A project célja a globális agyi ischemia következtében meginduló un. késleltetett sejtpusztulás (KSP) mechanizmusát vizsgálni patkány modelleken. Megfigyelték, hogy az agyi ischemia kiváltotta KSP mérsékelhető, ha a súlyos ischemiás állapotot órákkal vagy napokkal megelőzi egy rövid, tranziens ischemiás állapot. A jelenséget ischemiás prekondícionálásnak (IPK), míg az így elért neuroprotekciót ischemia toleranciának (IT) nevezzük. A projekt a KSP folyamatának legkorábbi szakaszára fókuszál, amikor a megemelkedett glutamát (Glu) szint következtében kialakuló excitotoxicitás a leginkább fenyegető jelenség. Azt vizsgáljuk, hogy milyen szerepet játszanak az excitatórikus aminósav receptorok (AMPA, NMDA) az IPK és IT létrejöttében, ill. ezek blokkolóinak adásával milyen mértékű neuroprotekció érhető el, különösen, ha ezeket kombináljuk az IPK legoptimálisabb paramétereivel. Különösen figyelünk az egyik endogén NMDA receptor-blokkoló anyag, a kinurénsav (KYNA) és szarmazékai neuroprotektív hatására. Új, hatékony (vér-agy gáton átjutó) KYNA származékokat is létre fogunk hozni. A neuroprotekció kérdésének teljesen új megközelítését jelenti, amikor az agyi Glu szintet úgy csökkentjük, hogy Glu scavengerek i.v. adásával fokozzuk az agyból az intravasalis térbe irányuló Glu tarnszportot. A pályázat egy széleskörű, már bejáratott hazai és nemzetközi együttműködésen alapul. Részt vesznek benne MSc-s és PhD-s hallgatók, ezzel is elősegítve a kutatói utánpótlás nevelését.
Summary
The aim of this project is to study the phenomenon of brain ischemia-induced (I-I) delayed cell death (DCD) on a global ischemic model of the rat. The level of DCD may be reduced if a sublethal ischemic episode occurs several hours or days before a severe insult. This is known as ischemic preconditioning (IPC) and the neuroprotection induced is called ischemic tolerance (IT). This project focuses on the early period of the I-I events, when the excitoxicity due to the enhanced glutamate (Glu) level is the most threatening phenomenon. We aim to study 1) the roles of AMPA and NMDA receptors in IPC and IT. 2) Special attention will be paid to the endogenous NMDA receptor antagonist kynurenic acid (KYNA) and its derivatives. We plan to develop new neuroprotective KYNA derivatives too. Our earlier results indicate that the probability of success is good. 3) The project additionally includes an absolutely new approach to neuroprotection: the Glu concentration in the brain will be lowered to a level below its excitotoxic threshold through the use of Glu scavengers. We plan collaborative work both with three institutes at our own university and with institutes abroad (in Israel, in Italy, in Slovakia). The project will clarify new details of the mechanisms of IPC and IT, and will result in new neuroprotective compounds which could be used in further preclinical studies. As undergraduates and PhD students will be included among those working on this project, it will contribute significantly to the training of the new generation of researchers.

A projekt fő célkitűzése agyi ischemiás modelleken neuroprotektív stratégiák kidolgozása és új neuroprotektív mulekulák előállítása és kipróbálása volt. A kitűzött célt három különböző módon közelítettük meg: 1) az ischemiás attack-ot követő extrém magas agyi glutamát (Glu) szintet szisztémásan adott glutamát scavengerek alkalmazásával csökkentettük. 2) olyan kinurénsav (KYNA) analógokat szintetizáltunk, melyek könnyen átjutnak a vér-agy gáton, tehát szisztémásan adhatók, ugyanakkor a központi idegrendszerbe bejutva, az NMDA receptorokhoz kötődve, csökkentik az idegszövet-károsító hiperexcitabilitást. 3) a sejtek természetes védekező mechanizmusait aktiváló pre- és posztkondícionálási paradigmákat dolgoztunk ki. A fent részletezett három területen végzett kutatómunka lehetőséget biztosított a különböző neurodegenerítv betegségek pathomechanizmusában a közös biológiai momentumok felismerésére. A projekt keretében végzett munka az elméleti ismereteken túl, a jövőben gyakorlati (gyógyszerfejlesztési ill. klinikai) jelentőséggel is bírhat.
Results in English
The main goal of this project was to develop new neuroprotective strategies and molecules on ischemic brain models. To approach this goal we used three different ways: 1) to reduce the excess glutamate (Glu) levels within the brain following an ischemic attack with administration of glutamate scavengers. 2) To develop such kynurenic acid (KYNA) analogues which are able to cross the blood-brain barrier (BBB) and able to reduce the hyperexcitability by blocking NMDA receptors. 3) To develop such pre- and postconditioning protocols which are able to activate the endogenous self-protective mechanisms resulting in neuroprotection after an ischemic attack. In the course of these studies, we had chance to realize the common elements in the pathomechanisms of different neurodegenerative disorders. Taken together our results, we hope that they may have pharmaceutical or clinical importance in the future.
