Az élő sejtek atomerő mikroszkópos megfigyelése új dimenzióval járult hozzá a biológiai kutatáshoz. Fontos lehetőség, hogy az élő sejtek folyadékban, saját természetes környezetükben figyelhetőek meg. Az alaki megfigyelés mellett erőméréssel a sejt mechanikai tulajdonságai, mint például a rugalmassági állandója határozható meg.
Eddigi kísérleteink során, a endotél sejtek rugalmassági moduluszának a meghatározásakor, az eredmények szokatlanul nagy szórást mutattak. Ennek megértéséhez kezeletlen konfluens sejtkultúrán, több órán keresztül mértük a sejtek rugalmasságát. A hosszú időbázisú méréseink az endotél sejtek rugalmasságában lassú, kb. 150 perces oszcillációt, mutattak, ami magyarázatot ad a rugalmassági modulusz változásaira. Bár élő sejteken többféle elektromos és mechanikai oszcillációt leírtak már, de az általunk megfigyelt lassú mechanikai oszcillációról írott információt nem találtunk. Ezt a jelenséget szeretnénk alaposabban megmérni és megérteni. Az endotél sejtek nagyon érzékenyek külső paraméterek változására. Az oszcilláció jellemzőinek a függését vizsgálnánk ezen külső paraméterek függvényében Az ismeretek felhasználásával élő endotél sejteken, vizsgáljuk különböző gyógyszer hatású vegyületek által a sejten létrejövő alaki és mechanikai tulajdonságok változásait. Ezen kutatások fontosak a gyógyszerek hatásmechanizmusának a vizsgálatában.
Társult kutatásként, tanulmányozzuk különböző antibakteriális peptidek, mint az indolicidin és néhány antioxodáns, mint a quercetin hatását szilárd hordozón létrehozott mesterséges membránokon. Egy másik vizsgálandó biokompozit a szén nanocsövek és fehérjék kölcsönhatásából kialakult elegy.
Summary
The atomic force microscopic study of the living cells gives a new dimension to the biological research. An important achievement that the living cells can be observed in fluid, in their natural environment. Besides observing the shape of the cells, their mechanical properties, such as the elastic modulus can be determined.
During our experiments with living endothelial cell culture the determination of the elastic modulus showed an unusual large scattering. To understand the phenomenon we monitored for several hours the elastic property of an untreated confluent endothelial cell culture. The long time-base measurement performed on endothelial cells, showed a very slow oscillation of the elasticity with about 150 minutes period. This explains the changes of the elastic modulus. Although different electrical and mechanical oscillations were described on living cells, about this very slow oscillation we could not find any information in the literature. We would like to measure and study this phenomenon. The endothelial cells are very sensitive to the change of the external conditions. We will study the change of the characteristics of the oscillation in the function of these external parameters. By the use of these informations we will study on living endothelial cells the change of the shape and mechanical properties during the treatment of the cell culture with drug and other chemicals. The research is important to understand the action of the drugs.
As a joint research we study the effect on solid supported membranes of some antibacterial peptide such as the indolicidin and antioxidant, such as the quercitin. Another biocomposite to study is the mixture of different proteins with carbon nanotubes.

Összefoglaló Fő kutatási témánk, a vér-agy gátat alkotó agyi endotél sejtek mechanikai tulajdonságainak a tanulmányozása atomerő mikroszkóp (AFM) segítségével. Az AFM élő sejtek mechanikai paramétereit képes meghatározni. Endotél sejtek alkotják a mikro-kapilláris vérereket, amelyek áteresztik az érfalon az agy működéséhez szükséges anyagokat és kiszűrik a testidegen vegyületeket. Ezt nevezzük vér-agy gátnak. Meghatároztuk az endotél sejtek sztatikus rugalmasságát, valamint a rugalmasság függését térben és időben. A rugalmasság időben több perces periódussal oszcillál. Két sejtet összenyomva és széthúzva meg lehetett határozni a két sejt között kialakuló adhéziós erőt. Meghatároztuk az endotél és a melanoma sejtek között kialakuló adhéziót és ennek hatását néhány az endotél sejtben levő inhibitorra. Mértük mesterséges és természetes membránok mechanikai paramétereit és antibakteriális peptid hatását ezekre a paraméterekre. Vizsgáltuk néhány nano-biokompozit, mint például a fotoszintetikus reakció centrum szén nanocső keverék tulajdonságait.

Summary Our main research project was the investigation of the mechanical properties of the blood-brain barrier forming cerebral endothelial cells by atomic force microscopy (AFM). This instrument is capable to determine the mechanical parameters of a living cell. Endothelial cells form the microcaplilary blood vessels, which lets through the materials important for the brain and stops the passage of other chemicals. It was determined the static elasticity of the endothelial cell and its spatial and temporal dependence. The time dependence of elasticity showed an oscillating with a periodicity of several minutes. By pressing together and retracting two cells the adhesion force between them could be calculated. It was determined the adhesion force between the endothelial and melanoma cells and its effect on several inhibitors. The mechanical parameters and the effect of the indolicidin, an antibacterial peptide, were measured on artificial and natural membranes. Also several nano-biocompozit, such as the mixture of photosynthetic reaction center and carbon nanotube was investigated.
