A növényi betegség ellenállóság során a reaktív oxigénfajták (ROF) kis koncentrációban a rezisztencia jelátvivői, míg nagy mennyiségben a gazdanövényt és a kórokozót is károsíthatják. Korábbi kutatásaink szerint a tünetmentes ún. nemgazda rezisztencia fő jellemzője a ROF gyors és korai felhalmozódása. Jelen pályázat keretében tisztázni kívánjuk, hogy a ROF felhalmozódás önmagában képes-e a hatékony - tünetmentes - növényi betegség ellenállóság kialakítására? Dohány modellnövényben vizsgáljuk a burgonya X vírus (PVX) ellen ható, tünetmentes (extrém) rezisztenciát meghatározó Rx1 gén ROF (szuperoxid és hidrogén-peroxid) termelésre gyakorolt hatását. Ha a ROF felhalmozódás valóban az extrém rezisztencia meghatározó tényezője, a gazdanövény antioxidáns kapacitásának fokozása (keresztezéssel, antioxidánsok infiltrálásával, ill. antioxidáns gének tranziens expressziójával) csökkenti vagy megszünteti a PVX-szel szembeni rezisztenciát, míg a fogékony dohány kezelése ROF-generáló ágensekkel képes legalább részleges PVX rezisztenciát kiváltani.
Előzetes kísérleteink szerint egy lisztharmat tüneteknek ellenálló hazai cseresznyepaprika fajta (cv. Szentesi) leveleiben fokozott szuperoxid felhalmozódás detektálható. Mind a rezisztencia, mind a szuperoxid felhalmozódás kertészeti oltással átvihető egy fogékony étkezési paprika fajtára (cv. Totál). Tisztázandó, hogy a szuperoxid-felhalmozódás tényleges szerepet játszik-e a lisztharmat rezisztenciában, ill. más (vírus) kórokozókkal szembeni védekezésben? Ezek a kutatások hozzájárulhatnak a gazdaságilag is jelentős tünetmentes növényi betegség rezisztencia mechanizmusának megértéséhez.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Saját korábbi kutatásaink szerint a tünetmentes, ún. nemgazda rezisztencia (= rezisztencia a más gazdanövényre specializálódott kórokozóval szemben) fő jellemzője a ROF (elsősorban a szuperoxid) gyors és korai felhalmozódása. Hasonló tünetmentes rezisztenciát idéztünk elő dohány mozaik vírussal (TMV) szemben is, ha a fogékony dohány növényeket ROF-generáló ágensekkel kezeltük a fertőzés utáni korai időpontban. Előkísérleteink szerint egy lisztharmat tüneteknek ellenálló hazai cseresznyepaprika fajta (cv. Szentesi) leveleiben szintén fokozott szuperoxid felhalmozódás detektálható.
A pályázat keretében tisztázandó, hogy a ROF felhalmozódás önmagában képes-e a gyors és hatékony - tünetmentes - növényi betegség ellenállóság kialakítására? A kérdés megválaszolásához kétféle kísérleti megközelítést tervezünk:
1/ A korai ROF felhalmozódás szerepe a vírusok elleni tünetmentes, ún. extrém rezisztenciában. A burgonya X vírus (PVX) elleni extrém rezisztenciát meghatározó Rx1 gént kifejező dohányban csökkenhet a vírus rezisztencia, amennyiben a gazdanövény antioxidáns kapacitását mesterségesen fokozzuk. Ebben az esetben feltételezhető az is, hogy a PVX-re fogékony dohány kezelése ROF-generáló ágensekkel képes legalább részleges PVX rezisztenciát kiváltani.
2/ Feltételezhető, hogy a fokozott szuperoxid felhalmozódást mutató cseresznyepaprika lisztharmat ellenállósága csökkenhet, ha a növény antioxidáns kapacitását növeljük. Ugyanakkor a ROF-generáló ágensekkel történő kezelés fogékony paprikában legalább részleges lisztharmat rezisztenciát válthat ki. A szuperoxid termelő cseresznyepaprika más (vírus) kórokozókkal szembeni rezisztenciáját is vizsgáljuk.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

A pályázat keretében elvégzendő kutatások tisztázhatják a gazdasági szempontból is jelentős tünetmentes növényi betegség rezisztencia egyik alapvető mechanizmusát, amelyben szerepet játszhat a ROF (pl. szuperoxid és hidrogén-peroxid) kórokozó behatoláshoz köthető gyors felhalmozódása. Új perspektívát jelenthet a termesztett növények ROF-túltermelésre történő szelekciója, az ilyen növények potenciálisan tünetmentes rezisztenciát mutathatnak akár többféle kórokozóval szemben is. A szuperoxid-felhalmozó cseresznyepaprika (cv. Szentesi) lisztharmatos tünetekkel szembeni rezisztenciája gyakorlati szempontból szintén fontos lehet, mivel az üvegházi paprikatermesztésben (a lisztharmat-betegség döntően itt jelent veszélyt) csökkentheti a nagymértékű és költséges növényvédőszer felhasználást, ill. az azzal járó környezetszennyezést.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

A növények képesek ugyan sikeresen védekezni az őket támadó kórokozók fertőzésével szemben, de ezt legtöbbször valamilyen nyilvánvaló mellékhatással is jár, pl. a megtámadott növényi szövetek elhalásával, lassabb növekedéssel, stb. A betegségek elleni védekezés akkor hatékony, ha gyors lefolyású, a kórokozót hamar hatástalanítja és így a növényi szervezet erőforrásait minél kevésbé köti le. Ilyenkor a gazdanövény tünetmentesen képes az adott kórokozó fertőzésének ellenállni. A növénynemesítés, ill. a mezőgazdasági gyakorlat számára ezért fontos lehet az olyan termesztett növényfajták előállítása, amelyek egy vagy több kórokozóval szemben tünetmentesen képesek védekezni.
A növényi betegség ellenállóság folyamatában fontos szerepet játszanak az ún. a reaktív oxigénfajták (ROF), amelyek minden élő szervezetben megtalálhatók. Növényekben ezek az anyagok kis mennyiségben többek között a fertőzések elleni védekezés irányítói, míg ha nagy mennyiségben és hosszú ideig termelődnek, a kórokozót és a gazdanövényt egyaránt károsíthatják.
A pályázat segítségével azt szeretnénk tisztázni, hogy a reaktív oxigénfajták gyors, koncentrált, a kórokozó behatoláshoz köthető felhalmozódása önmagában képes-e a hatékony - tünetmentes - növényi betegség ellenállóság kialakítására? Kísérleteinkben ezért azt vizsgáljuk, hogy
1/a ROF-vegyületek felhalmozódásának mesterséges visszaszorítása csökkenti-e a növények tünetmentes betegség-ellenállóságát
2/ növények ROF-vegyületekkel történő kezelése képes-e a tünetmentes betegség ellenállóság kialakítására?
Ezek a kutatások tisztázhatják a gazdasági szempontból is jelentős tünetmentes növényi betegség rezisztencia egyik alapvető mechanizmusát.

Reactive oxygen species (ROS) have a dual function during plant disease resistance. In low concentrations, ROS transduce resistance signals, while in high amounts they may damage both the host plant and the pathogen. According to our previous research, a main characteristic of symptomless non-host resistance is the rapid, early accumulation of ROS. In this project, we will clarify if ROS accumulation per se is capable to elicit effective, symptomless plant disease resistance. Tobacco plants will be used as a model to investigate the effect of the Rx gene determining symptomless (extreme) resistance to Potato virus X (PVX) on the production of ROS (superoxide, hydrogen peroxide). If ROS accumulation is indeed a key factor of extreme resistance, enhancing antioxidant capacity of the host (by crosses, infiltration of antioxidants, transient expression of antioxidant genes) should reduce or eliminate resistance to PVX. On the other hand, treatment of susceptible tobacco with ROS-generating agents will likely induce at least a partial resistance to PVX.
Our preliminary experiments show that enhanced accumulation of superoxide occurs in leaves of Hungarian cherry pepper (cv. Szentesi) resistant to powdery mildew symptoms. Both superoxide accumulation and resistance can be transmitted to a susceptible sweet pepper cultivar (´Totál´) by grafting. It should be clarified whether superoxide accumulation indeed plays a pivotal role in resistance to powdery mildew and other (virus) pathogens? This research will contribute to a better understanding of the mechanism of economically important symptomless plant disease resistance responses.

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.

According to our previous research, the main characteristic of symptomless non-host resistance (i.e. resistance to pathogens adapted to other host plants) is the rapid, early accumulation of ROS, primarily superoxide. We could elicit a similar, symptomless resistance to Tobacco mosaic virus (TMV), if susceptible plants were treated with ROS-generating agents early after inoculation. Our preliminary experiments demonstrate that enhanced accumulation of superoxide also occurs in leaves of Hungarian cherry pepper (cv. Szentesi) resistant to powdery mildew.
In the present project we will investigate whether ROS accumulation per se is capable to elicit rapid, effective, symptomless plant disease resistance responses. Two experimental approaches will be applied:
1/ Role of early ROS accumulation in the symptomless, so-called extreme resistance to viruses. In tobacco that express the Rx gene determining extreme resistance to Potato virus X (PVX) virus resistance could be reduced if antioxidant capacity of the host is artificially enhanced. In this case it is also likely that treatment of PVX-susceptible tobacco with ROS-generating agents will induce at least a partial resistance to the virus.
2/ It is anticipated that powdery mildew resistance of cherry pepper displaying enhanced superoxide accumulation will be reduced if antioxidant capacity of the host is enhanced. However, treatment of susceptible pepper with ROS-generating agents should confer at least a partial resistance to powdery mildew. We will also investigate resistance of superoxide-accumulating cherry pepper to other (virus) pathogens.

What is the significance of the research?

Research planned in the framework of this proposal may clarify a basic mechanism of economically important symptomless plant disease resistance, where the rapid accumulation of ROS (e.g. superoxide and hydrogen peroxide) associated with pathogen ingress may play a role. A new perspective in this regard could be the selection of crop plants for ROS-overproduction, since such plants may potentially display symptomless resistance to one or more pathogens. Resistance of superoxide-accumulating cherry pepper (cv. Szentesi) to powdery mildew symptoms is also significant from a practical point of view, since it may reduce large scale and expensive pesticide use and associated pollution in greenhouse pepper production (powdery mildew disease is a hazard especially under such conditions).

Although plants can successfully defend themselves against the infection of invading pathogens, this is usually accompanied by obvious side-effects, e.g. the death (necrosis) of invaded plant tissues, slower growth, etc. Defense against diseases is effective if it is a rapid process, eliminating the pathogen in due time and not overusing resources of the plant organism. In this case the host plant is able to resist infection by a given pathogen without the development of symptoms. Therefore, introduction of crop cultivars that display symptomless resistance to one or more pathogens is of primary importance for plant breeding and crop production.
Reactive oxygen species (ROS) can be universally found in living organisms and they play important roles in the process of plant disease resistance as well. In plants these compounds, when present in small amounts, are involved e.g. in orchestration of defense to pathogenic infections. However, when produced in large amounts for prolonged periods, ROS may harm the host plant and the pathogen as well.
With the aim of this project we should clarify whether rapid, concentrated ROS-accumulation associated with pathogen ingress is capable in itself to elicit an effective, symptomless plant disease resistance? Therefore, we will investigate whether
1/ artificial suppression of ROS-accumulation can reduce symptomless disease resistance in plants
2/ plant treatments with ROS can develop symptomless disease resistance?
These studies should clarify a basic mechanism of economically important symptomless disease resistance in plants.

Kimutattuk, hogy egy reaktív oxigénfajta (ROS) a szuperoxid (O2•–) felhalmozódása fontos tényezője a tünetmentes (extrém) rezisztenciának: 1/ dohányban a burgonya X vírussal (PVX) szembeni tünetmentes (HR-típusú nekrózis nélküli) ún. extrém rezisztenciának (ER), 2/ paprikában a lisztharmattal (Leveillula taurica) szembeni, oltással átvihető tünetmentes rezisztenciának. A szuperoxid (ROS) felhalmozódás csökkentése a PVX-szel szembeni ER során részlegesen megtöri az ER-t, míg a szuperoxid szint növelése PVX-re fogékony dohányban részleges rezisztenciát idéz elő: HR-szerű nekrotikus tünetek jelennek meg és csökken a PVX titer. Lisztharmat-rezisztens paprikában (cv. Szentesi, Szentesire oltott cv. Totál) a szuperoxid felhalmozódás korrelál a NADPH-oxidáz aktivitással és az oltással átvihető tünetmentes rezisztenciával. a rezisztencia biokémiai markerei (NADPH oxidáz aktivitás, magas CaPR1, CaPR2, alacsony CaMlo1, CaMlo2 génexpresszió) jól öröklődnek a lisztharmat rezisztens paprika utódaiban (rezisztens és fogékony növények keresztezésének, ill. az oltott, rezisztens növények utódaiban). Eredményeink szerint a szuperoxid (ROS) felhalmozódás és a szuperoxid termelést szabályozó gének expressziója szorosan összefügg a tünetmentes (extrém) rezisztenciával, amely oltással vagy keresztezéssel átvihető a kórokozókra fogékony növényekbe. További kutatások szükségesek ahhoz, hogy pontos képet kapjunk a szuperoxid (ROS) tünetmentes növényi betegség rezisztenciában játszott szerepéről.
Results in English
We have shown that accumulation of the plant reactive oxygen species (ROS), superoxide (O2• –) contributes to symptomless (no HR-type necrosis) resistance: 1/ in tobacco, extreme resistance (ER) to Potato virus X (PVX) and 2/ in pepper, graft-transmissible resistance to powdery mildew (PM, Leveillula taurica). Reduction of superoxide (ROS) accumulation during ER to PVX partially breaks ER, while elevation of superoxide levels in PVX susceptible tobacco confers partial PVX resistance: HR-necrosis appears and PVX titers decrease. In PM-resistant pepper (‘Szentesi’, Szentesi+Totál grafts), superoxide accumulation correlates with NADPH oxidase activity and graft-transmissible PM resistance. Biochemical markers of graft-transmissible PM resistance (NADPH oxidase activity, high CaPR1, CaPR2, low CaMlo1, CaMlo2 expression) are inherited in progeny of resistant pepper (in crosses of resistant and susceptible plants and progeny of grafted, resistant plants). Taken together, our results demonstrate that superoxide (ROS) accumulation and expression of defense-related genes regulating superoxide are tightly associated with symptomless (extreme) resistance that is transmissible to susceptible plants by grafting or crossing. Further research should clarify function(s) of superoxide in symptomless plant disease resistance.
