Az elmúlt két évtizedben vált ismertté, hogy a FXIII-A szerepe nem korlátozódik a véralvadásban betöltött funkciójára, hanem intracelluláris transzglutaminázként is működik a monocytákban és makrofágokban. Legutóbbi vizsgálatainkban a monocyták makrofággá való differenciálódásának folyamatát követve azt tapasztaltuk, hogy a FXIII-A expresszió az alternatív makrofág aktiváció markere. A dermatohisztopatológiában a dermális makrofágok és dendritikus sejtek (melyek – legalább részleges - monocyta-eredete elfogadott) azonosításának számos diagnosztikai vetülete van, de a leletek interpretálása nem egységes. Ezért megalapozott az a feltételezés, hogy a FXIII-A immunhisztokémiai kimutatásának, mint az alternatív úton aktiválódott makrofágok marker reakciójának diagnosztikai értékesítése új lehetőséget teremthet bizonyos bőrgyógyászati betegségek osztályozására. Jelen projekt keretében normál és FXIII-A deficiens monocytákból differenciálódó dendritikus sejtek gén expressziós profiljának összevetését és a FXIII-A pozitív sejtek karakterizálását tervezzük különböző malignus és nem-malignus bőrgyógyászati betegségek kialakulása és progressziója kapcsán. A vizsgálatok eredményeként egyrészt a FXIII-A monocyta/makrofág/DC differenciócióban, másrészt a bőrgyógyászati kórképek kialakulásában betöltött szerepének jobb megismerésére nyílik lehetőség.
Summary
Over the last two decades it became known that the role of FXIII-A is not restricted to the area of haemostasis but it also acts as an intracellular transglutaminase in monocytes and macrophages. In our most recent study designed to monitor the changes in FXIII-A gene expression during differentiation of monocytes into macrophages we found that FXIII-A expression is a marker for alternatively activated macrophages. In dermatohistopathology identification and characterization of dermal macrophages and dendritic cells (DCs) supposed to have – at least partly – the same monocytic origin has significant diagnostic approach, but severe interpretation difficulties. Consequently using the immunoreaction for FXIII-A as a marker for alternatively activated macrophages opens new vistas to cluster dermatological diseases upon the activation status of the macrophages. In the framework of this project proposed we are planning to compare the gene expression profiles of normal and FXIII-A deficient monocyte-derived DCs, as well as to see how FXIII-A positive cells are labelled for different DC/macrophage markers and distributed in different malignant and non-malignant skin lesions at stages of their development and progression. This comparative analysis provides a unique opportunity not only to characterize the repertoires of genes expressed differently in the presence and absence of functionally intact FXIII-A during differentiation of monocytes into DCs, but to create attractive hypothesis about the possible role of FXIII-A in the development of skin diseases.

Vizsgálataink a XIII-as véralvadás faktor A alegysége (FXIII-A) gén expresszióban játszott szerepének és immunhisztokémiai kimutatása markerrekcióként való értékesítéshetőségének jellemzésére irányultak. IL4 jelenlétében differenciáltatott normál és FXIII-A deficiens humán monociták/makrofágok microarray analízissel nyert gén expressziós profiljának összevetésével megállapítottuk, hogy a vizsgált 24398 gén-szekvencia közül a deficiens sejtek esetében 1017, míg a normál sejtek esetében 596 szekvencia expressziója volt IL4-szenzitív, melyek közül 307 a normál és a deficiens sejtekben azonosnak bizonyult. A Cytoscape/BiNGO és Ingenuity Pathways Analysis programokkal elvégzett "functional clustering" analízis szerint a domináns génexpressziós eltérések az immunregulációban és a sebgyógyulásban involvált géncsoportokban voltak észlelhetők. Granuloma annulare (GA), necrobiosis lipoidica (NL) valamint szemhéji xanthelasma szöveti mintákon kettős jelzésű immunhisztokémiai rendszerekben megállapítottuk, hogy a GA és NL betegségekben a CD163+ makrofágokat FXIII-A pozitivitás jellemzi (CD163+/FXIII-A+) szemben a xanthelasma FXIII-A negatív (CD163+/FXIII-A-) makrofágjaival. GA és NL esetében a CD163+ és CD11c+ sejtek teljesen elkülönülő populációkat alkottak. A FXIII-A kettős jelölése ezen markerekkel egyértelműen megkülönbözteti a makrofágokat (CD163+/FXIII-A+) a dedritikus sejtektől (CD11c+/FXIII-A-), ami a dermatohisztológiai diagnosztikában jól értékesíthető.
Results in English
The objectives of our studies were to characterize the possible role of FXIII-A in the regulation of gene expression and to define whether the immunohistochemical detection of FXIII-A can be used as a marker reaction. Microarray analyses carried out on DNA samples of monocytes/macrophages from FXIII-A-deficient patients and healthy controls after a 48-hour differentiation in the presence of IL4 1017 probes out of the 24398 expressed in macrophages from FXIII-A- deficient samples were IL4 sensitive, while only 596 probes were IL4 sensitive in wild-type samples. Of these genes, 307 were induced in both the deficient and the wild-type macrophages. Functional clustering of the target genes carried out using Cytoscape/BiNGO and Ingenuity Pathways Analysis programs showed that, in the absence of FXIII-A, the most prominent differences are related to immune functions and to wound response. In our studies on samples of granuloma annulare (GA), necrobiosis lipoidica (NL) and palpebral xanthelasma (PX) by using double labelling immunohistochemical reactions it was demonstrated that in GA and NL samples CD163+ macrophages are labeled for FXIII-A while in PX they are negative for FXIII-A. In GA and NL CD163+ and CD11c+ cells represent two distinct populations. FXIII-A positivity clearly distinguished macrophages (CD163+/FXIII-A+) from dendritic cells (CD11c+/FXIII-A-), which makes the immunohistochemical detection FXIII-A a valuable tool in diagnostic dermatopathology.
