A kutatás összefoglalója, célkitűzései szakemberek számára
Itt írja le a kutatás fő célkitűzéseit a témában jártas szakember számára.
A biofizika és molekuláris biológia újabb eredményei arra utalnak, hogy a mechanikai erők biológiai szerepe sokkal fontosabb, mint azt korábban gondoltuk. Jóllehet a támasztó, kötő- és kontraktilis szövetek mechanikai szerepe régóta ismert, mára egyértelművé vált, hogy a mechanikai erők fontos folyamatokat szabályoznak a celluláris, molekuláris, sőt az intramolekuláris szerveződési szinteken is. Az azonban továbbra sem ismert pontosan, hogy a mechanikai erők milyen mechanizmusokon keresztül fejtik ki celluláris és molekuláris hatásaikat. A mechanikai erők biológiai jelentőségének felismerésével létrejött egy új tudományág, a mechanobiológia, amelynek célja a mechanikai erők hatására fellépő biológiai válaszreakciók mechanizmusainak feltárása. Pályázatunk célja a mechanikai erők szerepének vizsgálata a hierarchikus biológiai szerveződés különböző szintjein, az egyedi molekuláktól a szupramolekuláris, celluláris és szöveti rendszereken át az élő szervezetig. Kísérleti céljaink eléréséhez olyan modern technológiákat alkalmazunk, amelyekkel lehetőség nyílik egyedi molekulák és szupramolekuláris rendszerek vizualizálására és nanomechanikai manipulálására, továbbá kontraktilis szövetek szerkezetének és működésének élő szervezetben való követésére. Model rendszerként a harántcsíkolt izom citoszkeletális rendszerét vizsgáljuk.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.
Általános célkitűzésünk a kontraktilis citoszkeleton szerkezetének, dinamikájának, mechanikájnak és kölcsönhatásainak vizsgálata a biológiai hierarchia számos szerveződési szintjén, a szubmolekuláris és egyedi molekula szinttől egészen az élő organizmusig. Kutatási kérdéseink ebből az általános célkitűzésből adódnak:
A titin óriásfehérje alacsony mechanikai stabilitású doménjei befolyásolják-e a molekula extenzibilitását a fiziológiás erő- és időtartományokban?
Milyen mechanizmusok húzódnak a natív titin komplex, erővezérelt gombolyodási trajektóriája hátterében?
A titin domének milyen szerepet játszanak a fehérje komplex erőválaszának kialakításában?
A titin globális szerkezeti dinamikája milyen összefüggésben áll a specifikus doménszerkezeti elemekkel?
Mi a bipoláris miozin vastag filamentum polimerizációs mechanizmusa?
Hogyan kooperálnak a miozin motorfehérjék egy kollektív környezetben?
Milyen tényezők befolyásolják az aktin filamentum diffúziós (reptációs) tulajdonságait?
Az aktin filamentumon belül mekkora a monomérek mechanikai stabilitása?
A titin PEVK domén képes-e szabályozni az aktin filamentumok polizerizációját/depolimerizációját?
Milyen a dezmin intermedier filamentum exakt erőválasza egyedi molekula szinten?

Mi a kutatás jelentősége?

Mechanobiológiai pályázatunk jelentősége a sokrétűségben és hierarchiában rejlik. Új metodikák alkalmazásával egy közös kérdésre keressük a választ a biológiai hierarchia minden szintjén: milyen szerepet játszik a mechanikai erő a biológiai folyamatokban? Eredményeink fontos biológiai jelenségek pontosabb megértéséhez vezethetnek. Projektünk tématerületei a modern sejt- és molekuláris biológia, biofizika és orvosbiológia legintenzívebben tanulmányozott területei közé tartoznak. Kutatásunk fontos szerepet játszhat az izom mechanobiológiájának megértésében és az általános mechanobiológiai elvek lefektetésében, továbbá hozzájárulhat új diagnosztikai és nanotechnológiai eszközök kifejlesztésében. Kísérleteink mintegy melléktermékeként bizonyos innovatív technológiák kifejlődésére is lehetőség adódhat. Ilyen technológia a “kontraktoszóma”, amely széles alkalmazásra kerülhet kompartmentalizált biomolekuláris rendszerek in vitro vizsgálatában. A harántcsíkolt izomszerkezet in vivo vizsgálatára optimalizált GRIN-lencse alapú mikroendoszkópia a jövőben akár humán biomedicinális alkalmazásra is kerülhet.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.
Bár bizonyos biológiai szövetek, például az izom- és csontszövet mechanikai szerepe régóta ismert, csak az utóbbi időben ismertük fel annak jelentőségét, hogy a mechanikai erők számos biológiai és biokémiai folyamatot szabályoznak. Ilyen folyamatok a szöveti fejlődés és differenciáció, és patológiás elváltozások kialakulása. A mechanobiológia egy nemrégiben kifejlődött tudományág, amelyet éppen a mechanikai erők biológiai folyamatokban betöltött szerepének jobb megértése hívott életre. Kutatási tervünk a mechanikai erők szerepét és mechanizmusait kívánja a biológiai hierarchia számos szerveződési szintjén, kezdve egyedi molekuláktól egészen az élő szervezetig. Különleges kutatási eszközpark alkalmazásával vizsgáljuk a biomolekuláris és szöveti rendszereket. Kutatásunkban olyan analitikai módszerek kifejlesztésére is sor kerül, amelyek később akár orvosbiológiai alkalmazásokra is kerülhetnek.

Summary of the research and its aims for experts
Describe the major aims of the research for experts.
Recent observations of biophysics and molecular biology have revealed that mechanical forces play a much more important and significant role in biology than earlier thought. While the mechanical properties of certain connective and contractile tissues have long been recognized, it is now clear that mechanical forces regulate important processes at the cellular, molecular and even intramolecular levels as well. Nevertheless, the exact mechanisms of how mechanical forces exert their effects at the molecular and cellular level are very far from being fully understood. In recognition of the biological importance of mechanical forces mechanobiology, an interdisciplinary subject area dealing with the mechanisms of biological response to mechanical forces has started to emerge. Our proposal aims at investigating the role of mechanics across the biological hierarchy, ranging from individual molecules to the living organism. To reach our objectives we plan to employ an array of novel methodologies that enable us to visualize and manipulate single biomolecules, supramolecular complexes and cells, and allow us to take a peak into the inner structure of the contractile tissues of the living organism. As a model system we are investigating striated muscle as it best displays the structural hierarchy of biological organization and because the role of mechanics in this tissue is clearly determinant for its function.

What is the major research question?

Our general aim is to explore the structure, dynamics, mechanics and interactions of the contractile cytoskeleton at various levels of the biological hierarchy, ranging from the level of individual molecules and submolecular structures to the level of living tissue and organism. Stemming from this general aim, our specific questions and hypotheses are the following:
Do low-force unfolding domains play a role in titin mechanics in physiologically relevant time and force regimes?
What are the mechanisms behind the complex, force-ddriven refolding trajectory of full-length titin?
What is the role of different domains in the detailed force response of titin?
How is the global structural dynamics of titin connected to specific structural regions?
What are the mechanisms of bipolar myosin thick filament formation?
How do myosin motors operate in a collective environment?
What factors influence the diffusive dynamics of actin filaments?
What is the mechanical stability of actin monomers within the filament?
Does PEVK influence the polymerization/depolymerization dynamics of actin?
What is the exact force response of a single desmin filament across its cross-sectional area?
How are desmin mechanics revealed in the mechanical behavior of the entire muscle fiber?
What are the mechanisms of PEVK-actin interaction in situ?
What are the manifestations of interactions between contractile proteins within a microscopic compartment?
What is the operating physiological sarcomere length range? Is thick-filament structure influenced by loading and extension conditions of the sarcomere?

What is the significance of the research?

The major significance of our mechanobiology project lies in its multifaceted and hierarchical nature. By using novel methodologies we address a common question at nearly all levels of the biological organization hierarchy. The results may provide a significantly better understanding of important biological phenomena. The project research areas are of prime importance and represent intensely investigated fields of current cellular and molecular biology, biophysics and biomedicine. Furthermore, our proposal represents a very important step towards a better understanding of the mechanobiology of striated muscle in specific, and the principles of mechanobiology in general. As such, the project may have far-reaching consequences towards implementing improved therapeutics and novel nanotechnology devices. In the end, as a by-product, certain innovative technologies are expected to be established. The liposome-based “contractosome” is clearly such, as it can be used as a model system for the investigation of compartmentalized supramolecular systems. The highly optimized GRIN-lens-based in vivo imaging system for exploring the internal structure of skeletal muscle during contraction in vivo may eventually make it towards human biomedical utilization. Thus, the significance of our project extends beyond expanding our fundamental knowledge about mechanobiology, and may lead towards the invention of novel technologies as well.

Summary and aims of the research for the public
Describe here the major aims of the research for an audience with average background information. This summary is especially important for NRDI Office in order to inform decision-makers, media, and others.
Although the mechanical properties of certain biological tissues such as muscles and bone have long been recognized, it is now clear that mechanical forces regulate a multitude of other biological and biochemical processes, organ development and pathological conditions as well. Mechanobiology is a subject area that has recently emerged in need of well-focused research to explore the mechanisms and the implication of mechanical forces in biology. Our proposal aims at exploring the effect of mechanical forces on the biological system at several levels of its hierarchical organization: from single biomolecules to the living organism. We are employing a vast array of novel technologies in sophisticated experiments to achieve our experimental objectives. Our research is highly important as it lends to hope of revealing novel insights into biological mechanisms. Furthermore, new analytical tools will be developed along the way which may find their future application in biomedicine.

Pályázatuk utolsó, záró évében elsősorban projektjeink lekerekítésére és eredményeink közlésére fókuszáltunk. Összességében a pályázati időszak igen termékeny volt, és számos fontos felfedezést tettünk a harántcsíkolt izom citoszkeletális rendszeréről, és ezen belül különösen a titin óriásfehérje biofizikai tulajdonságairól. A titinben alacsony, fiziológiás erőknél kitekeredni képes doméneket fedeztünk fel; egy újonnan fejlesztett módszer, a meniszkusz erővel történő molekulanyújtás segítségével vizualizálni tudtuk a titin mentén kitekeredett doméneket; felfedeztük, hogy a titin képes erőkifejtésre a gombolyodó doménjei által; végül leírtuk, hogy a titin, a feltételezésekkel ellentétben, nem egy egyszerű geometriai templátja a miozin vastag filamentumnak, hanem annak hosszát dinamikus mechanizmussal szabályozza. A fenti eredmények mellett számos olyan kísérlet történt, egyéb biomolekuláris rendszereken (nanoskálájú lipid rendszerek, DNS, fibrin, amiloid fibrillum, bakteriofágok), amelyek hozzájárultak egymolekula biofizikai és nanotechnológiai metodikai arzenálunk továbbfejlesztéséhez.

In the final year of the proposal much attention has been devoted to wrapping up several projects and publishing our results. Altogether, throughout this grant proposal quite significant discoveries have been made about the muscle cytoskeleton, particularly about the biophysical behavior of the giant protein titin. We discovered domains in the molecule that unfold at very low, physiologically quite relevant forces; a novel method of meniscus-force molecule-stretching allowed us to visualize the location of the domains unfolded by mechanical force; we discovered that titin is able to generate force by the folding of its domains; and we uncovered that titin is not a geometrical template of the thick filament but is likely to regulate thick-filament length by a dynamic mechanism. In additon to our focus strictly on sarcomeric proteins, we also explored addional, related nanoscale systems which allowed us to develop novel methodologies. Accordingly, we explored the behavior of nanoscale lipid systems (liposomes and cochleates), the elasticity and high-resolution structure of filamentous biomolecules (DNA, fibrin, amyloid fibrils), and the nanomechanical behavior of bacteriophages.
