A pályázó és együttműködő partnerei, Prof. Bateman (Kew, UK) és Dr. Ovidiu Paun (University of Vienna, Austria) elsődleges célja, hogy új-generációs szekvenálási módszerrel (RAD-seq) kísérletes módszerrel teszteljék az Epipactis nemzetségben (Orchidaceae) megfigyelt fajképződési módot, amely „evolúciós zsákutcát” jelent a – feltehetőleg egymástól függetlenül képződött – autogám fajoknak. Célunk egy kétlépcsős kutatásban 1) egy európai léptékű molekuláris filogenetika felállítása, amely keretet ad további munkánknak, de önmagában is jelentős eredmény; 2) egy európai léptékű földrajzi almintán (Észak-Görögországtól Angliáig) RAD-seq alkalmazásával feltárni az Epipactis helleborine feltehető szülőfaj és belőle származott fajainak filogeográfiai szerkezetét. A javasolt kutatás konkrét célja mellett a pályázó fontosnak tartja, hogy folytassa egy kisebb pályázat segítségével megkezdett munkáját, amely a RAD-seq módszert hazai meghonosítását célozza. Ezzel a módszerrel messze mélyebb betekintést nyerhetünk az élőlények közötti evolúciós viszony feltárásába, így sikerrel alkalmazható közelmúltban elvált csoportok filogenetikai szerkezetének megismeréséhez.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Az Epipactis nemzetségben (Orchidaceae) megfigyelt fajképződési mód, amely széles elterjedésű allogám fajból (E. helleborine) kiinduló, többszöri alkalommal egymástól függetlenül kialakult autogám fajok keletkezését valószínűsíti, a feltételezések szerint „evolúciós zsákutcát” jelent a kialakult fajoknak. Sajnos ezt a feltételezést, a konvencionális DNS-technikák alkalmazásával (elsősorban kloroplaszt DNS-en alapuló PCR-RFLP és kiválasztott régiók szekvenálásának technikájával) nem sikerült regionális szinten igazolni, elsősorban a rendelkezésre álló technikák kis felbontó képessége miatt. Kutatási tervünkben egy forradalmian új módszerrel, RAD-szekvenálással igyekszünk feltárni az Epipactis helleborine csoport európai léptékű filogeográfiáját, ezáltal empirikusan tesztelve az „evolúciós zsákutca” jelenséget. A kutatás alapkérdése, hogy az önmegporzó fajok valóban „evolúciós zsákutcát” jelentenek-e a nemzetség evolúciójában. Feltételezzük, hogyha igaz az evolúciós zsákutca hipotézis, úgy a viszonylag széles elterjedésű autogámok egymástól függetlenül, az allogám E. helleborine-ből leágazva jelentek meg, amit a kellő felbontást adó RAD-seq-kel nyert filogeográfiai kép tesztelhet; ha akár részben azt találjuk, hogy ezek egymásból alakultak ki, akkor az „evolúciós zsákutca” voltukat cáfolja.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

Az evolúciós zsákutca hipotézis számos esetben feltételezik a növényvilág evolúciójában, ahol gyakori az önmegporzó fajok képződése. Mindazonáltal ezt a jelenséget még nem vizsgálták meg az igazán nagy filogenetikai felbontó képességű új-generációs szekvenálási módszerekkel. Ezért a javasolt kutatás úttörő lehet ezen a téren, hiszen bizonyíthatja, hogy egy egyébként kevés filogenetikai információt hordozó csoportban is tesztelhető RAD-seq módszerrel finom léptékű mikro-evolúciós speciációs folyamatok. A nemzetség szintjén ez a vizsgálat elsőként adhat betekintést annak evolúciós viszonyaiba, megteremtve ezáltal azt a kiforrott taxonómiai alapot, mely egy jobban megalapozott természetvédelmi kezelésekhez segíthet hozzá. A RAD-seq módszer hazai megvalósítása pedig biztosítja, hogy ezt az egyértelműen a jövőbe mutató, nagy felbontású filogenomikai módszert Magyarországon is meghonosítsuk, melyet sikerrel alkalmazhatunk problematikus növény- és állatfajok, csoportok evolúciós viszonyainak feltárásában.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

A hazai orchideákat képviselő Epipactis nemzetséget magas fajképződési ráta jellemzi, melyet rovarmegporzású ősökből több alkalommal képződött önmegporzó fajok spontán keletkezése tart fenn, melyek nem járulnak hozzá a génusz további evolúciójához, ezért „evolúciós zsákutcának” tekinthetők. Sajnos eddig ezt nem tudtuk tesztelni, de most rendelkezésünkre állnak olyan, új-generációs szekvenálási módszerek, melyekkel a fenti elképzelés helyessége megvizsgálható. A kutatás ilyen, ún. molekuláris filogenomikai (a teljes DNS-állományt összehasonlító) módszert (RAD-seq) kíván használni. Egy európai léptékű földrajzi területen meg fogjuk vizsgálni a feltételezett szülőfaj genetikai szerkezetét, és összehasonlítjuk a feltételezett utódfajokéval, így tesztelve a fenti hipotézist. Az elméleti eredményeken túlmutatóan kutatásunk hozzájárul a nemzetség evolúciós viszonyainak, így taxonómiai helyzetének tisztázásához, ezen keresztül pedig megalapozott természetvédelméhez. A vizsgálati módszer hazai alkalmazásával pedig ennek a kétségkívül jövőbe mutató, nagy felbontású filogenomikai módszer magyarországi meghonosítását célozzuk meg, melyet sikerrel alkalmazhatunk problematikus növény- és állatfajok evolúciós viszonyainak feltárásában.

The primary goal of the joint effort of the applicant and his collaborators, Prof. Bateman (Kew, UK) and Dr. Ovidiu Paun (University of Vienna, Austria) is to experimentally test the “evolutionary dead-end” hypothesis proposed to describe speciation in the European genus Epipactis (Orchidaceae) by examining two levels of genetic variation by: i) building Europe-wide phylogeny to provide a robust phylogenetic framework for the work; ii) testing the evolutionary dead-end nature of the autogamous species by exhaustively examining a geographical subsample of European material with a phylogenomic approach utilising next-generation sequencing techniques (RAD-seq).This high-throughput technique assures resolution at micro-evolutionary scale by providing insight into the phylogeography of the allogamous E. helleborine as putative parent, and its presumed direct descendent autogamous daughter species. Besides the main goal of the current proposal, the applicant would like to continue his just started small-scale researches with RAD-seq for the ultimate goal of implementing this method into Hungarian phylogenetics. Since we can gain far more detailed insight into the (micro-)evolutionary relationships of organisms than ever before, RAD-seq can be applied to reconstruct phylogenetic relationships of recently radiated groups.

What is the major research question?

The “evolutionary dead-end” hypothesis proposed to describe speciation in the European genus Epipactis predicts multiple, independent segregation of autogamous daughter species from the widespread allogamous species E. helleborine without the possibility to give rise to new autogams. Mostly because of the limited resolution of chloroplast PCR-RFLPs and candidate gene sequencing available to researchers, it was unfortunately impossible to examine this prediction on a broad geographic scale. The present proposal benefits from the high resolution power of a revolutionary new approach, RAD-sequencing, which can help us to gain insight into the fine-detailed phylogeography of the Epipactis helleborine alliance thus testing empirically the "evolutionary dead-end" hypothesis. Therefore, the main research question is whether the autogamous species do really represent evolutionary dead-ends in the evolution of the genus? Our prediction is if this is really the case, than the autogamous species with considerably large distribution will arise on the RAD-seq phylogenetic tree from the allogamous E. helleborine independently from each other as short "sub-branches" of the same species. Otherwise, if the autogamous species give rise to some new autogams (i.e. sub-branches include more than one autogamous species), that will question the evolutionary dead-end nature of autogamous species in the genus Epipactis.

What is the significance of the research?

The evolutionary dead-end hypothesis is usually invoked when describing evolution of plant groups particularly rich in autogams. Nonetheless, this hypothesis has not been revisited by high-resolution power next-generation sequencing techniques. Therefore, the proposed research can be pioneering in this field of research by testing micro-evolutionary speciation processes with RAD-seq techniques in groups with otherwise low phylogenetic signal. At the level of this genus, this can be the first insight into the micro-evolutionary relationships thus providing taxonimic basis to a much stronger basic for conservational efforts. Finally, the Hungarian implementation of RAD-seq, this futuristic method of high phylogenetic resolution power, can provide important yardstick to Hungarian biology to understand evolutionary relationships of problematic plant and animal species and groups.

Genus Epipactis, representing European terrestrial orchids, is characterised by high speciation rate, which is supposedly fuelled by recurrent independent splitting off of autogamous (self-pollinating) species from more widespread allogamous (insect-pollinated) species. These new species will supposedly not contribute further to the evolution of the genus, making them to be 'evolutionary dead-ends'. Unfortunately, we had no technical possibility yet to test this hypothesis, but recent development in sequencing techniques, namely next-generation sequencing, opened a new avenue of research. The proposed research utilises such technique, the so-called molecular phylogenomics (genome-wide comparison of DNA) method of RAD-sequencing. We will examine the geographical structure of the putative parental species on a European scale, and compare it to the structure unravelled for the putative daughter (descendant) species, thus testing the above hypothesis. Beyond these theoretical goals, our planned research will contribute to the understanding of the evolutionary processes, thus providing basis for a well-established taxonomy, on which in turn we can base our conservation of this charismatic orchid genus. Finally, the Hungarian implementation of RAD-seq, this futuristic method of high phylogenetic resolution power, can provide important yardstick to Hungarian biology to understand evolutionary relationships of problematic plant and animal species and groups.

Az elsődleges célkitűzése volt a fajképződés mozgatórugóinak megértése a nőszőfű (Epipactis) nemzetségben. Ehhez mintákat gyűjtöttem a Kaukázustól Walesig a feltételezett szülőfajból (E. helleborine), és ezen a területen belül 28 kisfajról, melyeket az előbbi leszármazottjának tartanak. A RAD-seq módszerrel generált adatok alapján úgy rekonstruáltuk a nőszőfüvek fajképződését, hogy a keresztbeporzó, nagy genetikai diverzitású E. helleborine adja azt a genetikai alapot, melyből a kisfajok leválhatnak. Ebből emelkednek ki a különböző mértékben önmegporzó kisfajok, mégpedig úgy, hogy minél inkább hajlamosak az önmegporzásra, annál nagyobb sebességgel (annál hatékonyabban) izolálódnak. A fenti eredmények nagyszámú taxonómiai következtetés levonását is lehetővé tették, először a fajcsoport történetében. A pályázat emellett felvállalta a bangó (Ophrys) nemzetségen belüli taxonómiai csoportok feltárást is RAD-seq módszerrel. Itt a korábbi megközelítésekhez képest soha nem látott felbontást hozott a genomi módszer alkalmazása, így számos ponton sikerült élesíteni a képet a nemzetség evolúciós közelmúltjáról (ld. Annals of Botany 121(1): 85-105.). A pályázat másodlagos célja volt a RAD-seq módszer hazai implementálása, amelynek sikerét jelzi, hogy a pályázat végén egy újabb hazai pályázatot nyertem el, amely a RAD-seq módsezr alkalmazás körül forog. A pályázat során összesen 15 publikáció született filogenetika és orchidea ökológia témában összesen 34.508 IF eredményezve.
Results in English
The primary aim was the understanding of speciation mechanism in the helleborines (genus Epipactis). To achieve this goal, I have collected samples of E. helleborine, the proposed progenitor species, from the Caucasus Mts. to Wales, plus samples of 28 ‘microspecies’ on the above area which are believed to be derivate species of the previous one. The RAD-seq technique gained a profound insight into the speciation mechanism of helleborines and identified the outcrossing, genetically diverse species E. helleborine as progenitor species that provides genetic background for the isolation of the microspecies. The microspecies with different levels of autogamy can emerge from this; specifically, the more autogam a microspecies is, the more effective and fast its isolation from the ‘plexus’ E. helleborine provides. These results also provided several taxonomic conclusions first in the history of studying this species group. Besides this, we aimed to gain taxonomic insights into the bee-orchids using the RAD-seq method. Here, an unprecedented phylogenetic insight was gained with the genomic approach, thus, we successfully sharpened details of the taxonomy of this notoriously challenging group of orchids. The secondary aim of the researches was the implementation of the RAD-seq method in Hungary, which was successful as I have won a research grant using RAD-seq as the focal point at the end of this one. Altogether, I co-authored 15 publications during this grant totaling 34.508 IF.
