Ezzel pályázattal arra a kérdésre keresünk választ, hogy milyen kapcsolat van a gyökérfejlődés és a szárazságadaptáció között fűféléknél. Korábbi munkánk során különböző búza genotípusok szárazságtűrését vizsgálva azt tapasztaltuk, hogy egyes, jól adaptálódó fajták meg tudják változtatni a gyökér/hajtás fejlődés egyensúlyát, fenntartva a gyökérnövekedést és visszafogva a hajtásfejlődésüket korlátozott vízellátás esetén. Ennek a jelenségnek a molekuláris hátterét kívánjuk feltárni a szálkaperje (Brachypodium distachyon) kísérleti rendszeren. A szálkaperje a mérsékeltövi fűfélék új, és dinamikusan fejlődő modellrendszere, szerepe a kétszikűeknél áttörést eredményező lúdfőhöz (Arabidopsis) hasonlítható. Létrehozunk egy olyan kísérleti rendszert, mely alkalmas a szálkaperje gyökérfejlődésének vizsgálatára kontrollált vízellátás mellett. Meghatározzuk száraz és kiegyensúlyozott klímához adaptálódott ökotípusok gyökérfejlődésének jellemzőit. Összehasonlítjuk erős gyökérfejlődésű és érzékeny ökotípusok gyökerének transzkript mintázatát. Élesztő két-hibrid kölcsönhatásra alkalmas cDNS-könyvtárakat készítünk szárazság-stressznek kitett szálkaperje gyökércsúcsából és bokrosodási csomójából. Azonosítunk LOB-domain transzkripciós faktorokat, melyek korábbi eredményeink szerint kapcsolatot jelentenek a gyökérfejlődés és a sejtciklus szabályozása között, és jellemezzük ezeket a szabályozó komplexekkel és más fehérjékkel való kölcsönhatásuk, valamint DNS-kötő sajátságaik alapján.
Summary
Our previous studies on long term drought adaptation of different wheat genotypes revealed that some of the well adapting cultivars can alter the balance of root/shoot development favoring the maintenance of root growth and reducing shoot growth under limited water supply. Molecular background of this phenomenon is to be revealed using Brachypodium distachyon as a new and improving model system for temperate grasses similar to that of Arabidopsis in dicots. Experimental system of Brachypodium is suitable to study root development under controlled water supply will be established. Root growth characteristics of ecotypes adapted to arid and balanced climate will be determined. Transcript profile of roots of strong rooting and sensitive ecotypes will be compared. Yeast two-hybrid (Y2H) cDNA libraries of root tips and crown tissues of Brachypodium subjected to drought stress will be constructed. LOB-domain transcription factors that link root development and cell cycle regulation according to our previous results will be isolated and characterized by their interactions with cell cycle regulatory complexes and other proteins as well as by their DNA binding specificity.

Beállítottunk egy olyan kísérleti rendszert, ami alkalmas arra, hogy a gyökérfejlődést tanulmányozzuk Brachypodiumon. Ez a rhizotronra alapozott rendszer lehetővé teszi, hogy kutatások során tovább vizsgáljuk a kérdést: hogyan alakítják át a füvek, illetve gabonaféléink a gyökérzetük felépítését a korlátozott vízellátáshoz való alkalmazkodásuk során, és melyek azok a génjelöltek, melyek szerepet játszanak a gyökér architektúrát érintő, jól megfigyelhető morfológiai változásokban. Brachypodium genotípusok széles körét teszteltük eltérő szárazságtűrésük szempontjából, és a kiválasztott vonalakat részletesen jellemeztük gyökérnövekedésük és architektúrájuk, valamint általános növekedési paramétereik meghatározásával rhizotronban. Az elvégzett főkomponens-elemzés is megmutatta, hogy az erős és mélyre hatoló elsődleges és nodális gyökerek fontosak a hosszútávú szárazsághoz való jó alkalmazkodáshoz. A gyökér-architektúrát befolyásoló génjelöltek között elsősorban a szálkaperje ún. LOB-domén (oldalszerv-határ meghatározó) transzkripciós faktor (TF) családját tanulmányozzuk. Eddig tizenegy LOB-domén kódoló TF gént jellemeztünk transzkripciós szinten. Különböző szervspecifikus kifejeződését mutattuk ki e géneknek, egyikük kivételesen magas gyökércsúcs specifikus működést mutat. A gén promóterével készült GFP riportergén konstrukció megerősítette ennek a génnek az expresszióját gyökércsúcsban és azt is kimutattuk, hogy promótere aktív a porzószálban is.
Results in English
At first, experimental system, similar to the one used for wheat in recent years, was established to study the root development of Brachypodium. This rhizotron based system enables the continuation of research an the same question: how can grass plants reshape their root system as the consequence of limiting water supply and what are the candidate genes playing role in the changes of the root architecture that we observe on morphological level in genotypes accommodating to drought stress well. Wide range of Brachypodium genotypes differing in drought tolerance was tested, selected lines were characterized in details in rhizotron in terms of root growth and architecture and general growth parameters. Main component analysis revealed that thick and deep growing primary and nodal roots are important for good long term drought adaptation. As candidates of root architecture influencing genes, we study lateral organ boundary (LOB) domain transcription factor family of Brachypodium. To date, eleven genes coding for LOB domain transcription factors were characterized at transcript level. Various organ specificities were found, one of them possesses exceptionally high root tip specificity. The promoter-GFP construct of this gene confirmed the expression in root tip and also revealed promoter activity in stamen.
