Az uborka mozaik vírus (Cucumber mosaic virus, CMV) egyike a legsúlyosabb károkat okozó vírusoknak, köszönhetően rendkívül széles gazdanövénykörének és földrajzi elterjedésének. A vírus felépítése igen egyszerű, egy köpenyfehérje burkon belül találjuk a vírus osztott genomú örökítőanyagát, mely összesen öt fehérjét határoz meg. A vírus teljes "életciklusát" ez az öt, multifunkcionális fehérje szabályozza, szoros együttműködésben a gazdanövény fehérjéivel. A vírusfehérjék különböző funkcióinak előtérbe illetve háttérbe kerülésének szabályozásáról jelenleg nagyon keveset tudunk.
Minden eukarióta sejtben a fehérjék transzláció utáni módosítása (például foszforilációja) igen fontos szabályozási funkciót lát el, mégis a vírusfehérjék módosításáról alig áll rendelkezésünkre adat. A CMV esetén eddig két esetben írtak le ilyen módosítást, egyrészt a 2a fehérje foszforilációját vírusfertőzött növényben, másrészt a 3a fehérje foszforilációját transzgénikus növényben, de ezekben az esetekben a foszforilációnak sem a pontos lokalizációját, sem a vírusfertőzés során betöltött funkcióját nem tisztázták. Jelen pályázat során szeretnénk a vírusfehérjék foszforilációjának hatását vizsgálni a CMV fertőzése során. Elsősorban a vírus mozgásában résztvevő fehérjék (3a fehérje, köpenyfehérje, 2b fehérje) esetén szeretnénk a foszforilációs helyeket azonosítani, majd mutáns vírusok készítése után vizsgáljuk ezeknek a fertőzés folyamatában, a tünetek kialakításában és a gazdanövénykör meghatározásában betöltött szerepét Nicotiana sp. tesztnövényeken és a CMV természetes gazdanövényein.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Korábbi munkánk során (Gellért és mtsai., 2006) homológia modellezésen alapuló funkcionális analízissel a CMV köpenyfehérjéjében in silico foszforilációs, amidációs és glikolizációs helyeket azonosítottunk. Ezek közül a lehetséges protein modifikációs helyek közül két foszforilációs hely nem csak a CMV izolátumokban, hanem a többi, a cucumovírus nemzetségbe tartozó vírus köpenyfehérjéjében is jelen van. Két másik foszforilációs hely és egy amidációs hely mindegyik CMV izolátumban megtalálható. Korábbi OTKA pályázatunk során a 2b fehérje “alanin-scanning” mutagenezise során azonosítottunk egy nem fertőző vírus variánst, ahol az aminosav változás (sps40-42aaa) egy jósolt foszforilációs helyen található. Ezen korábbi munkáink eredményeit felhasználva szeretnénk szisztematikusan vizsgálni a 2b, 3a és köpenyfehérjén található lehetséges foszforilációs helyek szerepét a vírusfertőzés során. Egyrészt a vírusfehérjék és mutáns változataik foszforilációját in vitro vizsgáljuk, majd a megváltoztatott (foszforilációra nem képes) vírusfehérjék biológiai hatását in vivo fertőzőképes klónok felhasználásával fogjuk tesztelni. A kísérleteket a CMV tesztnövényei mellett fontos gazdanövényein, paradicsomon, paprikán, uborkán fogjuk végezni.

Mi a kutatás jelentősége?

Számos növényi rezisztencia gén tartalmaz kináz domént, és kórokozók támadása során a növényekben számos MAP kináz aktiválódik. Így a vírusfehérjék foszforilációjának a kompatibilis illetve inkompatibilis vírus-gazda kölcsönhatásoknak kulcsszerepe lehet. A tervezett munka fontos adatokkal járulhat hozzá, hogy megismerjük a CMV fehérjék módosításainak jelentőségét, ezek biológiai, növénykórtani vonatkozásaival együtt. A foszforilációs események lehetséges terápiás célpontul is szolgálhatnak, ahogy ilyen kísérletek herpesz vírusok esetében már folyamatban vannak.
A hosszabb távú gyakorlati vonatkozások mellett természetesen munkánk új információkkal szolgál az egyik legfontosabb növényi vírus fertőzésének, az általa okozott tünetek kialakulásának folyamatáról, valamint a vírus viselkedéséről rezisztens és fogékony fajták esetén. Mindemellett betekintést nyerhetünk a multifunkcionális vírusfehérjék különböző funkcióinak összehangolásának módjába is.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

Az uborka mozaik vírus gazdaságilag az egyik legfontosabb vírus, több mint 1000 növényfajt fertőz, melyek között zöldségnövényektől a fás szárú növényekig sok faj megtalálható. Az egész világon elterjedt, a trópusi területektől egészen az északi régiókig. Az uborka mozaik vírus igen egyszerű felépítésű, egy fehérjeburokban található a vírus osztott genomú örökítő anyaga, mely összesen öt fehérjét kódol. Ez az öt fehérje felelős a vírus "szaporodásáért", terjedéséért a fertőzött növényen belül, illetve a növények között, valamint a betegségtünetek kialakításáért. Természetesen mindegyik fehérjének több feladata van a vírusfertőzés során, tehát multifunkcionálisak, illetve szorosan együttműködnek a gazdanövény fehérjéivel. Ezek a folyamatok megkívánják a vírusfehérjék működésének pontos szabályozását. Közismert hogy a növényi sejtekben a fehérjék több mint harminc százaléka foszforilálódhat, ami a biokémiai folyamatoknak egy alapvető szabályozó mechanizmusa. Növényi vírusfehérjék foszforilációjáról és ennek a szabályozó mechanizmusnak a szerepéről a vírusfertőzés folyamatában alig érhetők el adatok. Pályázatunk során az uborka mozaik vírus in silico meghatározott módosítási helyeinek vizsgálatát tűztük ki célul. Szeretnénk meghatározni, hogy a fehérje módosítás milyen mértékben érinti a vírus mozgásában és terjedésében szerepet játszó fehérjéket. Meg fogjuk vizsgálni, hogy ezeknek a módosításoknak milyen hatása van a vírus gazdanövénykörének, tüneteinek meghatározására, valamint hogy vírusfertőzésre fogékony és rezisztens fajták esetében különbözik-e a vírusfehérjék módosítása. Munkánk alapját képezheti új, vírusok elleni védekezési stratégia kialakításának.

Cucumber mosaic virus (CMV) is one of the economically most important viruses with extremely wide host range and economical importance. The devided genome of the CMV encodes only five proteins. The primary function of the 1a and the 2a protein is the participation in the virus replication, the 2b protein is the gene silencing suppressor of the virus, the 3a is the movement protein and the coat protein is encapsulate the viral RNA. Beside these primary functions, all the viral proteins multifunctional and participate in the formation of pathological characteristics (for example host range, symptom phenotype). The regulation of the different functions of the proteins is still unknown.
No much data is available in the respect of phosphorylation of plant viral proteins as it is the case also with the phosphorylation of CMV proteins. Only the phosphorylation of the 2a protein and the 3a protein in transgenic plants was reported, but the function and the strict localization of the phosphorylation sites is still in question. In this project we would like to extend our knowledge in the role of protein modification during the infection cycle of CMV. We would like to precisely identify as much protein modification sites (primarily phosphorylation sites) as possible on the movement related proteins of CMV (2b, 3a and the coat protein) and investigate the biological significance of these modifications. The role of the significant modification will analyse in different plant-pathogen systems including resistant and susceptible economically important hosts of CMV. Based on this information new defence strategies can be developed.

What is the major research question?

In our previous study (Gellért et al., 2006) we have predicted phosphorylation, amidation and glycolisation sites in the CP of cucumoviruses according to the homology modeling based functional analysis. There are at least two predicted phosphorylation sites conserved in the amino acid sequence of all cucumovirus CPs, and two more phosphorylation and one amidation recognition sequence conserved in the case of all CMV isolates. In our previous OTKA project we also identified an amino acid triad in the 2b protein essential for virus infection and located in a predicted phosphorylation site. In the present project we would like to extend the computer based protein modification site prediction for the 2b protein and MP. We will analyze the biological significance of the predicted phosphorylation sites during the virus life cycle using specific in vitro mutants of Rs-CMV. The original and the mutated proteins will be assayed for in vitro phosphorylation in cell free soluble fraction of N. benthamiana plant. We are going to analyze the effect of the phosphorylation on different economically important host plants of CMV (tomato, pepper, cucumber). Tolerant and susceptible cultivars will be included the experiments.
Generally this project would like to describe the role of protein modification in the infection cycle of one of the economically most important viruses namely cucumber mosaic virus.

What is the significance of the research?

A number of resistance genes in plants contain kinase domains and even specific MAPKs are activated by plant responses to pathogens. Thus phosphorylation can be a key element in compatible or incompatible virus-host interactions. Our work can significantly broaden our knowledge in respect of the opportunity and biological significance of phosphorylation of different cucumovirus proteins, and can display the differences between susceptible and resistant cultivars. Even the phosphorylation events during viral infections provide potential therapeutic targets for virus infection.
Beside this practical aspect, this project can significantly increase our knowledge on the role of protein modification in a plant virus infection cycle. Even if the protein phosphorylation is one of the best studied biochemical processes, in the field of plant virology limited data are available. Since 30% of the proteins of the eukaryotic cells are phosphorylated, there is negligible choice of the viral proteins to escape modification. Since protein phosphorylation occurs mostly in a sequence-specific and reversible manner it allows a complex regulation of biological events and it could become a new and valuable field in the research of cucumber mosaic virus.

Cucumber mosaic virus (CMV) is one of the economically most important viruses infecting over 1000 plant species in more than 85 plant families. It is prevalent all over the world from the tropical territory to the north regions. The CMV has a simple organization, its genome is enclosed in an isometric shell made up of many small protein molecules. The CMV codes only five proteins responsible for virus multiplication, induction of disease symptoms, spreading the virus inside and between plants. The viral proteins are multifunctional, and tightly co-operate with the proteins of the host plant. This cooperation requires the precise phasing and regulation of the different functions of the viral proteins. A potential possibility for the regulation is the reversible modification of the viral proteins. Even if about 30% of the proteins in a plant cell are modified, hardly any information is available regarding to the modification of plant viral proteins. Our research scope is the analysis three CMV proteins in this respect, recognizing the significance of these modifications in the viral life cycle, in host range and symptom formation. The widening our knowledge in this respect could provide potential therapeutic targets.

Results in Hungarian
Generally the viruses has limited number of proteins, in the case of cucumber mosaic virus (CMV) that means five proteins altogether. The viral proteins has several function during the infection cycle, and the post translational modification offer an excellent opportunity to switch between the different functions. We have analyzed the phosphorylation of the 2b protein, the coat protein (cp) and movement protein (mp). We have demonstrated the phosphorylation of 2b protein in vivo, both in infected N. benthamiana and in infiltrated patches. Mutants containing aspartic acid in the phosphorylation site accumulated only in the cytoplasm indicating that phosphorylated 2b protein could not enter the nucleus. We identified a conserved dual phosphorylation switch in CMV 2b protein, which equilibrates the shuttling of the 2b protein between the nucleus and the cytoplasm, and regulates the suppressor activity of the 2b protein We have also verified the cp phosphorylation and proved that it has a central role in the cycling symptom phenotype of CMV. The phosphorylation of the CMV mp was also analyzed, and we have determined that the mp phosphorylation could have an effect on symptom formation in connection with plasmodesmata localization.

A vírusok genomja csupán néhány fehérjét kódol, például az uborka mozaik vírus (cucumber mosaic virus, CMV) összesen öt fehérjével rendelkezik. Mindegyik fehérje multifunkcionális, számos különböző funkciót tölt be a vírus életciklusa során. A fehérjék poszttranszlációs módosítása kiváló lehetőséget biztosít a különböző funkciók szabályozására. A pályázat során a 2b fehérje, a köpenyfehérje (coat protein, cp) és a mozgási fehérje (movement protein, mp) foszforilációját és ennek a módosításnak a szerepét vizsgáltuk. Megállapítottuk, hogy a 2b fehérje foszforilálódik infiltrált levelekben és fertőzött N. benthamiana növényekben is. A foszforilációt modellező mutánsok csak a citoplazmában halmozódtak fel, nem jutottak be a sejtmagba. Bizonyítottuk, hogy egy dupla foszforilációs hely határozza meg a 2b fehérje sejten belüli lokalizációját valamint a foszforiláció a 2b fehérje szuppresszor aktivitásának szabályozásában is részt vesz A CMV cp foszforilációját is bizonyítottuk, és megállapítottuk, hogy központi szerepe van a CMV-re jellemző ciklikus tünetek kialakításában. A CMV mp legalább három helyen foszforilálódhat, és szerepe van a vírustünetek kialakításában, illetve a mp plazmodezmata lokalizációját szabályozza.
