A serdülőkor az emberi agy fejlődésének kiemelt kritikus periódusa. Hipotézisünk szerint a felnőttre jellemző megbízható agykérgi hálózatok kialakulásának közvetlen előzményeként még a primer látó- és mozgatókérgi hálózatok is átesnek egy átmeneti destabilizációs és újraszerveződési fázison. Ez a fokozottan alakítható periódus segít megbírkózni a fejlődés és tanulás kettős követelményével: lehetővé teszi, hogy a hálózat hasznos kapcsolatai megerősödjenek, miközben a hibás, szükségtelen kapcsolatok felszámolódnak. Hasonló elképzelések korábban is megfogalmazódtak, például a szürkeállomány serdülőkorban bekövetkező térfogati csökkenésének, és a neurális szinkronizáció párhuzamos hanyatlásának magyarázatára. A viselkedéses teljesítmény és annak neurofiziológiai (EEG) korrelátumai azonban még nem kerültek szisztematikus vizsgálat alá ebben az értelemben. Munkánkban ezt a hiányt szeretnénk pótolni a perceptuális és motoros készségtanulás feltehetőleg nem-monoton fejlődési görbéi és az ezekhez kapcsolódó EEG korrelátumok feltárásával. A különböző agykérgi területek eltérő érési sebességének megfelelően a kamaszkor korai és késői stádiumában tervezünk méréseket. A tervezett kísérletsor a humán procedurális tanulás fejlődésével kapcsolatban végzett korábbi munkánkra alapoz, melyben primer vizuális és motoros kérgi területek kanonikus hálózatainak humán-specifikus fejlődési mintáit a késői serdülőkor kezdetéig jellemeztük, és a tanulást és az agyi érést meghatározó alvási paramétereket azonosítottunk. Jelen kutatásban szeretnénk kiterjeszteni vizsgálatainkat az emberi fejlődés talán legérdekesebb szakaszának, a kamaszkor egészének részletes vizsgálatára.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

1. Kutatási cél. A perceptuális integráció fejlődési-tanulási görbéjét fogjuk meghatározni késői serdülőkorban, az agykérgi hosszú távú kapcsolatok fejlődési mintázatának feltérképezésére kifejlesztett kontúrintegrációs feladatot alkalmazva. Nemlineáris változásokat várunk a késői kamaszkor mintázatában. Vizsgáljuk azt a kérdést, hogy vajon a kamaszkori fokozott agykérgi plaszticitás megnövekedett tanulási kapacitáshoz is vezet-e, vagy ezzel ellentétben, a kérgi hálózat éppen zajló strukturális átalakítása miatt kevésbé lesz képes új összeköttetések kiépítésére.
2. Kutatási cél. Teszteljük azt a hipotézist, mely szerint a motoros teljesítmény a kamaszkor alatt lejátszódó ellentétes irányú szürke és fehér állomány változások szelektív indikátora. A nem-szekvenciális ’finger-tapping’ feladat alkalmazásával maximális motoros sebességet mérünk. Az itt megjelelnő monoton fejlődési mintázat a kortikospinális összeköttetések vezetési sebességének, a mieliniziáció előrehaladásának korrelátuma. Ezzel szemben a szekvenciális ’finger-tapping’ feladatatban nyújtott teljesítmény, amely a motoros kéreg kiterjedt horizontális hálózatán alapul, feltehetően nem-monoton fejlődési görbét mutat, a hálózat átmeneti destabilizációját tükrözve.
3. Kutatási cél. Kapcsolatot szeretnénk találni a kamaszkori alvás neurofiziológiai mutatói valamint a perceptuális és motoros teljesítmény között, ambuláns poliszomnográfia és kvantitatív EEG analízis alkalmazásával. Életkorfüggő változásokat keresünk az alvási EEG-ben a lassú hullámú alvás (NREM) spektrális sűrűségében, a lassú hullámok amplitúdójában, meredekségében és a lassú hullámú tevékenység topografikus eloszlásában.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

Az emberi agy hosszúra nyúló szerveződési fázisának eredményeként igen sokáig befogadó a környezeti ingerekre, ami egyaránt magában hordozza az alkalmazkodás és a sérülékenység aspektusait. Míg a sokáig tartó plaszticitásának nyilvánvaló evolúciós előnyei vannak, egyelőre nem tudjuk, hogy milyen mechanizmusok teremtik meg a kifejlett agy stabilitását, a gyermekiből a felnőttszerű állapotba való lépést. Kutatásunk ennek a kérdésnek a tisztázásához járul hozzá.
Ugyan régóta ismert, hogy fontos agyműködési mutatókban meredek visszaesés tapasztalható kamaszkorban (pl. szinaptikus sűrűség, agyi metabolizmus, lassú hullámú alvás amplitúdója), a vonatkozó viselkedéses mérések egyelőre hiányoznak. Ezeket a méréseket szeretnénk pótolni a perceptuális és motoros működés tekintetében, és megvizsgálni az elektorfiziológiai és viselkedéses paraméterek korrelációit. Kutatásunk a következő jellemzők kombinációja miatt egyedülálló: 1. Longitudinális vizsgálatot tervezünk, mely eddig fel nem tárt összefüggéseket mutathat meg. 2. Az egyszerű életkori skála használata helyett (ami pontatlan adatokhoz vezet, hiszen a serdülőkor kezdetét tekintve akár hat év egyéni eltérés is előfordulhat), hormonális teszteket használunk a kamaszkor kezdetének megállapítására, és az adatokat egy jóval pontosabb, korrigált életkori skálán mutatjuk be. 3. Terveinkben szerepel egy nagy felbontású EEG készülék használata, amely pontos forrás-lokalizációt, és nagyon megbízható kérgi funkcionális méréseket is lehetővé tesz.
Elsősorban tipikusan fejlődő gyermekekkel és serdülőkkel fogunk dolgozni, adataink azonban segíthetnek a rendellenességek megértésben és a szükséges intervenciók megtervezésében. Oktatási és terápiás stratégiák tervezésében kiemelten fontos lehet az agyi struktúrák genetikus és környezeti hatásokra való érzékenységének időbeli meghatározása. Bízunk abban, hogy alvás EEG adataink hasznosak lesznek klinikai endofenotípusok és biológiai markerek meghatározásában, melyek segítségével a különböző jellegű terápiás módszerek között választani lehet.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

A felnőttkor határán rendkívüli átalakulás következik be a kognitív és érzelmi területeken. Ebben a korban zajlik egy sor identitást alakító fontos esemény, míg ezzel párhuzamosan emelkedik a droghasználók száma, a korai terhességek, a szexuális úton közvetített betegségek és az autóbalesetek száma. A fokozott kockázatvállaláson túl érdemes hangsúlyozni azonban ennek a periódusnak a fokozott rugalmasságát is. A jelentős mértékű plaszticitás a nagy lehetőségek idejévé teszi a serdülő éveket: bizonyos értelemben az agy érésének kritikus periódusává.
Ez alatt a kritikus időszak alatt az agykérgi hálózatok átmeneti destabilizáción és újraszerveződésen mennek keresztül mielőtt a stabil, felnőttre jellemző hálózatok kialakulnának. Feltételezzük, hogy a destabilizáció elengedhetetlen feltétele annak, hogy a hasznos kapcsolatok megszilárdulhassanak, ugyanakkor a nem adaptív agykérgi kapcsolatok megszűnjenek. Azt gondoljuk, hogy ez érinti az érzékléssel és mozgással kapcsolatos agyterületeket is. Célunk, hogy megragadjuk ezt az átmeneti szakaszt az érzékelési és mozgásos készségtanulás fejlődési görbéiben, és az ehhez tartozó alvás EEG korrelátumokban.
Az emberi fejlődés kiemelkedően fontos szakaszának alaposabb megértésén, valamint a kamaszok oktatóinak és szüleinek nyújtott információn túl, munkánk klinikai és terápiás jelentőséggel is bírhat. Számos neuropszichiátriai zavar, mint például a skizofrénia, a fejlődésnek ebben a szakaszában jelenik meg, ami arra enged következtetni, hogy a serdülőkorban lejátszódó agyi újraszerveződésben bekövetkező hibáknak életre szóló hatásai vannak. Kutatásunk segíthet a neuropszichiátriai zavarok korai és specifikus jeleinek feltárásában.

Adolescence is a critical period in the protracted course of human brain development, when the cortical networks are going through a transitional destabilization and reorganization phase, just preceding the emergence of established adult cortical networks. We assume that the destabilization phase is necessary to cope with the double constraints of strengthening useful connections while eliminating flawed ones even within the primary sensory and motor cortices. While similar hypotheses have been formulated in order to explain the observed cortical grey matter reduction during puberty, or the decreased task-related neural synchrony, the relationship between EEG measures and behavioral performance in adolescents has never been studied before. We aim to capture this transitional phase in non-monotonic behavioral developmental curves of perceptual and motor skill learning, and associated correlates of sleep EEG in early and late adolescence, corresponding to the different maturational rates of different cortical areas. The proposed work is based on the research team’s earlier efforts with respect to the human development of procedural learning, where we have characterized the human-specific development of the “canonical circuits” of primary sensory and motor cortices, and defined relevant aspects of slow-wave sleep determining learning and brain maturation. Here we would like to expand these efforts into the most fascinating period of human brain development.

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.

Specific Aim (1) is to establish the developmental-learning curve of perceptual integration in a late adolescent (15 to 19 y) group. A contour integration task, developed by the research team to address the human development of long-range connectivity in the primary visual cortex, will be employed involving a perceptual learning procedure. We expect to observe nonlinear performance variations, and ask whether the enhanced plasticity of the adolescent brain leads to enhanced learning in a perceptual task, or, alternatively, the cortical network is less capable to build in new connections while it is under construction.
Specific Aim (2) is to test the hypothesis that motor performance is a selective indicator of changes in white- and grey matter volume during adolescence. Employing a non-sequential finger-tapping task we measure maximum motor speed, and ask whether we can find the predicted monotonous development and correlation with the conduction velocity of the corticospinal tract. In the sequential finger-tapping task, which relies on the extensive horizontal network of the primary motor cortex, we expect to see a non-monotonic developmental curve, reflecting the temporary destabilization of the network.
Specific Aim (3) is to bridge the relationship between the neurophysiology of sleep and perceptual and motor performance in adolescents by using ambulatory polysomnography and quantitative EEG analysis. We will look for age-dependence in the power spectral density of (NREM) sleep EEG (slow wave activity), the amplitude of sleep slow waves, the steepness of slopes of sleep slow waves, and in the topographical distribution of sleep slow wave activity.

What is the significance of the research?

The extremely long phase of cortical organization provides the human brain with an extended sensitivity for environmental impact, including aspects of both adaptability and vulnerability. While it is clear that the protracted plasticity of the human brain should have evolutionary advantages, it has not been clarified yet what neural mechanisms bring the cortical networks to a fully developed stability, and permit a child to enter adulthood. Our research will contribute to the clarification of this issue.
Although it has been demonstrated before that relevant measures of cortical function are steeply declining in adolescence (e.g., synaptic density, cerebral metabolism and sleep slow wave amplitude), behavioral measures correlating with these are still missing. We offer to provide these missing measures in the perceptual and motor domains, and to investigate correlations between electrophysiological and behavioral parameters. Our proposed research will be unique for three reasons: (1) We are proposing a longitudinal that will uncover relevant developmental trends; (2) Instead of using a simple chronological age-scale (leading to noisy data since the onset of puberty can vary by as much as 6 years), we offer to use hormonal tests to determine the onset, and present the data on a corrected age-scale with a much increased precision; (3) We propose to use a high density EEG equipment with high spatial resolution to allow precise source-localization, and more reliable cortical functional measurements.
Although we plan to include only typically developing children and adolescents, these data will provide a baseline to assess pathology and to guide interventions in the case of atypical development. Information about the exact time when certain brain structures are particularly sensitive to genetic or environmental influences during development might have imperative educational and therapeutic implications. We also hope that our sleep EEG data will prove to be useful in providing endophenotypes, biological markers that have the prospective to define subtypes of disorders responding to different treatments.

Fascinating “nonlinear” transformations in the cognitive and emotional domains take place at the brink of adulthood. This is the age when identity-forming life-events occur, and, at the same time, the prevalence rates of drug abuse, early pregnancies, sexually transmitted diseases, car accidents are on the rise. Beyond the predisposition towards risk-taking behavior, the adaptive nature of increased malleability in the adolescent brain has also been emphasized. This enhanced plasticity makes the teen years a time of great possibilities: in some sense, a “critical window” of brain maturation.
During this critical period, the cortical networks are going through a transitional destabilization and reorganization phase, just preceding the emergence of established adult cortical networks. We assume that the destabilization phase is necessary to cope with the double constraints of strengthening useful connections while eliminating flawed ones even within the primary sensory and motor cortices. We aim to capture this transitional phase in non-monotonic behavioral developmental curves of perceptual and motor skill learning, and associated correlates of sleep EEG.
In addition to understanding this important phase of human development better, and providing useful information for educators and parents of typically developing teenagers, we believe that our work will also have clinical and therapeutic relevance. A number of neuropsychiatric disorders, including schizophrenia, emerge during this period, indicating that errors in brain reorganization during puberty might have life-long effects. Our research may potentially reveal early and very specific signs of such disturbances.

A felnőttkor határán egy sor identitást alakító fontos esemény zajlik, és ezzel párhuzamosan kialakulnak a felnőttre jellemző agykérgi hálózatok. Az volt a célunk, hogy megragadjuk ezt az átmeneti szakaszt az érzékelési, mozgásos és magasabb szintű fejlődést közvetítő hálózatok tekintetében, valamint az ezekhez tartozó alvás EEG korrelátumokban. Poszterior-anterior agyi érésnek megfelelő fejlődési trajektóriákat kaptunk a viselkedésesen mért érzékelési, mozgásos és végrehajtó funkciók tekintetében, ami jól összeköti az idegtudományi és pszichológiai módszerekkel mért eredményeket. A több száz serdülő alany tanulmányozása közben egy olyan sikeres paradigmát fejlesztettünk ki, mellyel most először valóban figyelembe lehet venni az érettségből adódó eltéréseket. Az MTA és a PPKE támogatásával ezt a paradigmát már át is ültettük a gyakorlatba. Az alvásnak a fejlődésben betöltött szerepére vonatkozóan három jelentős eredményünk van. Egy új alvás-EEG szinkronizációt elemző módszer segítségével fontos fejlődési mintázatokat találtunk az agy nagyléptékű összeköttetéseiben. Az intellektuális fejlődés konkrét alvási korrelátumaira bukkantunk. Az alvási orsók és a motoros tanulás között kauzális összefüggést találtunk. Mindezek az eredmények megerősítik azt a hipotézisünket, miszerint az alvás a fejlődés „Achilles sarka” – amint az az NKFI honlapján, a „Meggyőző eredmények, példaértékű sikerek” rovatban is olvasható.
Results in English
Adolescence is a relevant period in terms of identity-forming life-events, and also in terms of the transition of cortical neural networks into an adult pattern of connectivity and function. Our purpose was to map puberty induced changes in the development of the “canonical circuits” of sensory, motor and frontal cortices, and to delve into the unresolved issue of sleep determined teenage development. We found that posterior-anterior anatomical brain maturation is reflected by the developmental trajectories of visual, motor and executive function, providing a link between neuroscientific and behavioural data. Studying hundreds of teenage subjects helped us to develop a novel paradigm that takes puberty onset times into real account for the first time. We have recently received HAS and PPCU support to apply this new paradigm. In terms of finding the exact developmental role of sleep in adolescence, there are three major results of the project. First, with our newly developed technique to analyze sleep-EEG synchronization, we found relevant typical and atypical developmental trends in large-scale cortical connectivity. Second, we found specific sleep correlates of intellectual development. And third, we found a causal relationship between sleep spindle activity and motor learning. Taken together, these findings point to the fact that sleep is the Achilles’ heel of brain development – detailed in the „Impressive results, exemplary achievements” section of the NRDI homepage.
