A búza nemesítési programok egyik fontos célja a tartalékfehérje frakciók mennyiségének és minőségének módosítása, hogy a lisztből készült termékek minősége minél jobban megfeleljen a fogyasztó igényeinek. A tartalékfehérjék expressziója nem csak a genetikai háttértől, de a környezeti hatásoktól is függ, melyek molekuláris hátteréről gabonafélék esetében nem sokat tudunk. Az elmúlt években csoportunkban elkezdett in silico vizsgálatok bizonyították, hogy egyes fehérje csoportok eltérő expressziós profilja egyértelmű kapcsolatba hozható a promóter régió szekvenciájában mutatkozó kis eltérésekkel. Ezek az összefüggések nehezen tanulmányozhatók a hexaploid búzában, de a modell gabonaként egyre szélesebb körben vizsgált diploid Brachypodium distachyon új lehetőségeket nyitott ebben a munkában is. A pályázatban megfogalmazott célunk, hogy az elmúlt 15 évben szerzett tapasztalatunk alapján vizsgáljuk a modell növény tartalékfehérjéinek expressziós profilját a szem fejlődés időszakában különböző módszertani megközelítések segítségével. Ezzel párhuzamosan vizsgáljuk a transzkripciót befolyásoló transzkripciós faktorokat is. A genomikai, transzkriptomika és proteomikai módszerek alkalmazásával kapott eredmények közelebb vihetnek a búza tartalékfehérje expresszió szabályozásának megértéséhez is, s hosszú távon hasznos információkat kaphatunk a minőségre történő nemesítéshez is.
Summary
Breeding programs are focused at the composition of the storage protein fractions related to end-use quality. The storage protein expression is tissue specific and expression levels of the fractions highly depends on stress and soil conditions. Our previous in silico studies on the promoter sequences of different prolamin groups revealed special relationships between regulatory elements and mRNA concentration in the endosperm. However the large genome size and the three genomes of bread wheat raise difficulties in the complex analysis of transcriptional regulation. To reduce these problem, a new model species Brachypodium distachyon has been suggested to use studying aspects of cereal development and gene functioning. Our aim is to take advantages provided by the smaller and known genome sequence, using all of our previous experience and knowledge we gained in the last 15 years involved in storage protein science. Our research is going to focus on the transcriptional regulation of seed storage proteins. Expression will be studied on various levels by different approaches like genomics, transcriptomics and proteomics. Transcription factors will also be investigated in this proposed project, to get deeper understanding of their regulatory mechanisms involved in grain filling. The result of the work carried out in our laboratories will broaden our knowledge and identify several factors playing relevant role in end-use quality determination in different environmental conditions.

A nemesítési programok egyik fontos célja a környezeti változásoknak ellenálló, stabil fehérje hozamot és minőséget produkáló búzafajták előállítása. Igazoltuk, hogy az egyes tartalékfehérje családok expressziója elsődlegesen a promoter régiók milyenségétől és a hozzájuk kapcsolódó transzkripciós faktorok időbeni és térbeni interakciójától függ. Ezek az összefüggések azonban a búzában elég nehezen tanulmányozhatóak. A Brachypodium distachyon, egyszerűbb genomjának továbbá az alacsonyabb kópiaszámban előforduló prolamin géneknek köszönhetően új lehetőséget nyitott ennek tanulmányozására. ‘Omikai’ vizsgálatokkal igazoltuk, hogy hő és szárazság stressz hatások a tartalékfehérje képzést felgyorsítják, ugyanakkor az egyes gének eltérő módon reagálnak a stressz hatásokra, ami az egyes fehérjék elérhető N és S raktározási képességével állhat összefüggésben. A fókuszált ko-expresszáló hálózatelemzés során igazoltuk, hogy a kódoló régiók hasonlósága ellenére a Brachypodium szemfejlődésének transzkripciós szabályozása számos egyedi sajátossággal bír, így a búza szemfejlődés modellezésére kevésbé alkalmas, azonban rendelkezik jó néhány olyan genetikai faktorral, mely szélsőséges környezeti viszonyokhoz való alkalmazkodási képességének megfelelően nagyobb abiotikus stressz adaptációs képességet feltételez. Ezeknek a stressz körülmények között is stabilan expresszáló genetikai faktoroknak a megismerése és a búza nemesítésben történő kiaknázása a projekt egyik legjelentősebb eredménye.
Results in English
One of the focus of wheat breeding is produce of sustainable wheat cultivars with high protein yield and quality. We have proved that the expression of prolamin storage protein genes is highly influenced by the composition of the promoter regions and the spatial and temporal interactions of trans-acting factors bound to these promoter motifs. However, these interactions are hard to study in the hexaploid wheat. Therefore Brachypodium distachyon, a diploid wild grass with a simple genome can serve as a good alternative in these investigations. ‘Omics’ approaches were used to prove that heat and drought stresses speed up the accumulation of storage proteins in the seed. However, the different gene types follow different expression patterns that can be related to the N and S reserving capabilities of the individual genes. Our focused co-expression network analysis have highlighted some differences in the transcriptional regulation of brachypodium and wheat prolamins. This phenomenon has also lead to the conclusion that Brachypodium is less suitable to model the mechanisms of wheat storage protein expression. However, due to its adaptability to wide range of environmental conditions it can posses some specific genetic and regulatory factors, less dependent on environment, leading to a better adaptation to abiotic stresses. The understanding of these genetic factors and possible utilization in the wheat breeding programs is one of the most important outcomes of the project.
