A kutatás összefoglalója, célkitűzései szakemberek számára
Itt írja le a kutatás fő célkitűzéseit a témában jártas szakember számára.

Kutatási programunk célja gerinctelen állatok között lezajló kémiai információ csere, kölcsönhatások, illetve a környezetből származó kémiai ingerek feldolgozásának interdiszciplináris úton történő elemzése, továbbá különböző, az alkalmazkodóképesség szempontjából meghatározó a vízi és szárazföldi környezetben előforduló magatartásformák kemorecepcióval történő befolyásolásának feltárása. Kutatásaink során egyrészt különböző kémiai ingerek, valamint növényi eredetű allelokemikáliák hatásait illetve neuronális feldolgozását fogjuk elemezni szárazföldi és vízi gerinctelenek magatartásformáival kapcsolatban, és mindenekelőtt puhatestűek kémiai szignalizációs folyamatait fogjuk feltárni, melyek meghatározzák az egyes környezeti kihívásokra adandó adekvát válaszokat. Kutatásainkat három fő témakörben fogjuk folytatni. i) Kemoszenzoros információk feldolgozásának központi és perifériás szerveződése és celluláris folyamatai gastropoda fajokban illetve a rendszerben bekövetkező lehetséges változások követése kon- és interspecifikus kémiai kommunkáció során. ii) A körrnyezet (kémiai) ingereire válaszoló perifériás végrehajtó (motoros) egységek idegi és neurokémiai szerveződése, különös tekintettel a működéssel és a molekuláris receptorok expressziójával kapcsolatos változásokra. iii) Intracelluláris szignalizációs változások nyomonkövetése faj (egyed)-környezet kölcsönhatások során, különös tekintettel a különböző kémiai ingerek által kiváltott szerv és sejtszintű peptid-protein profil változásokra.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Kutatási eredményeink alapkutatás jellegűek, de a várható eredmények rávilágíthatnak egyes gerinctelen állatcsoportok (puhatestűek) mint modellek esetében az élővilágot és környezetünket meghatározó öko- és populációs stratégiai folyamatokra, az egyed, faj és nagyobb élő együttesek között fennálló kölcsönhatásokra, illetve a kölcsönhatásokat meghatározó szabályozó folyamatok molekuláris, sejt, szerv, szervezet és magatartás szintű törvényszerűségeire. Ezek ismeretében az életfolyamatok és magatartásformák szerveződési, szabályozási, továbbáinter- és intracelluláris szignalizációs törvényszerűségeinek mélyebb megismerése válik lehetségessé, melyek végső soron lehetővé teszik a gerinctelen állatok számára a magas fokú és rendkívül sikeres alkalmazkodó képesség és populációs stratégiák kidolgozását. A tervezett kutatások kísérleti paradigmái, melyek a technikai megközelítések széles sorát foglalják magukba, modellként szolgálhatnak általános evolúciós neuroetológiai és kémiai ökológiai törvényszerűségek feltárásához. Az ilyen folyamatok megismerése nagy jelentőségű lehet a környezet- és a természetvédelem számára is hatékony beavatkozási pontok kijelölése céljából.

Mi a kutatás jelentősége?

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

Környezetünk és a benne létező élő rendszerek együttesének állapotát azok a kölcsönhatások határozzák meg, melyekben a növények és az állatok különböző kémiai jelzőrendszerek útján kialakítják azokat a magatartásformákat, amelyekkel az egyed és/vagy a populáció sikeres fennmaradása és szaporodása biztosított, és ami mint egész az élettér egészséges egyensúlyi állapotának a biztosítéka. Ebben a folyamatban meghatározó szerepet játszanak a törzsfejlődés alacsonyabb fokán álló gerinctelen állatok. A pályázat célja azoknak az érzőrendszereknek a vizsgálata alacsonyabb rendű, vízi és szárazföldi gerinctelen állatfajokban (kagyló és csigák), melyek egyrészt a növények által kibocsátott kémiai anyagok illetve a saját maguk által egymás között használt kémiai közvetítők hatását érzékelik, majd továbbítják az idegrendszer megfelelő szabályozó rendszerei felé, hogy azok végül kialakítsák az egyedek illetve élő együttesek megfelelő (együttműködő, elutasító vagy menekülő) válaszukat az őt körülvevő környezetben.

Summary of the research and its aims for experts

The objective of the research proposal is to analyze the interactions between different invertebrate species and between invertebrates and plants based on chemical stimuli mainly of plant origin, as well as to reveal important behavioral forms involved in complex interactions that occur in the aquatic and terrestrial nature, respectively. The research will focus partly on the effect of specific allelochemicals and partly on the effect of con-and interspecific chemical signaling between aquatic molluscs and its functional and developmental consequences underlying different behaviors to environmental challenges. Our research will deal with three major topics as follows. i) Central and peripheral representation and organization of sensory systems encoding environmental cues, with special attention to possible changes following con- and interspecific chemical communication. ii) Organization of peripheral (effector) organs responding to environmental (chemical) cues, with regard to the function and receptor expression changes upon chemical stimuli. iii) Identification of chemical signals resulting in intracellular biochemical changes during species-environment interactions related to different environmental chemical cues, including intracellular signaling systems and peptide-protein composition.

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.

Plants produce complex mixtures of organic compounds including secondary metabolites, the so called allelochemicals which fundamentally influence the behavior and interaction in the living surrounding. The basic question of the proposal to be investigated is how aquatic and terrestrial invertebrates, first of all mollusks, percept and decode these chemical signals, what are the physiological and behavioral consequences, and how do they elaborate proper responses be the stimulus attractive or repellent when they search for food, feed or develop. As a first step we want to analyze basic questions such as the chemical communication between adult-to-embryo and adult-to-adult as well as the influence of different environmental (chemical) signals on this interaction, both determining the self-maintenance (survival) and reproduction. The organization of the sensory systems and their intercellular signaling processes related to the regulation of the motor activity of the effector organs, both determining the interpretation of chemical (odor) stimuli evoking either approaching (feeding, mating) or escape, will also be studied. Finally the long-term changes of the intracellular molecular cascades and peptide composition evoked by the alterations of the chemical or allelochemical surrounding, both reflecting the cellular consequences of chemical interactions, will be the aim of the project.

What is the significance of the research?
Describe the new perspectives opened by the results achieved, including the scientific basics of potential societal applications. Please describe the unique strengths of your proposal in comparison to your domestic and international competitors in the given field.

The proposal and the planned investigations belong to basic research, however, with possible high practical consequences, concerning certain and less studied aspects of behavioral processes related to environmental interactions in ecosystems. Hopefully novel data will be presented in a yet rather unknown field of evolutionary neuroethology and molecular ecology. By this way, our knowledge will be more complete in understanding the neural basis of different behaviors, making invertebrates to elaborate successful accommodation capacities and population strategies which are responsible for shaping and sustaining the balance in the nature. The different research points of the proposal are strongly related to and represent a “network” of experimental paradigms wherein cellular, chemical and neurochemical as well as behavioral studies would serve as models for analyzing evolutionary neuroethological and chemical ecological rules in the entire animal kingdom. To have an insight in the complicated network of these processes might be of significance for environmental protection, to know how and where to intervene if needed.

Summary and aims of the research for the public

The actual state of our environment and the state of the living systems existing in it are strongly determined by the interactions in which different forms of animal behaviors are influenced by chemical signals emitted by different plants or the animals themselves. The precise execution of these behaviors is also important for the successful survival and reproduction of the individuals, the species and the populations which is the warranty for the optimal balance in the entire ecosystem as well. In this complicated process an important role is played by the invertebrate fauna. The aim of the planned investigations are to analyze and characterize the sensory systems of lower aquatic and terrestrial invertebrate species (bivalves and snails) which are responsible to detect the chemical signaling molecules playing role in their interactions both among them and with the environment, as well as affecting their development and corresponding (co-operative, dismissive or escape) behavior when responding to the environmental challenges.

Kutatási programunk célja kemoszenzoros rendszerek szerepének feltárása volt puhatestűek adaptív magatartásában, melynek során adatokat kívántunk nyerni az ökoszisztémában lezajló kölcsönhatásokról, különös tekintettel egyedfejlődési, funkcionális neuroanatómiai és fiziológiai folyamatokra. Megállapítottuk, hogy i) az invazív vándorkagyló (Dreissena sp.) korai embrionális érzőrendszerét 5-HTerg és peptiderg (FMRamid) elemek alkotják, melyek alapvető szerepet játszanak a mozgásaktivitás szabályozásában; ii) perifériás kolinerg, aminerg és peptiderg szenzo-motoros rendszerek komplex intercelluláris kapcsolatokat alkotnak a nagy mocsári csigában (Lymnaea); iii) allelokemikáliák (tanninsav) közvetlenül nem, csak a perifériás érzőrendszer közvetítésével hat a táplálkozási aktivitást szabályozó központi neuronhálózatra; iv) nitrogén monoxid (NO) volumen, szinaptikus és ko-transzmitterként is funkcionálhat a központi idegrendszerében (Helix, Lymnaea); v) a szaglóközpont neuronjaiban kimutatott NO/cGMP/PKA szignalizációs út meghatározó szerepet játszik az étcsiga, Helix, táplálkozási magatartásának szabályozásában; vi) Lymnaea ritmikus táplálkozási aktivitásának szabályozásában az 5-HT egy gátló 5-HT1 és egy serkentő 5-HT6,7 receptor útján vesz részt; vii) humán fogamzásgátlók és aktív bomlástermékeik gátolják vízi gerinctelenek embriógenezisét (Lymnaea), szaporodását (Daphnia), illetve a táplálkozási neuronhálózat (Lymnaea) működését.
Results in English
Our research project aimed at the analysis of the role of chemosensory systems in adaptive behavior of mollusks, in order to get insight into species-environment interactions in the ecosystem. Special attention was paid to system organization, functional neuroanatomy and physiological aspects. It has been shown that i) early embryonic sensory systems consist of 5-HTergic and peptidergic (FMRFamide) elements in the invasive bivalve (Dreissena sp.) that are involved in the regulation of the embryonic swimming behavior, including response to chemical stimuli; ii) peripheral cholinergic, aminergic and peptidergic sensory-motor elements form a complex intercellular connection system in the pond snail, Lymnaea; iii) the allelochemical, tannic acid influences the behavior (locomotion, feeding) of Lymnaea through the peripheral sensory system, without affecting directly the central regulatory network, iv) NO can function as volume, synaptic and co-transmitter in the Helix and Lymnaea CNS; v) an NO/cGMP/PKA signaling pathway in the olfactory lobe (procerebrum) cells is involved in the regulation of the feeding behavior of the terrestrial snail, Helix; vi) 5-HT exerts in its regulation on Lymnaea feeding activity through an inhibitory 5-HT1-like and an excitatory 5-HT6,7-like receptor; vii) human contraceptives and their active degradation products represent a real negative challenge to the development (Lymnaea), reproduction (Daphnia), and feeding network (Lymnaea).
