Itt írja le a kutatás fő célkitűzéseit a témában jártas szakember számára.
A fotoszintetikus szervezetek különböző struktúrák és mechanizmusok közreműködésével védekeznek a túlzott fénysugárzás káros hatása ellen. A fotoprotekció legfontosabb módja az abszorbeált fény hő-disszipációja (TD, thermal dissipation) a fénybegyűjtő komplexekben (LHC), a gerjesztési energia ill. a fluoreszcencia nem-fotokémiai kioltása. Az LHC-ben a TD szabályozása összetett, és számos, egymással időben átfedő, visszaható mechanizmust tartalmaz, amelyekben valószínűleg különböző LHC szubpopulációk vesznek részt. A közelmúltban világossá vált, hogy a növényekben a TD aktiválása in vivo az LHC-k makroszerkezeti átrendeződével jár. A TD önmagában is okozhat makroszerkezeti változásokat (ez az úgynevezett termo-optikai hatás). A pályázat célja, hogy feltárja a makroszerkezeti változások és a fotoprotekciós TD kapcsolatát növényekben és algákban és egyúttal tisztázza a különböző, az irodalomban javasolt fényvédő mechanizmusok élettani szerepét. Ezek a következők: a hő disszipációja a PS II antennában, az LHC-k leválása a reakciócentrumokról és fotoprotektív töltésrekombináció a reakciócentrumokban. E célból pikoszekundumos feloldású és spektrálisan bontott fluoreszcencia mérések segítségével növényi és alga modell rendszereken kinetikai modellszámításokat fogunk végezni, hogy megismerjük a különböző módon aktiválódó disszipációs utakat és ezek hozzájárulását a fotoprotekcióhoz. Ezzel párhuzamosan cirkuláris dikroizmus spektroszkópiával és a kisszögű neutronszórással nyomon követjük az LHC és a membránrendszer szerkezetváltozásait in vivo, hogy választ kapjunk arra, hogy ezek a szerkezetváltozások milyen összefüggésben vannak egymással és a TD-val.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.
• Milyen időskálán zajlanak az LHC makrostruktúrális változásai, aktiválódása és relaxációja, ill. milyen kapcsolatban van ez a nem-fotokémiai fluoreszencia kioltás kinetikai komponenseivel (qE, qT, qI)?

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!
A fotoszintetizáló szervezetek - köztük is túlnyomó többségben növények és algák – látják el közvetve vagy közvetlenül energiával a Föld minden élő szervezetét. A fotoszintézis a légköri szén-dioxid kivonásával hűti a klímát és biztosítja a táplálékot a bioszféra valamennyi élőlénye számára; ez fedezi az emberiség által felhasznált energia nagy részét is. Bár a fosszilis tüzelőanyagok használatát csökkentenünk kell, továbbra is megmarad az igény a fotoszintetikusan termelt energiára pl. bioüzemanyag formájában, és a napenergia átalakítás mesterséges és biomimetkus rendszerek termékei révén is. A fotoprotekció nélkülözhetetlen a fotoszintetizáló szervezetek számára, de nem kevésbé fontos a mesterséges rendszerek esetében - a tartósság és a fenntarthatóság megoldása több rendszerben nem megoldott. A pályázat alapvető tudományos problémáinak a megválaszolása hozzá fog segíteni a fotoszintézis összetett szabályozásának a pontosabb megértéséhez és a fotoprotekció mechanizmusának felderítéséhez. A projekt megvalósítása során egy felhasználó-barát időfelbontásos fluoreszcencia spektroszkópiás mérési technika rendszerbe állításával jelentősen bővítjük azt a technikai arzenált, amivel a növények élettani funkcióit vizsgálni tudjuk. A nagy léptekkel haladó technológiai fejlesztések a félvezető lézerek, az elektronika és a számítástechnikai terén magában hordozza a lehetőségét annak, hogy rutinszerűen alkalmazhatóvá válik ez a mérési módszer a növénybiológusok részére is, kiegészítve a jelenleg használatos, lényegesen kevesebb információt szolgáltató hagyományos klorofill fluoreszcencia mérési technikákat. A Szegedi Biológiai Kutatóközpontban íly módon rendelekezésre álló ’bench-top’ ultragyors időfelbontásos fluoreszcens spektrofotométer a pályázat megvasósításán túl rendelkezésre fog állni más projektekben felmerülő problémák megoldásához is.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.
Az életet a Földön a Nap táplálja. A fotoszintetizáló szervezetek, növények és algák, a napfény-energia felhasználásával széndioxidból szénhidrátot szintetizálnak. A bioszféra energiaháztartása erre az alapfolyamatra épül. A fotoszintézis hozta létre a Föld oxigénben dús atmoszféráját és a fosszilis energiaforrásokat is. A fotoszintetikus fényenergia átalakítás pontos megértése hozzásegíthet bennünket az alternatív, környezet-kímélő energia-forrás technológiai kifejlesztéséhez.
Az úgynevezett fénybegyűjtő antennák segítségével a növények képesek a fényenergiát igen hatékonyan összegyűjteni. Ezen folyamat közben, magas fényintenzitáson a ’felesleges, fotoszintetikusan nem hesznosítható gerjesztési energia tönkreteheti magát az energiaátalakító rendszert és az organizmust is megölheti. Ezt elkerülendő a növények rendelkeznek aktív szabályozó, fotoprotektív mechanizmussal, mely gyorsan reagál, ha túl sok a fény – pl. a fénybegyűjtő antenna átkapcsol ”disszipációs” üzemmódra, ami során az energia gyorsan és biztonságos módon képes hővé átalakulni. Tanulmányozni fogjuk ennek a folyamatnak a mechanizmusát, a szerkezeti változások szerepét és természetét a fotoprotekciós rendszer működése során, különböző modern biofizikai, biokémiai és molekuláris biológiai módszer együttes alkalmazása segítségével különböző szervezettségű rendszereken. Poláros fény vagy neutron sugár segítségével képesek leszünk az energiaátalakító rendszer molekuláris és kiterjedt membrán szerkezetét tanulmányozni; időfelbontásos fluoreszcencia spektroszkópia alkalmazásával pedig követni tudjuk azokat az ultragyors energiadisszipációs folyamatokat is.

Photosynthetic organisms possess various systems and mechanisms to protect themselves against harmful effects of excess light irradiation. A key photoprotective process is the thermal dissipation (TD) of absorbed light energy in the light-harvesting antenna complexes (LHC), often referred to as non-photochemical fluorescence quenching. The regulation of TD in LHCs itself is complex and is driven by several feedback mechanisms overlapping in time and probably affecting different subpopulations of LHCs. Recently it has become clear that the activation of TD in plants involves macrostructural rearrangements of the LHCs in vivo. Conversely, TD itself may cause macroorganization changes (so-called thermo-optic effect). This project aims to elucidate the relationship between macroorganization and photoprotective TD in plants and algae. We plan to clarify the physiological roles of different proposed photoprotective mechanisms: TD of the Photosystem II antenna, functional decoupling of the LHCs from the reaction centres, photoprotective charge recombination in the reaction centres. For this purpose we perform picosecond time- and wavelength-resolved fluorescence experiments with plant and algal model systems in different conditions and apply kinetic modeling analysis to detect the activation of different energy dissipation pathways and to evaluate their contribution to actual photoprotection. In parallel we will monitor the macroorganization of LHC in the photosynthetic membranes by circular dichroism spectroscopy and small-angle neutron scattering in order to answer the question of how LHC macrorganization and membrane ultrastructural changes and TD are interrelated.

What is the significance of the research?

Photosynthetic organisms, the vast majority of them represented by plants and algae, are the ultimate source of energy for all living organisms on Earth. Photosynthesis sequesters greenhouse carbon from the atmosphere, cools the climate, and ultimately provides all the energy and food for all living organisms, and most of the energy for human activities. Mankind is posed to decrease the usage of fossil fuels for its energetic needs but will still rely strongly on photosynthesis either as biofuels or as solar energy conversion by artificial and biomimetic systems.
Photoprotection is crucial for living photosynthetic organisms but not less important for artificial systems where durability and sustainability is currently a major setback in some systems. Answering the basic scientific problems of this project will drive further our knowledge and understanding of the complex regulation of photosynthesis and shed more light on the mechanisms of photoprotection. In the process of getting these answers we will continue the development of our methodology for probing the functional state of plants by time-resolved fluorescence spectroscopy. With the booming technological advancements in solid-state lasers, electronics and computational power, this methodology has the potential to become routinely available to plant scientists, far surpassing the reliability and information content of the current ubiquitous chlorophyll fluorescence methods. The bench-top ultrafast time-resolved fluorescence instrument that will be installed in the BRC will most certainly be of special value for many other areas of research.

Summary and aims of the research for the public

Life on Earth is driven by energy from the Sun, which lets photosynthetic organisms, in particular plants and algae, to produce sugar from carbon dioxide. Practically all living organisms directly or indirectly rely on this basic process for their energetic needs. Photosynthesis has also produced the oxygen in Earth’s atmosphere and the fossil fuels used by mankind. Plants have solved their energy problem and so can we, if we fully understand the inner workings of their remarkable energy-converting machinery.
With the help of the so-called light-harvesting antenna proteins, plants can capture light very effectively. But this poses a serious challenge – too much light is dangerous and when the photosynthetic capacity is exceeded, the unused light energy can destroy the machinery and kill the organism. That is why plants are equipped with actively regulated photoprotection systems that are quickly engaged when light is too much – for example, the light-harvesting antenna switches into “dissipative” mode, whereby the energy is quickly and safely converted into heat. By a combination of advanced biophysical, biochemical and molecular biology techniques we will study the mechanisms of this process, in particular the structural changes controlling the photoprotective switch in different organisms and at different levels of structural complexity. Using polarized light or neutron beams we can probe the molecular structure of the light harvesting antennae and the membrane ultrastructures in leaves and algae, while with time-resolved fluorescence spectroscopy we will follow the extremely fast processes of energy dissipation, occurring in less than one billionth of a second.

A projekt során részletesen tanulmányoztuk a fotoszintetikus fénybegyűjtő antennák (LHA) gerjesztési energijának hőleadására (TD) fordított biofizikai folyamatait, kapcsolódva ezeknek a folyamatoknak a fényvédelemben és fényadaptációban betöltött szerepéhez, figyelembe véve az LHA fotoszintetikus membránjainak bonyolult összefüggését TD és a makroorganizáció között. Ebből a célból szisztematikus vizsgálatokat végeztünk LHA komplexeken, illetve izolált mintákon TD vagy nélküle indukálódó folyamatokban, rekonstituált komplexeken, mesterséges membránokon, és pigment cserélt komplexeken hajtottuk végre. Strukturális és makrostrukturális szempontokat figyeltünk meg abszorpciós, lineáris és cirkuláris dikroizmus spektroszkópiai, ezen felül a kis-szögű neutron szórásos, atom-erő mikroszkópiai és elektronmikroszkópiai vizsgálatokkal. A rendszerek funkcionális tulajdonságait vizsgáltuk, figyelve az energiaátviteli dinamikára, fotokémiai és TD méréseket végeztünk, amely vizsgálatoknál ultragyors tranziens abszorpciós spektroszkópiai, többdimenziós elektronikus spektroszkópiával és kinetikai modellezéssel kapott adatokat vizsgáltunk. Használtunk még fluoreszcens spektroszkópiai módszereket, ezt a berendezést a projekt keretein belül terveztük meg. Részletes bizonyítékokat szereztünk a makroorganizáció és a TD-kétirányú kapcsolatáról, valamint betekintést nyertünk az LHA-ban lejátszódó energia- átadási és veszteségi utak dinamikájába illetve fizikai mechanizmusokba és utakba.

The project revealed details on the biophysical process of thermal dissipation (TD) of excitation energy in the photosynthetic light-harvesting antenna (LHA) in the context of its role in photoprotection and light adaptation focusing on the intricate relationship between TD and macroorganization of the LHA in photosynthetic membranes. To this end we performed systematic studies on LHA complexes from plants and algae in their native membranes or in isolated states with or without inducing TD (e.g. by light or controlled aggregation), reconstituted complexes, artificial membranes, and complexes with exchanged pigments. Structural and macrostructural aspects were monitored by absorption, linear dichroism and circular dichroism spectroscopy, and additionally by small-angle neutron scattering, atomic-force microscopy and electron microscopy. The functional properties of the systems under study, especially regarding the pathways and dynamics of energy transfer, photochemistry and TD, were examined by steady-state and time-resolved fluorescence spectroscopy, the latter performed with equipment planned and built within the project, as well as ultrafast transient absorption spectroscopy, multidimensional electronic spectroscopy and kinetic modeling. Detailed evidence for the two-way relationship between macroorganization and TD was obtained as well as insights into the physical mechanisms, pathways and dynamics of transfer and dissipation of energy in the LHA.
