A kutatás összefoglalója, célkitűzései szakemberek számára
Itt írja le a kutatás fő célkitűzéseit a témában jártas szakember számára.
A projekt célkitűzése olyan jelátvitel pályák azonosítása, melyek az I. és III. típusú interferonokkal (IFN) és más oldott faktorokkal együttműködve képesek a vírus és baktérium ellenes immunválasz kimenetelét befolyásolni. Figyelembe véve a vírusfelismerés által kiváltott molekuláris folyamatok összetettségét, vizsgálatainkban a vírus felismerésben kiemelt szerepet játszó retinsav indukált gén I (RIG-I) által szabályozott jelpályák együttműködését vizsgáljuk. Célunk az egymást segítő és/vagy gátló folyamatok molekuláris hátterének feltárása és azonosítása. Hipotézisünk szerint a mintázat felismerő receptorként (PRR) működő RIG-I specifikus (5’ppp) stimulálása DC-ekben sejtekben az aktivált jelpályák és a szecernált citokinek révén egymásra kölcsönösen ható folyamatokat indít el. Ez a párhuzamos jelpályák együttműködése vagy egymást gátló hatása révén is érvényesülhet és befolyásolhatja a sejtválasz kimenetelét. Mivel a mintázat felismerő receptorok kifejeződése csak bizonyos sejttípusokra, köztük a mieloid és a plasmacitoid dendritikus sejtekre korlátozódik, míg a citokinek és interferonok szekréciójára a szervezet számos sejttípusa is képes, az oldott faktorok által közvetített szabályozás az aktiválódó sejtekre is hat. Ez a működési elv különösen fontos az interferon termelés szabályozásában, mivel interferon stimulált gének aktivációját is eredményezi. A tervezett vizsgálatok kutatási eredményei várhatóan a RIG-I szerű receptorok új, eddig nem ismert funkcionális sajátságainak feltárását és az interferonok gyulladási folyamatokra kifejtett gátló és/vagy stimuláló hatásainak szabályozására deríthetnek fényt.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.
A kutatás alapkérdése az, hogyan módosítják az I és III típusú interferonok a különböző sejttípusok funkcionális sajátságait. Ennek feltárására vizsgáljuk az IFNβ fokozó és gátló hatását bél epitél sejtekre és tanulmányozzuk, hogy a human DC-ek polyI:C stimuláció hatására milyen módon váltanak ki ellentétes hatást a RIG-I és MDA5 kifejeződésére. Az mTOR szerepét a polyI:C-vel indukált antivirális és gyulladásos válasz során az mTOR stimuláló és gátló hatása alapján human DC altípusokban vizsgáljuk. A TLR és RLR együttműködés során egymás ellen ható szabályozást is feltételezünk, amit a RIG-I kifejeződés emelkedése mellett a TLR3,7/8,9 és szelektált kommenzális baktériumok együttes adása után a PRR expresszió mérésével vizsgálunk. A reguláló faktorok expressziós mintázatát a mikrobiális stimulációt követően az 5’ppp-dsRNA aktiváció hatására módosult RIG-I és citokin expresszió segítségével követjük. A baktérium indukált moDC stimuláció során az I típusú IFN és PRR jelátvitel elkülönítését az aktivált útvonal komponensek és a foszforilált transzkripciós faktorok azonosításával végezzük. Nagy áteresztő képességű protein array segítségével mérjük a TLR/RLR receptorok kifejeződését és az interferon stimulált gének (ISG) szintjét. A vizsgálati eredmények az IFNAR1 és PRR útvonalak jelátviteli eseményeinek részletes feltérképezését teszik lehetővé, ami az immunrendszer manipulálására alkalmas stratégiák citokin szintű tervezésének lehetőségét rejti magában.

Mi a kutatás jelentősége?

A Toll-szerű receptorok kulcs mikrobiális érzékelők, melyek stimulálják a természetes immunitás védelmi programjait. A TLR3 ligandum kötődése sok sejtben I típusú interferonok termelését váltja ki, míg a TLR7,8,9 receptorok cDC és pDC sejtekben I típusú I IFN szekréciót váltanak ki, ami az IFN kiemelt szabályozó szerepét igazolja. A pályázat fő kérdése az, hogyan vesznek részt az I és III IFN a vírus ellenes immunválasz szabályozásában. A projekt pDC és cDC sejtekben az RLR-ok általi vírus felismerés következményeinek vizsgálatára összpontosít az IFN függő és független útvonalak jelátviteli pályáinak elemzésével. A vírusok felismerésére több specifikus receptor szakosodott, melyek a sejten belül a vezikuláris membránban vagy a citoszólban helyezkednek el. A RIG-I receptorok a citoplazmában található, dupla vagy szimpla szálú RNS és DNS felismerésére képesek. A RIG-I receptor specifikus liganduma az 5’ppp és 5’ppRNA. A receptor általi felismerés során a vírust hordozó sejtekből nagy mennyiségben szabadulnak fel I és III típusú interferonok, melyek rövid távú védelmet nyújtanak a további fertőzéssel szemben, miközben számos sejttípusra hatva befolyásolják a sejtválasz kimenetelét. Az interferon válasz egyedi sajátsága, hogy a vírus felismerést követően számos interferon stimuláló gén is (ISG) aktiválódik, melyek szintén rendelkeznek antivirális aktivitással és termelődésük visszahat a saját és a környező sejtekre. Bár az interferon termelés a vírus eliminációt szolgálja, az interferonok túlzott, nem szabályozott szekréciója számos, köztük az immunrendszer sejtjeire hatva autoimmun betegségek kialakulását eredményezheti. A dendritikus sejtek vírussal történő fertőzése anti-virális állapotot hoz létre, ami sejt-autonóm módon gátló hatást fejt ki a fertőzött és szomszédos sejtekben. Az I. típusú interferonokat (IFN-α és β) krónikus vírus fertőzésekben (hepatitis C virus) és autoimmune betegségekben (sclerosis multiplex) terápiás célra is alkalmazzák. A monoterápiák hatékonysága azonban nem megfelelő, ezért az IFN alapú kombinált terápiák fejlesztése új kihívást jelent a vírus ellenes és autoimmune betegségek terápiájában.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.
A vírusok a környezetünkben nagy számban előforduló ártalmatlan, vagy fertőző képességgel rendelkező parányi élőlények. Egyedi sajátságaiktól függően az emberi szervezet eltérő sejt típusaiban telepednek le és fertőzést követően sokszor magas lázzal járó betegséget váltanak ki. A vírusok szervezetbe jutását az immunrendszer erre szakosodott receptoraik segítségével gyorsan érzékelik és válasz lépésként a vírus ellen védelmet biztosító oldott faktorokat, interferonokat termelnek, melyek képesek a vírusok semlegesítésére. Ennek a védelmi lépésnek fontos sajátossága, hogy visszahat a fertőzött és a szomszédos sejtekre is. Ennek eredményeként olyan gének aktiválódnak, melyek továbbra is rendelkeznek vírus ellenes hatással, ezzel biztosítva a vírus ellenes védelmet, de emellett visszahatnak az aktivált sejtekre is. Ez a működési elv azonban lehetőséget ad arra, hogy a vírus által kiváltott védelmi reakció olyan mértékben felerősödik, hogy gyulladásos betegségek kiváltását idézi elő. Az interferonok azonban gátló hatást is kifejthetnek, ezért bizonyos betegségek gyógyítására is alkalmazhatók. Ezek az eljárások azonban még további fejlesztést és új terápiás stratégiák kidolgozását igényli. A pályázat keretében végzett vizsgálatok tervezésekor az interferonok és az őket irányító szabályozó molekulák együttműködésének lehetőségeit tanulmányoztuk.

Summary of the research and its aims for experts

The major aim of the project is to identify signalling pathways, which upon collaborating with type I and III interferons and other soluble factors have the potential to modulate the outcome of anti-viral and anti-bacterial mmune responses. Considering the complexity of molecular events induced by viral recogition we aimed to analyse the signaling cascades induced by retinoic acid gene-1 (RIG-I) family receptors (RLR) playing a key role in viral recognition. We plan to identify the molecular background of collaborating and/inhibitory mechanisms with the potential to modulate the outcome of anti-viral responses. Based on our hypothesis the ligand-specific stimulation of RLRs, acting as pattern recognition receptors (PRR) of dendritic cells (DCs) induce mutual interactions at the level of cytokines and transcription factors. These interactions can induce both parallel and inhibitory effects with the potential to modulate the final outcome of the response. As the expression of PRRs is restricted to unique cell types, among them myeloid and plasmacytoid dendritic cells, while the production of interferons and cytokines are produced by many cell types we suggest that the regulation at the level of soluble factors acting mutually on activated cells have an impact on the outcome of the overall response of the cell. Thus the results of the project will uncover new aspects of immune regulation lead by interferons in concert with other cytokines present in the actual microenvironment of the affected cell.

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.
The basic question is how type I and III interferons modulate functional activities of various cell types, the expression levels of transcription factors upon triggering anti-viral responses. We analyse the effects of IFNβ on colon epithelial cells to identify enhancing and inhibitory effects. We also test how the stimulation of human DCs with polyI:C could exert different effects on MDA5 and RIG-I expression. The role of mTOR in the regulation of polyI:C-induced antiviral and inflammatory responses will also be tested by the stimulatory or inhibitory effects of mTOR on human DC subtypes. We also predict counter regulatory events during the collaboration of TLRs and RLRs monitored by the consequences of RIG-I up-regulation induced by simultaneous triggering of TLR3,7/8,9 with selected commensal bacteria to measure the expression levels of PRRs. Expression profiles of regulatory factors during microbial stimulation followed by a temporally designed treatment protocol, where moDCs are exposed to short term bacterial stimulation and are re-stimulated by the ligand 5’ppp-dsRNA to induce RIG-I expression and cytokines. Dissection of type I IFN and PRR signaling involved in bacterium-evoked moDC stimulation will be performed by the identification of the activated pathway components by detecting the phosphorylated transcription factors. A high density protein array will be used for monitoring changes in TLR/RLR receptor expression levels of interferon stimulated genes (ISGs). We expect the detailed mapping of signalling events mediating the IFNAR1 and PRR pathways that will allow us to design strategies for manipulating the immune response at the level of cytokines.

What is the significance of the research?

Toll-like receptors are key microbial sensors that stimulate innate immune defense programs. The ligation of TLR3 results in type I IFN production in many cell types, while TLR7, 8, 9 induce type I IFN secretion in cDCs and pDCs indicating the prominent role of these cytokines in regulating immune responses. The main question of the proposal is how type I and III IFNs are involved in the regulation of anti-viral responses. We focus to molecular consequences of virus recognition by cytosolic RLRs capable of inducing IFN secretion by both pDCs and cDCs with a potential to modulate the outcome of immune responses. We plan to study IFN-dependent an independent regulation of PRRs under inflammatory conditions induced by commensal bacteria known to elicit robust immune responses in normal tissues. We aim to identify the conditons of signaling pathways that result in synergistic effects or activate mechanisms with opposing regulatory effects. Recognition of viruses is mediated by specific virus receptors that are expressed in the vesicular membrane or in the cytosol of the cell. RIG-I receptors recognize single stranded or double stranded RNA and also DNA. The cytosolic RIG-I receptor detects 5’termini (5’ppp and 5’ppRNA). The unique feature of IFNS is that upon viral recognition high levels of type I and III IFNs are produced and multiple interferon stimulated genes (ISG) get activated, which also exhibit anti-viral activities and create an antiviral environment that affects neighbouring cells. Although the production of IFNs serves the elimination of viruses, the production of interferons acts back on the stimulated cells and may cause inflammatory reaction. Thus the proper regulation and counter regulation of IFN responses is of high importance. Infection of DCs by viruses is known to establish an anti-viral state that develops by a cell autonomous mechanism and confers inhibitory signals for infected and neighboring cells. Type I interferons (IFN-α, IFN-β) are used in therapies against chronic virus infections and also for treating autoimmune diseases however, the efficacy of these mono-therapies is far from optimal efficacy.

Summary and aims of the research for the public

Viruses are abundant innocent or dangerous small parasites. Depending on their unique properties they home to different cell types of the human body and are localized to intracellular compartments. Accidentally they provoke inflammation with usually high fever. Entry of viruses to the cells is rapidly recognized by specific virus receptors, which upon binding to the virus also alarm the immune system and induces the production of soluble mediators (interferons) that have the potential to neutralize and eliminate the invading viruses. The unique feature of this protection is that interferons induce the production of a wide array of interferon induced genes which still have anti-viral activity and can act back on the infected and neighbouring cells. This mode of action bears the risk that the interferon response is scaled up and results in the development of autoimmune diseases. Interestingly, interferons can also exert inhibitory functions, which can be utilized for therapeutical intervention such as the treatment of some viral infections. However, the efficacy of these therapies has to be improved and the development of novels strategies is required. In this project we designed experiments aimed to analyse the multiple cellular interactions of interferons.

Az immunrendszer aktivitása akkor megfelelő, ha működése finoman szabályozott. A túlzottan aktív immunválasz krónikus gyulladáshoz és autoimmun betegségekhez vezethet, míg a gyenge immunreakciók megkönnyíthetik a rák és a súlyos, sőt halálos fertőzések kialakulását. A pályázat megvalósítása során olyan új mechanizmusokat azonosítottunk, amelyek képesek modulálni az immunválaszok kimenetelét. Kimutattuk, hogy a muramil-dipeptid, egy bakteriális sejtfal komponens, és a vele együtt alkalmazott IFNγ erős immunstimuláló hatást gyakorol az emberi dendritikus sejtekre, ami lehetőséget teremthet patogének vagy rákos sejtek elleni hatékonyabb vakcinák kidolgozására. Azt is megfigyeltük, hogy a flagellin, egy baktériumból származó különleges fehérje, nemcsak aktiválja a dendritikus sejteket, hanem képes elősegíteni az immunogén sejthalált is. A flagellinnek ez az egyedülálló képessége lehetővé teszi, hogy adjuvánsként alkalmazzák különböző terápiás körülmények között, elsősorban a tumor immunterápiájában. Leírtuk továbbá, hogy a saphenous vénák falából izolált mezenchimális őssejtek hasonló immunmoduláló tulajdonságokkal rendelkeznek, mint a csontvelőből származó mezenchimális őssejtek.

The activity of the immune system needs to be finely balanced. Hyperactive immune responses can lead to chronic inflammation and autoimmune disorders, while weak immune reactions may facilitate the development of cancer and severe or even fatal infections. During this project we have identified novel mechanisms with the potential to modulate the outcome of immune responses. We have found that muramyl dipeptide, a bacterial cell wall component, together with IFNγ exerts strong immune stimulatory effects on human dendritic cells, and thus provide a rationale for designing more potent vaccines against pathogens and cancer cells. We have also observed that flagellin, a unique bacterial protein, not only activates dendritic cells, but also has an ability to promote immunogenic cell death. This unique capability of flagellin designates it as a potent adjuvant, which can be readily harnessed for different therapeutic settings, preferentially in tumor immunotherapies. Furthermore, we have described that mesenchymal stem cells isolated from saphenous vein vessel wall have similar immunomodulatory properties as their bone marrow-derived counterparts.
