Tervezzük gyógyhatású szerves vegyületek esetében a vér-agy gát (BBB) átjárhatósága optimalizálásának vizsgálatát, in vivo (patkányon), és javaslatot teszünk a BBB átjárhatóság szempontjából optimális szerkezetre.
Számos nem-lipofil vegyület is átjut a BBB-en transzport mechanizmusok révén. A koncentráció növelése során a telíthetőség szinte valamennyi transzport folyamat jellemzője. A CNS-be való "intenzív/optimális bejuttatás" egyik módja a transzporter rendszer túlterhelésének megakadályozása. A vegyületek szöveti szintjeinek meghatározására nagyhatékonyságú folyadékkromatográfiás (HPLC) és kapilláris zóna elektroforézises módszereket (CZE) kívánunk alkalmazni, főleg az ultraibolya detektálásokra törekszünk (UV), alacsony koncentrációk esetén elektrokémiai, ill. fluorimetriás detektálást végzünk. Az eredményeink ellenőrzésére tömegspektrometriás (MS) méréseket is tervezünk. Különböző dózisok és adagolási módok alkalmazásával vizsgálnánk, hogy a GYO koncentráció a vér/agy, a vér/CSF, ill. az agyszövet/CSF arányok tekintetében független-e a beadott dózistól, vagy a dózis versus szöveti szint arány mutat-e telítési jelleget.
Következtetéseket tudunk majd levonni az egyes vegyületek in vivo vér-agy gát transzportjának mechanizmusára vonatkozóan, a vegyületek potenciális részvételére a BBB transzport folyamataiban. Kísérletes munkánk eredményei segítenek majd a hatásos és mellékhatásokkal kevésbé rendelkező szerves gyógyszer-vegyületek tervezésében, azaz a gyógyszerek olyan fejlesztésében, melyek a kívánt módon jutnak a támadáspontjukra. Adataink alapján a vegyületek megfelelő szerkezeti módosítását is elvégezzük, vagy erre javaslatot fogunk adni.
Summary
We aim to monitor penetration of drugs (and drug candidates) across blood-brain-barrier (BBB) and blood-cerebrospinal fluid-barrier (BCsfB). Even certain non-lipophilic compounds are able to have access to the brain utilizing one of the transport systems devoted for endogenous compounds. A saturation-type of dose versus degree of penetration is the common characteristic of transporter-facilitated movement across the BBB.
The presence or absence of transport process (active transport or transporter) will be studied including the saturation of these tentative transport processes. Conclusions will be drawn on the circumstances and conditions that regulate blood-brain and blood-CSF penetrations, including the determinant role of the chemical structures.
The experiments will be done using Wistar rats. The blood, cerebrospinal fluid and dissected brain tissues will be collected following drug administration at different doses and time points. Analyses will be carried out using HPLC, capillary zone electrophoresis (CZE) and HPLC-MS.
BBB penetration of K-27 and K-48 (very hydrophilic compounds); L-deprenyl, nordeprenil, L-DOPA and derivatives of L-DOPA; ecdysone and 20-hydroxyecdysone will be studied. Suggestions will be done how to alter the structure of an organic chemical compound for the sake of optimization the BBB penetration.

Három szerkezeti- illetve hatástani-csoportba tartozó gyógyszerek és vegyületek vér-agy gát átjárhatóságát vizsgáltuk (1) A limitált penetrálhatóságú pralidoximmal (antidotum) szemben a bisz-piridinum aldoximok (K027, K048, K203) agyi penetrációja fokozottabb. (2) A Parkinzon-kór kezelésében használt Jumex agyi penetrációja tökéletes, az agyban kimutatható szint nagyobb a vérszintnél, az agyi monoamin-oxidázhoz (MAO) való kötődés következtében. A kötődés azonban a Jumex p-fluor-származékánál jelentősebb. A Jumex és a p-fluordeprenil szervezeten belüli eloszlását nem befolyásolja szignifikánsan sem az adagolás módja (i.v., i.p., p.o.), sem az alkalmazott kísérleti állat (patkány, nyúl vagy Beagle kutya). A Jumex áthalad a vér-here gáton, kötődik a herében a MAO enzimhez. Független szerzők közölték, hogy patkányok kezelését követően a Jumex fokozza a spermiumok számát, mozgékonyságát, és növeli az utódok számát is. A Jumexhez hasonlóan a p-fluordeprenil is jelentősen feldúsul a herében. (3) Ekdiszteroidok esetében a 20-hidroxiekdizon nem hatol át a vér-agy gáton, de kevésbé hidrofil származékai jelentős agyi koncentrációt adtak. Központi idegrendszeri hatás elérésére a K203 antidotumként, p-fluordeprenil, mint Jumex helyettesitő javasolható bizonyos hatások elérésére. Kísérleteink eredményei alapján rendszeres vizsgálatok elvégzése ajánlott további Jumex ((-)-deprenil) analógok humán termékenység sikerességét növelő hatásának vizsgálatára!
Results in English
Blood-Brain Barrier (BBB) penetration was experimentally scouted. (1) Pralidoxim (mono-pyridinium aldoximes, antidotes of organophosphate intoxication) has limited BBB penetration. BBB penetration was essentially improved when using its bis-pyridinium analogues (K027, K048, K203). (2) BBB penetration of Jumex ((-)-deprenyl), used in the treatment of Parkinson’s disease is excellent. Its level in the rat brain is higher than that in the serum as Jumex is bound to the monoamine oxidase of brain. BBB penetration is even more expressed in (-)-p-fluorodeprenyl. Distribution of neither Jumex nor p-fluorodeprenyl is influenced by the mode of administration (i.v., i.p., p.o.). Moreover similar distribution patterns were found using rats, rabbits and Beagle dogs. Both Jumex and p-fluorodeprenyl can penetrate through the blood-testis barrier. Administration of Jumex facilitates reproduction of rats, increases the number and motility of sperms and also increased the number of offspring, as independent colleagues published. (3) 20-hydroxyecdysone does not penetrate BBB, but its less hydrophilic analogues do. K-203 is suggested as antidote in the case of organophosphate intoxication. p-Flourodeprenyl can substitute Jumex in certain special cases. Positive effects of Jumex in reproduction parameters (of rats) suggest further investigation of Jumex and p-fluorodeprenyl to elucidate how they can positively influence the number and quality of sperms both in animals and human beings.
