A vírusokról alkotott képünk koránt sem teljes és napjainkban nagymértékben változik. A vírusok a legszámosabb, legsokszínűbb és legsikeresebb "lakói" a földi bioszférának. Konzervatív becslések szerint több mint 100 millió vírusfaj (ezen belül 1 millió gerinces vírusfaj) létezhet, melynek jelenleg csak 0,0025%-át ismerjük. A kutatás célja, hogy korábban nem ismert, új vírusfajokat, vírusnemzetségeket (esetleg víruscsaládokat) azonosítsunk, határozzunk meg, majd elemezzünk egészséges és beteg állatoktól és emberektől származó legkülönbözőbb mintákból. A vizsgálatokhoz elsősorban korszerű, igen hatékony szekvencia-független random amplifikációs molekuláris módszereket alkalmaznánk, melyek képesek gyakorlatilag bármilyen ígéretes kiindulási biológiai mintából, az "idegen/gazda" eredetű nukleinsavak elbontását követően, a vírus-kapsziddal védett nukleinsavak megtartására, annak relatív felsokszorozására (virális metagenomika). Ezt követően nagy áteresztő képességű új generációs szekvenáló és bioinformatikai eszközökkel és módszerekkel határoznánk meg szekvencia virális eredetét nagy adatbázisokkal való összehasonlítás segítségével. Ezzel az eljárással, egyidőben pontosabb, valóságosabb (komplexebb) képet szeretnénk kapni az adott élőlényből származó biológiai minta (minták) virális összetevőiről (virom) és lehetőség szerint taxonómiailag is új vírusok azonosítására törekszünk. A már azonosított és további új vírusok teljes genomjainak meghatározásával a vírusok diverzitásáról, rokonsági viszonyairól, evolúciójáról, társ-fertőzéseiről, fajok közötti átviteléről, kóroki szerepéről szeretnénk új információkat gyűjteni, ezzel bővítve a vírusokkal kapcsolatos ismereteket.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Konzervatív becslések szerint több mint 100 millió vírusfaj (ezen belül 1 millió gerinces vírusfaj) létezhet a földi bioszférában, melynek jelenleg csak 0,0025%-át ismerjük. Ez azt jelenti, hogy a vírusok csak igen szerény (majd hogy nem elhanyagolható) részét ismerjük, azaz, ahhoz, hogy megfelelő mennyiségű és főleg valós ismereteink legyenek a vírusok számáról, jelentőségéről és szerepéről (pl. egyes akut és krónikus kórképekben, a mikrobiális flóra tagjaként, az élő organizmusok evolúciójában vagy akár a földi élet kialakulásában) további intenzív kutatások szükségesek. Napjaink technikai forradalma a molekuláris biológia és a bioinformatika területén új, hatékony eszközöket adtak a kezünkbe és megteremtették a lehetőségét annak, hogy gyakorlatilag bármilyen kiindulási biológiai mintában, egyidejűleg, nagy mennyiségű információ/adat rövid időn belül elemzésével bepillantsunk annak virális összetételébe. Így napjainkban, egy-egy biológiai minta vizsgálata immár komplexebb, valóságosabb képet mutathat a virális alkotókról és a klasszikus virológiai módszerekkel szemben alkalmasabb új vírusok azonosítására is. Vizsgálatainkkal egészséges és beteg állatoktól és emberektől származó legkülönbözőbb típusú mintákból szeretnénk taxonómiailag is új vírusokat azonosítani, majd teljes genomjukat meghatározni. Ennek segítségével a vírusok diverzitásáról, rokonsági viszonyairól, evolúciójáról, társ-fertőzéseiről, fajok közötti átviteléről, kóroki szerepéről szeretnénk új információkat gyűjteni. Vizsgálatainkkal, a jelen tudásunk és kapacitásunk szerint, néhány 10 taxonómiailag is új vírussal növelhetjük az ismert vírusok számát és a vírusokkal kapcsolatos ismereteket.

Napjaink technikai forradalma a molekuláris biológia és a bioinformatika területén új, hatékony eszközöket adtak a kezünkbe és megteremtették a lehetőségét annak, hogy gyakorlatilag bármilyen kiindulási biológiai mintában, egyidejűleg nagy mennyiségű információ/adat rövid időn belül elemzésével bepillantsunk annak valósabb virális összetételébe (virom). Egészséges és beteg állatoktól és emberektől származó legkülönbözőbb típusú mintákból szeretnénk taxonómiailag is új vírusokat azonosítani, majd teljes genomjukat meghatározni. Ennek segítségével a vírusok diverzitásáról, rokonsági viszonyairól, evolúciójáról, társ-fertőzéseiről, fajok közötti átviteléről, kóroki szerepéről szeretnénk új információkat gyűjteni. A kutatás segítségével növelni szeretnénk az ismert, taxonómiailag is új vírusfajok, vírusnemzetségek (esetleg vírus családok) számát, az azonosított vírusok teljes genomjainak meghatározásával és elemzésével. Az eredmények a mikrobiológia, ezen belül a virológia tudományán túl a megelőzés, a humán és állatorvos-tudomány, a közegészségügy-járványügy, az élelmiszer-, vér-, szervellátás biztonság legkülönbözőbb területein közvetlenül is hasznosulhatnak.

A vírusokhoz az átlagemberek körében sokféle hiedelem és félelem kapcsolódik. A vírusokról alkotott kép a kutatók számára sem teljes és napjainkban nagymértékben változik. A vírusok a legszámosabb, legsokszínűbb és legsikeresebb "lakói" a földi bioszférának. Konzervatív becslések szerint több mint 100 millió vírusfaj (ezen belül 1 millió gerinces vírusfaj) létezhet, melynek jelenleg csak 0,0025%-át ismerjük; azaz jelenleg ilyen igen csekély ismeret alapján van bármilyen elképzelésünk is a vírusokról. A kutatás célja, hogy korábban nem ismert, új vírusfajokat, vírusnemzetségeket (esetleg víruscsaládokat) azonosítsunk és írjunk le a ma legkorszerűbbnek számító, egyben igen hatékony szekvencia-független random apmlifikációs/molekuláris/metagenomikai és nagy adatmennyiség feldolgozására alkalmas bioinformatikai módszerek segítségével a legkülönbözőbb mintákból, melyek egészséges, illetve beteg állatoktól, emberektől származnak. Ennek segítségével pontosabb, valóságosabb (komplexebb) képet kaphatunk többek között a vírusok számáról, sokszínűségéről, rokonsági viszonyairól, evolúciójáról, társ-fertőzéseiről, fajok közötti átviteléről, kóroki szerepéről, mely eredmények a megelőzés, a humán és állatorvos-tudomány, a közegészségügy-járványügy, az élelmiszer-, vér-, szervellátás biztonság legkülönbözőbb területein hasznosulhatnak.

Summary of the research and its aims for experts

Our world view about viruses is far from complete and nowadays it is changing fundamentally. Viruses are the most abundant, diverse and successful inhabitants of the Earth’s biosphere. Conserved estimates shows that there are potentially more than 100 million virus species (including 1 million virus species in vertebrates) of which only 0.0025% is presently known. The aims of the study is to identify, characterize and analyze previously unknown novel virus species, genera (maybe family/ies) in wide range of specimens collected from healthy and diseased animals and humans. We would like to use modern, up-to-date and highly effective sequence-independent random molecular amplification methods which are able to maintain and relative over amplification of enriched capsid-protected viral nucleic acid (viral metagenomics) after degradation of “non-viral/host” origin nucleic acids from any promising biological specimens. Subsequently, we would like determine the viral origin of the sequences using high-throughput next generation sequencing and bioinformatics methods with help of online sequence database comparisons. With this procedure, we would like to give more exact, reliable (complex) synchronous view about the viral composition (virome) of the biological specimen(s) of the sampled organisms and to detect taxonomically novel viruses as far as possible. Based upon of the complete genome characterization of novel viruses identified (but nor characterized) previously and will identify during this study we would like to collect/expansion of data/information about the diversity, relationships, evolution, co-infections, interspecies-transmission and etiological role of viruses.

What is the major research question?

Conserved estimates shows that there are potentially more than 100 million virus species (including 1 million in vertebrates) present in Earth's biosphere, however, only the 0.0025% are presently known. This means, that the woefully small (virtually negligible) part of the viruses are known. If we would like an adequate amount, and more importantly, a more real knowledge about the number, importance and role of viruses (e.g. in certain acute/chronic diseases, in microbial flora, in evolution of living organisms or either in creation of Earth’s life) further intense research are needed. The present day technical revolution in molecular biology and bioinformatics give us a novel, effective tools and initiation of the possibility of the real time overview of more exact viral composition (virome) analysis using high amount of data/information within a short period of time in any type of biological specimens. Using these techniques, nowadays, it is possible to collect more exact, reliable (complex) synchronous view about the viral composition (virome) of the biological specimen(s) and it is better suited to detect taxonomically novel viruses than classic virological methods. In this study, we would like to identify and than characterize taxonomically novel viruses in wide range of different type of specimens collected from healthy and diseased animals and humans. With this help, we would like to collect novel information/knowledge about the diversity, relationships, evolution, co-infections, interspecies-transmission and etiological role of viruses. Based upon of our present knowledge (and capacity) it is possible to increase the taxonomically novel viruses with some tens.

What is the significance of the research?

The present day technical revolution in molecular biology and bioinformatics give us a novel, effective tools and initiation of the possibility of the real time overview of more exact viral composition (virome) analysis using high amount of data/information within a short period of time in any type of biological specimens. In this study, we would like to identify and than characterize of the complete genome of taxonomically novel viruses in wide range of different type of specimens collected from healthy and diseased animals and humans. With this help, we would like to collect novel information/knowledge about the diversity, relationships, evolution, co-infections, interspecies-transmission and etiological role of viruses. We would like to expansion of the number of the known, taxonomically novel virus species, genera (maybe family/ies) based upon the complete genome characterization and analysis of the discovered viruses. The results will be utilized directly not only in microbiology especially in virology but also in wide range of areas including prevention, medical and veterinary science, communicable diseases, food-, blood and organ supply safety.

There are plenty of beliefs and fears related to viruses in laymen. The views about viruses among specialist are also far from complete and nowadays it is changing fundamentally. Viruses are the most abundant, diverse and successful inhabitants of the biosphere. Conserved estimates shows that there are potentially more than 100 million virus species (including 1 million virus species in vertebrates) present in Earth's biosphere, however, only the 0.0025% of the virus species are presently known; it means that our present understanding about viruses is woefully poorly. The aims of the study are to detect and characterize novel - previously unknown - virus species, virus genera (possibly virus families) in wide range of specimens collected from healthy and diseased animals and humans using up-to-date, highly effective sequence independent random amplification/molecular/metagenomic and high capacity bioinformatic methods. This study gives us more exact, reliable (complex) view about viruses including the number, diversity, relationships, evolution, co-infections, inter-species transmission and etiological role of viruses which results could be used in wide range of areas including prevention, medical and veterinary science, communicable diseases, food-, blood and organ supply safety.

A 4 kutatási év alatt összesen 21 taxonómiailag új vírusfajt és 10 új vírus nemzetséget azonosítottunk és határoztunk meg, melyeket 35 peer-reviewed tudományos közleményben (ΣIF=133,779) és 22 könyvfejezetben publikáltunk. Az ígéretes mintákat (n=133 pool) virális metagenomikai és új generációs szekvenáló módszerekkel vizsgáltuk. Az új vírusok teljes genomjait kombinált molekuláris módszerekkel határoztuk meg, melyek egy részét laboratóriumunkban fejlesztettük ki. Kombinált módszereket alkalmaztunk az azonosított vírusok számosságának, előfordulásának, diverzitásának, evolúciós kapcsolatainak és potenciális klinikai és etiopatogenetikai hátterének vizsgálatára. A témavezető (RG) részt vehetett kutatási partnerénél egy 21 napos tanulmányúton a Blood Systems Research Institute-ban (San Francisco/USA). Összefoglalva, az ízeltlábúak, halak, kétéltűek, hüllők, madarak és emlősök osztályából állatfajok viromját vizsgáltuk virális metagenomikai/újgenerációs szekvenáló módszerekkel és taxonómiailag új RNS és DNS vírusokat fedeztünk fel 7 víruscsaládból (Picornaviridae, Astroviridae, Parvoviridae, Hepeviridae, Rhabdoviridae, Picobirnaviridae, Retroviridae). Új, genetikailag távoli vírusokat azonosítottunk különböző gazdafajokból és egyes esetekben a pathogenezist is sikeresen feltártuk. Fenntartottuk a nemzetközi együttműködésünket a virális molekuláris epidemiológia és az új vírusok felfedezése területén a világ vezető laboratóriumaival és szakértőivel.
Results in English
During the 4 research years total of 21 taxonomically novel virus species and at least 10 novel virus genera were identified, characterized and published in 35 peer-reviewed scientific papers (ΣIF=133.779) and in 22 book chapters. Promising samples (n=133 pools) were subjected to viral metagenomics and next generation sequencing methods. The complete novel viral genomes were determined by combined molecular methods some of them were developed in our laboratory. Combined methods were also used to investigate the abundance, endemic presence, diversity, evolutional connections and potential clinical and etiopathogenetic background of the identified viruses. The senior scientist (RG) was participated in a 21-day-long study tour at the research partner Blood Systems Research Institute (San Francisco/USA). In summary, the virome of animal species (in class of arthropod, fish, amphibian, reptile, bird and mammal) were investigated using viral metagenomics/next generation sequencing methods and taxonomically novel RNA and DNA viruses related to 7 virus families (Picornaviridae, Astroviridae, Parvoviridae, Hepeviridae, Rhabdoviridae, Picobirnaviridae, Retroviridae) were discovered. Novel, genetically divergent viruses were detected in different host species and in certain cases the pathogenesis was also successfully confirmed. We maintained the international co-operation in the fields of viral molecular epidemiology and novel virus discovery with world-class laboratories and experts.
