500 pL) és oldószer (1-10 μL) fogyasztást, gyors meghatározási sebességet (0.5-2 perc), egyszerű elemzési sokszorosítást (egy chipen több mérőrendszer, kromatográfiás töltet is található).
A tervezett kutatást szoros nemzetközi együttműködésben végezzük.
A támogatás felhasználásának célja nemzetközileg elismert eredményeket produkáló hazai mikrofluidikai analitikai kutatás megalapozása.
Summary
Although there is a high demand for miniaturized chromatographic techniques, numerous recent surveys of the literature showed only a few examples of chip-based chromatographic systems compared to chip-based CE devices. In our recent works we described the fabrication of a PDMS-based microfluidic chip containing reversed-phase silica beads without the use of frits or other barriers. The packing of the silica beads into the microchips is made possible by the hydrophobic nature and elasticity of PDMS. Different retaining and stabilizing effects appearing in the packed channel have been observed. The chromatographic packing of this new type of chips can be utilized to separate a large variety of compounds. Further investigation and characterization of the packing and use of other types of particles are being investigated.

The planned research is based on our experiences and knowledge on electrophoretic and microfluidic research:
1, designing and microfabrication of microfluidic devices,
2, separation (CE, LC, CEC) in open tubular and packed channels, integration of new stationary phases into the chip,
3. application of microfluidic devices in clinics, pharmacology and environment.
Using the developed disposable chips, we would like to utilize the advantages of our microfluidic devices: minute volume of consumed samples (10-500 pL) and reagents (1-10 μL), fast analysis (within 1-2 min), cheap parallelization (2 or more separation units on a single chip).
This research is strongly connected to international cooperations.
The main benefit of the obtained support that a modern microfluidic research internationally recognized can be established in our country.

A mikrofluidikai kutatásaink egyik területe a kromatográfiás elválasztási technikák miniatürizálása, a különféle kromatográfiás töltetek mikrocsipben való kialakítása és alkalmazása volt. A kialakított tölteten kromatográfiás és elektrokromatográfiás elválasztást írtunk le. A mikrofluidika lehetőséget nyújtott arra is, hogy több analitikai rendszer - például 12 kromatográfiás elválasztóegységet - helyezzünk el egyetlen mikrocsipen, így egyidejűleg több kromatográfiás elválasztást is végrehajthattunk. Mikrocsipen történő elektroforetikus elválasztásokhoz hatékony injektálási módszert dolgoztunk ki néhányszor száz pikoliternyi térfogatú folyadékminták mikrocsipbe való injektálásához. Az új, nyomással történő injektálási eljárás kidolgozásával kiküszöböltük az elektrokinetikus injektálásokra jellemző, mennyiségi meghatározások pontosságát csökkentő hibákat. Munkánk során egy olyan mikrocsip alapú kapilláris elektroforézis rendszert fejlesztettünk ki, melynél a mintainjektálás a mikrocsipben, az elektroforetikus elválasztás pedig - hasonlóan, mint a hagyományos CE készülékekben - egy kvarckapillárisban történik. Az általunk kifejlesztett miniatürizált kapilláris elektroforetikus rendszert mikrofluidikai platformok segítségével felületi plazmon rezonancia spektrometriával is kapcsoltuk. A kutatásainkat nemzetközi együttműködésben végeztük. A támogatás felhasználásával nemzetközileg elismert eredményeket produkáló hazai mikrofluidikai analitikai kutatást tudtunk megalapozni.

One of the main field of our microfluidic research was the miniaturization of the chromatographic separation techniques, the preparation of different chromatographic packings and their applications. Several chromatographic and electrochromatographic separations were carried out using the different packings. The microfluidics provides possibility to integrate several analytical systems - for instance 12 chromatographic separation units - into a single microchip, thus several chromatographic separations can be simultaneously performed. For the microchip-based electrophoretic separations an effective injection method was developed for the liquid samples of few-hundred-picoliter volumes to be injected into the microchip. Using this new, pressure-driven injection method, the sampling and matrix biases typical for electrokinetic injection are avoided. In this project we developed a microchip based capillary electrophoretic system, in which the sample introduction was performed in a polydimethylsiloxane microchip, and the electrophoretic separation - similarly as in conventional capillary elctrophoresis instruments - occurs in a fused silica capillary. This developed miniaturized capillary electrophoretic system with microfluidic platforms was hyphenated with surface plasmon resonance spectrometric detection. Our research was carried out in international cooperation. This support obtained from OTKA established a domestic analytical microfluidic research laboratory, which demonstrated internationally recognized results on this field.
