A kutatás összefoglalója, célkitűzései szakemberek számára
Itt írja le a kutatás fő célkitűzéseit a témában jártas szakember számára.
A tervezett projekt célja a gyulladásos folyamatok alapjainak megismerése Drosophila modellszervezet alkalmazásával. A projekt a sokmagvú óriás vérsejt felfedezésén és az azt követő vizsgálatainkon alapul, melynek során felismertük, hogy ez, a Drosophila ananassae alcsoport fajaira jellemző sejttípus nagyfokú hasonlóságot mutat a gerincesek granulómáiban előforduló sokmagvú óriássejttel. Korábban, a Drosophila immunrendszerének a vizsgálatára alkalmas molekuláris és genetikai markereket hoztunk létre, jellemeztük a vérsejt alpopulációkat és vérsejtképző szöveteket. Az eddigi eredményeinkre építve a következő célokat tűztük ki magunk elé:
i) A D. ananassae csoportba tartozó fajok vérsejtjeinek vizsgálatára alkalmas markermolekulákat azonosítunk és jellemezzük a molekulák funkcióit.
ii) Transzgenikus törzseket és genetikai markereket hordozó törzseket hozunk létre D. ananassae-ban.
iii) Génexpressziómintázat analízist végzünk D. ananassaeból származó naiv vérsejteken, immunindukált egyedek plazmatocitáin és óriássejtjein. Jelölt géneket szelektálunk további vizsgálatokra.
iv) Az antiparazita választ és a parazita elpusztításában résztvevő faktorokat jellemezzük.
v) Keressük az azonosított gének/molekulák gerinces homológjait és szelektált targetmolekulák funkcióit vizsgáljuk gerinces modellszervezeten.
vi) Kutatási lehetőséget biztosítunk a molekuláris immunológia iránt érdeklődő fiatalok számára.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.
A Drosophila immunrendszerének vizsgálatára alkalmas molekuláris és genetikai eszköztárt hoztunk létre, melynek alkalmazásával egy eddig ismeretlen sejttípust, a sokmagvú óriás vérsejtet azonosítottuk a Drosophila ananassae alcsoport fajaiban. Ez a sejttípus nagyfokú hasonlóságot mutat a krónikus gyulladások során képződő egyes granulómákat jellemző sokmagvú óriássejttel. Mivel igen sok jelátviteli útvonal kulcseleme erősen konzervált, vizsgálatainkból az állatvilágra általánosan érvényes következtetéseket vonhatunk le. Kísérleteinkben azt vizsgáljuk, hogy a veleszületett immunitás elemei hogyan vesznek részt a krónikus gyulladások, a granulómaképződés és a szarkoidózis során. Drosophila modellszervezeten vizsgáljuk az óriássejtképződés során megvalósuló fúziós folyamatokban résztvevő felismerő struktúrákat továbbá a testidegen részecskék felismerésében és a patogének elpusztításában részvevő receptorokat. Továbbá vizsgálni szeretnénk az eddig ismeretlen, kizárólag az óriás vérsejtekben azonosított struktúráknak, a mikrovezikuláknak a tartalmát, mely feltehetően kapcsolatban áll az óriássejtek példátlan parazitaölő képességével. A Drosophilán és a laboratóriumi egéren nyert eredmények összehasonlítása a következtetések evolúciós vonatkozásainak vizsgálatára is lehetőséget ad és a krónikus gyulladásban résztvevő specifikus, konzervált faktorok azonosítását segítheti.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!
Korábban megmutattuk, hogy a Drosophila sejt közvetítette immunitása nagyfokú funkcionális hasonlóságot mutat a gerinces szervezetek sejt közvetítette immunfolyamataival, a bekebelezéssel és a granulómaképződéssel. Igazolást nyert az is, hogy ez a hasonlóság a molekuláris szinten zajló folyamatokra is kiterjed valamint, hogy a Drosophila és a gerinces szervezetek immunsejtjeinek fejlődésbiológiai és a morfológiai jellemzői is hasonlítanak egymáshoz. A sokmagvú óriás vérsejt azonosítása Drosophilában lehetővé teszi a krónikus gyulladás sejtes folyamatainak nagyfelbontású vizsgálatára alkalmas, genetikailag jól definiált és kontrollált rendszer létrehozását. A kísérletek várhatóan eddig ismeretlen jelátviteli hálózat(ok)at és effektor mechanizmusokat fednek fel, melyeknek jelentősége emlősökben vizsgálható. A fuzionált makrofágok gyakran előfordulnak granulómákban, ezért a sokmagvú óriássejtek fúziójában résztvevő faktorok azonosítása érdemben hozzájárulhat a granulómaképződéssel járó kórképek, mint például a tuberkulózis, vagy a szarkoidózis jobb megértéséhez és gyógyító kezeléséhez. Az óriás hemociták létrejöttében szerepet játszó faktorok azonosítása olyan evoluciósan konzervált faktorok azonosítását is lehetővé teheti, melyek a sokmagvú óriássejtek és az óriás hemociták képződésben egymáshoz hasonlóan vesznek részt.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.
Az immunrendszer a szervezetet érő külső és a belső kihívásoktól védi a szervezetet és kulcsszerepe van a kóros folyamatokkal - egyszerű megfázástól kezdve az anyagcsere betegségeken keresztül a daganatos betegségekig - szembeni védekezésben. Miután az immunrendszer működését szabályozó faktorok konzerváltak a törzsfejlődés során, sikeresen vizsgálhatók modellszervezetekben. A jelen projekt célja a gyulladásos folyamatok során zajló szabályozó folyamatok megismerése Drosophila modellszervezet alkalmazásával. A projekt a sokmagvú óriás vérsejt felismerésre és az azt követő irányra épül, melynek során kimutattuk, hogy hogy ezek a sejtek nagyfokú hasonlóságot mutatnak a gerincesek granulómákra jellezmő óriássejtekkel. A tokképzés és a granulóma képződés az immunrendszernek a krónikus ingerekre adott tipikus válaszreakciói. Valójában, a Drosophilában a parazitoid darazsakkal szemben zajló tokképző reakció vizsgálata a granulómaképződés folyamatának mélyebb megértését teszi lehetővé. A reakcióban résztvevő faktorok parallel vizsgálata rovar modellen és gerinces szervezetben az evolúciós vonatkozásokon túlmenően lehetővé teszi az eredményeknek az alkalmazott orvostudományba történő átvételét, mivel a veleszületett immunitás szabályozó elemeinek léteznek gátló faktorai. Újonnan azonosított molekulák targetek lehetnek a granulómaképződéssel járó kóros folyamatok (tuberkulózis, szarkoidózis) gyógykezelésében, de a kártevőkkel szembeni védekezésben is hasznosak lehetnek. Summary of the research and its aims for experts
Describe the major aims of the research for experts.
The specific aim of the present project is to receive insight in the regulatory and developmental processes of the inflammatory reactions using Drosophila as a model organism. The project is built on our original observations, the discovery of the multinucleated giant hemocyte (MGH) and a track we followed when showed that these cells, discovered in the ananassae subgroup of Drosophilidae, bear significant similarities with the characteristic cell type in different granulomas of vertebrates, the multinucleated giant cell (MGC). So far we generated molecular and genetic tools for the analysis of the Drosophila immune system, and characterized the hemocyte subsets and compartments with respect their origin and function. Based on these findings we propose a project with the following aims:
i) to identify novel molecular markers for further analysis of the cell mediated immunity of species belonging to the ananassae subgroup of Drosophilidae and to analyze the molecules with respect to their functions;
ii) to generate genetic markers and transgenic stocks in D. ananassae as tools for further analysis of MGH differentiation;
iii) to carry out a gene expression profile analysis of uninduced hemocytes, immune induced plasmatocytes and MGHs, and selection of candidates for further studies;
iv) to analyze the effective anti-parasite immune response and killing mechanism;
v) to find vertebrate homologs of the newly identified immune-genes/molecules via a computational analysis and to study their function in a vertebrate model;
vi) to provide a stimulating atmosphere for students who have an interest in molecular immunology;

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.
Our laboratory has generated molecular and genetic tools for the analysis of the Drosophila immune system and recently discovered a so far unidentified cell type, dubbed the multinucleated giant hemocyte, in the ananassae subgroup of Drosophilidae. This cell shows remarkable similarities to multinucleated giant cells, a characteristic feature of granulomas, a manifestation of chronic inflammation in vertebrates. As the core components of many genetic pathways - including those of blood cell development and innate immunity - are conserved from insects to humans our research will help to understand the regulation of blood cell development and innate immunity in general. The basic question of our studies is how the differentiation of the cellular elements of innate immune system is regulated in the chronic inflammation, more specifically in granuloma formation and in sarcoidosis. Using a Drosophila model we ask questions on the factors regulating the cell-cell recognition in the fusion process of activated macrophages as well as on the receptors involved in the recognition of the foreign particles and pathogens. Also, we wish to understand the possible link between the so far unrecognized ultrastructural elements, the micro vesicles, in the cytoplasm of the MGHs and their possible involvement in the unprecedented parasite killing capacity and related functions. A comparison of the results obtained from Drosophila ananassae and the subsequent experiments in laboratory mouse have the potential to provide an evolutionary context to our conclusions, and may highlight specific conserved events/factors involved in the chronic inflammation.

What is the significance of the research?

Previously we have shown that Drosophila cellular immune responses bear functional similarities with cellular immune reactions of vertebrates namely with phagocytosis, and granuloma formation. It was revealed too that the similarities extend into the molecular level and that the developmental and the morphological features of the immune cells in Drosophila and in vertebrates bear similarities. Describing the multinucleated giant hemocyte (MGH) in Drosophila, a multinuclear giant cell found for the first time in insects, allows the construction of a genetically well-defined and controlled system to study specific steps of chronic inflammation at a high resolution. Our experiments will reveal novel regulatory networks and effector mechanisms in Drosophila whose importance can subsequently be tested in mammals. The fused macrophages are common in granulomas therefore finding factors that initiate MGH formation can be important to find new therapeutic approaches in granuloma associated human diseases like tuberculosis, sarcoidosis. Identification of factors that initiate MGC formation will allow the extension of the study in the analysis of factors that are evolutionary preserved and play a similar role in initiation of MGH and MGC formation.

The immune system acts to internal and environmental challenges and has a key impact on pathological processes going from common cold to metabolic diseases, and cancer. As the regulatory factors of the immune system are evolutionarily conserved, they can be successfully studied in model organisms. The specific aim of the present project is to receive insight in the regulatory and developmental processes of the inflammatory reactions using Drosophila as a model organism. The project is built on the discovery of the multinucleated giant hemocyte and a track we followed when showed that these cells bear significant similarities with the characteristic cell type in granulomas of vertebrates, the multinucleated giant cell. The encapsulation reaction and granuloma formation in the animal kingdom are typical responses to chronic inflammatory stimuli of the innate module of the immune system. In fact, the encapsulation reaction and the capsule formation of the Drosophila larva in response to parasitoid infection serve as a viable model through which to study granulomas. The possibility of investigating candidates parallel in an invertebrate and a vertebrate system not only supports and extends the evolutionary aspect of the work, but may also allow the future translation of the results into applied medicine as the regulators and the mechanisms of innate immunity have chemical inhibitors making them attractive targets for therapeutic intervention in granuloma associated human diseases like tuberculosis, sarcoidosis, or, also in pest control. The project will also provide a comprehensive overview of the innate immune response against microbes and parasites in general.

Egy, a közelmúltban általunk felfedezett sejttípust, a sokmagvú óriás vérsejtet jellemeztünk egysejtes transzkriptom, bioinformatikai és ezeket kiegészítő funkcionális és transzmissziós elektronmikroszkópos vizsgálatokkal. Az állatvilágban a sokmagvú óriássejtek a granulóma képződéssel járó krónikus gyulladások során jelennek meg. A sokmagvú óriás vérsejtet azokban a Drosophila fajokban azonosítottuk, amelyek különleges hatékonysággal védekeznek a parazita darazsakkal szemben, és amelyekben az óriás vérsejtek granulómához hasonló tokot képeznek a darázs petéje és lárvája körül. A kísérleteink célja az volt, hogy információt gyűjtsünk a sejtek differenciálódását és funkcióit szabályozó folyamatokról. A kísérletek eredményeként az óriássejtekben több, ezekre a sejtekre jellemző gén- és fehérjehálózatot azonosítottunk, melyeknek jelentőségét a morfológiai és a funkcionális tesztek is megerősítették. A pályázat segített a korábban kialakult szakmai együttműködések folytatásában, továbbá egy új együttműködés kialakításában, mely egy a Drosophila genomba horizontális géntranszferrel integrálódott biotoxin géncsalád vizsgálatára irányul.

We characterized a novel cell type of innate immunity, the multinucleated giant hemocyte, by single cell transcriptome, bioinformatics and complementary functional and transmission electron microscopic analysis. Multinucleated giant cells are associated with chronic inflammation in the animal kingdom. In Drosophilidae the multinucleated giant hemocyte (MGH) was discovered in our laboratory and found to be involved in a highly effective anti-parasite cell mediated immune response, bearing similarities to granuloma formation in vertebrates. The aim of our studies was to find clues to the differentiation and function of these cells. We have identified genetic and protein networks as well as supporting morphological queues which may lead to the understanding of the unique features of these cells and their effector functions. The grant helped to continue the already existing collaborations and led to the development of a new, fruitful collaborative project on a horizontally transmitted biotoxin-gene family in the Drosophila genome, supporting the evolution and integration of diverse phenotypic modules in the animal kingdom.
