A rheumatoid arthritis súlyos autoimmun megbetegedés, melynek pathomechanizmusa a mai napig nem teljesen ismert. A kezelésére használt szereknek súlyos mellékhatásai vannak. Az elmúlt évtizedben a hidrogén-szulfid (H2S) a farmakológiai kutatások előterébe került. A H2S szerepe az állati és emberi gyulladásos betegségekben bizonyított. A TRPA1 receptor a nociceptív primer szenzoros neuronokon expresszálódik. Az utóbbi időben bizonyították, hogy a H2S aktiválja a TRPA1 receptort és neuropeptid (pl. szomatosztatin) felszabadulást okoz. A szomatosztatin gyulladásgátló hatását az sst4 receptoron keresztül váltja ki. A TRP csatornák szerepet játszanak az arthritis pathomechanizmusában. Új adatok szerint a TRP csatornák közül a TRPC5 (TRP canonical 5) szintén részt vesz az ízületi gyulladás kifejlődésében. Az együttműködés célja akut és krónikus ízületi gyulladásos egérmodellek kifejlesztése (CFA kiváltotta akut és krónikus ízületi gyulladás, immuntranszfer indukálta polyarthritis) és vizsgálata mindkét kutatóhelyen. Felhasználva az angol és magyar laboratóriumok közös munkáinak eddigi eredményeit megvizsgáljuk, hogy az exogén és endogén H2S milyen hatásokat fejt ki a fenti TRP csatornák közvetítésével. Célunk felderíteni a H2S TRP csatorna-függő és attól független hatásait akut és krónikus ízületi gyulladás modellekben és bizonyítani az sst4 szomatosztatin receptor esetleges szerepét a gyulladásgátló hatásban. A pályázati támogatás segítségével magyar és brit PhD hallgatók kölcsönös tanulmányútjait kívánjuk elősegíteni a két laboratórium között.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Feltételezésünk, hogy a hidrogén-szulfid TRP és sst4 receptorokhoz kapcsolható mechanizmusok által tudja az ízületi gyulladást befolyásolni. Kutatásunk a következő kérdésekre ad választ:
1. Hogyan befolyásolja az exogén és endogén H2S az akut és krónikus arthritis patomechanizmusát egérmodelljeinkben?
2. Van-e szerepe a TRPA1 receptoroknak a H2S ízületi gyulladásban kifejtett hatásainak közvetítésében?
3. Hatással lehet-e a H2S egyéb TRP csatornák, mint például a TRPC5 csatorna, működésére az arthritis súlyosságának befolyásolásában?
Célunk, hogy egér arthritis modelljeink segítségével feltérképezzük azokat a mechanizmusokat, melyekkel a H2S és a TRP csatornák valamint a szomatosztatin képesek a fájdalmat és gyulladást befolyásolni. Komplex vizsgálatot tervezünk funkcionális (dinamikus plantáris eszteziometria, plethysmometria, digitális mikrométer), morfológiai (in vivo fluoreszcens és lumineszcens képalkotó vizsgálatok, szövettan, micro CT), kémiai (H2S meghatározás amperometriával) és immunológiai (gyulladásos sejtek elemzése flow cytometriával, gyulladásos mediátorok meghatározása ELISA-val, gerincvelői c-Fos immunhisztokémia) módszerekkel.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

Kísérleteink eredményeket fognak szolgálni a H2S szerepére akut és krónikus arthritis modellekben, valamint, hogy ezek a hatások TRPA1, TRPC5 vagy sst4 receptor által mediáltak vagy ezektől függetlenek. Eredményeink új perspektívákat kínálhatnak a gyógyszerfejlesztés számára a rheumatoid arthritis kezelésének területén. Ennek eredményeképpen a jövőben lehetőség nyílhat a TRPA1, TRPC5 és az sst4 receptor ligandok, H2S-donorok vagy endogén H2S szintézist befolyásoló szerek klinikai gyakorlatba való beillesztésére.

A reumatoid artritisz gyulladásos autoimmun betegség. A betegség előrehaladó és súlyos rokkantságot okozhat. Kialakulásának mechanizmusa nem teljesen ismert, kezelése máig nem teljesen megoldott. Az antirheumás szereknek számos súlyos mellékhatása van. A betegség molekuláris mechanizmusának és befolyásoló tényezőinek jobb megértése hatékonyabb, kevesebb mellékhatással bíró gyógyszeres kezelés kifejlesztését segítheti elő.
A hidrogén-szulfid (H2S) az emberi test endogén gáz halmazállapotú jelátvivő molekulája, ami különböző élettani és kórélettani folyamatokban vesz részt. Ismert, hogy szerepet játszik az ízületi gyulladásban, de ennek pontos mechanizmusa még nem tisztázott. A legújabb adatok szerint a H2S gyulladásgátló hatású lehet ízületi gyulladásban. Neurogén faktorok – különösen az érző idegrostokból felszabaduló neuropeptidek – befolyásolhatják a gyulladásos elváltozásokat az ízületekben. Az érzőideg végződések TRP csatornáit különböző kémiai ingerek aktiválhatják, köztük a H2S-sel. Pályázatunk keretében szeretnénk megvizsgálni az endogén és exogén H2S szerepét és pontos hatásmechanizmusát, valamint a TRP csatornák közvetítette összetevők részvételét ízületi gyulladás egérmodelljeiben. A jelen kutatás célja új célmolekulák azonosítása és a hozzájuk kötődő vegyületek gyulladásos folyamatok elleni hatékonyságának igazolása reumatoid artritisz állatmodelljeiben.

Rheumatoid arthritis (RA) is a serious inflammatory autoimmune disease. Its pathomechanism has not been elucidated yet. Anti-rheumatoid drugs elicit severe adverse effects. Over the last decade hydrogen sulfide (H2S) has arisen to the front line of pharmacological research. Participation of H2S in animal and human inflammatory diseases has been established. TRPA1 receptor is expressed in nociceptive primary sensory neurons. Recently, H2S has been reported to activate neuronal TRPA1 receptors and to elicit neuropeptide (e.g. somatostatin) release. Anti-inflammatory effect of somatostatin is mediated by sst4 receptors. It is now realised that other TRP receptors have the potential to be involved in arthritis. Of these TRPC5 (TRP canonical 5) is under early study. The principal aims of the present study are to develop murine models of acute and chronic arthritis that we can both use in our grants and this is the nature of the collaboration. The present application aims to further understand the role of the interaction of H2S with TRPA1 and TRPC5 receptors in animal models of acute and chronic arthritis and determine the possible role of somatostatin in anti-inflammatory changes. Here, we are planning to use CFA-evoked knee inflammation as an acute arthritis model. CFA- and immune transfer-induced polyarthritides will serve as chronic models of joint inflammation showing more similarities to human RA. With the help of the grant we are planning bilateral visits for the Hungarian and British Ph.D. students between the two laboratories.

What is the major research question?

The hypothesis is that hydrogen sulfide interacts with TRP and somatostatin-related mechanisms to influence the pathophysiology of arthritis.
2.1 How does the endogenous or exogenous H2S influence the pathomechanism of acute and chronic arthritis in our animal model?
2.2 Does the TRPA1 receptor play a role in the mediation of the effects of H2S in joint inflammation?
2.4 Can H2S influence other TRP receptors such as the TRPC5 receptor to influence arthritis?
2.3 Do H2S and TRPA1 interactions influence somatostatin release? We would also like to investigate involvement of somatostatin-mediated anti-inflammatory components using sst4 receptor knockout mice.
2.4 The aim is to utilise murine animal models of arthritis to analyse possible mechanisms by which H2S and the TRP receptors, together with somatostatin are involved in influencing the pain and inflammation. We will provide a comprehensive analysis of arthritis using functional (dynamic plantar aesthesiometry, plethysmometry, digital micrometer), morphological (in vivo fluorescent and luminescent imaging, histology), biochemical (amperometric detection of H2S levels) and immunological (characterization of inflammatory cells by flow cytometry and detection of inflammatory mediators by ELISA, immuno-histochemical detection of spinal c-Fos expression) methods.

Our proposed project will provide valuable data on how H2S influences murine models of acute and chronic arthritis that we know to be influenced by TRPA1, TRPC5 and sst4 mediated components or independently from these receptor structures. Our results will allow us and others to translate TRPA1, TRPC5 or sst4 receptor ligands or H2S donors and modulators of endogenous H2S synthesis into clinical therapy of arthritis in the future. The results of the basic science could open new ways for the drug developmental processes against the rheumatoid arthritis.

Rheumatoid arthritis is regarded as an inflammatory autoimmune disease. It is progressive and can result in serious disability of the patients. Its pathomechanism has not been fully elucidated, the therapy is still an unresolved issue. Antirheumatoid agents have a wide range of serious adverse effects. Better understanding of the molecular pathomechanism and modulatory factors would be a new gateway to develop more efficient, less harmful pharmacotherapy of the disease.
Hydrogen sulfide (H2S) is an endogenous gaseous mediator in the human body, which mediates different physiological and pathophysiological processes. It is known to be involved in arthritis, but it is too early to understand the precise mechanisms. According to recent data, H2S appears to play an anti-inflammatory role in arthritis. Neurogenic factors, especially neuropeptides released from the sensory fibres can modulate the inflammatory changes in the joint. TRP channels on the sensory nerve endings can be activated by different chemical stimuli, such as H2S. In our study we will investigate the role and precise mechanism of action of endogenous and exogenous H2S as well as the participation of TRP channel-mediated components of joint inflammation in murine models. The aim of the present research is to identify novel drug targets and prove the efficiency of their ligands against inflammatory processes in animal models of rheumatoid arthritis.

A hidrogén szulfid (H2S) egy gázmediátor, ami szerepet játszik számos fiziológiai és pathofiziológiai mechanizmusban, mint például a neuroimmun moduláció, metabolikus változások, gyulladás, fájdalom, stb. Az érzőidegeken és a nem-neurális sejteken levő tranziens receptor potenciál (TRP) kation csatornák mechanikai, termális és kémiai stimulusokkal aktiválhatók. Projektünkben kimutattuk, hogy a TRPA1 receptor izgatása H2S donorok és poliszulfidok által gyulladásgátló hatású az ízületi gyulladás állatmodelljeiben. Mivel a receptor stimulációja a gyulladásgátló neuropeptidként jól ismert szomatosztatin felszabadulását fokozza, feltételezhető, hogy részben ez a peptid felelős az artritiszes tünetek csökkenéséért. Munkánkban szintén rámutattunk arra, hogy TRPA1 hiányos, artritiszes állatokban a sulfidok fokozták a fájdalmat és a gyulladást. Eredményeink alapján projektunk fő megállapítása az, hogy ízületi gyulladás állatmodelljeiben a szulfid vegyületek a TRPA1 receptor jelenlétében gyulladásgátló hatásúak lehetnek, viszont a receptor hiányában inkább aktiválják a gyulladásos kaszkádot.

Hydrogen sulfide (H2S) is a gaseous mediator in various physiological and pathological processes, including neuro-immune-modulation, metabolic pathways, inflammation and pain etc. Transient Receptor Potential (TRP) cation channels on the sensory nerves and non-neuronal cells can be activated by mechanical, thermal or chemical triggers. In our project we have shown, that activation of TRPA1 receptors by H2S donors or polysulfides exerts an anti-inflammatory role in the animal models of arthritis. Since stimulation of TRPA1 channels resulted in increased release of the anti-inflammatory neuropeptide somatostatin, we presume that somatostostatin is partly responsible for the reduction of arthritic changes. Our work also also pointed out that sulfides and polysulfides increase pain and inflammation in the TRPA1 KO animals. The main conclusion of our study is that sulphide compounds can activate anti-inflammatory changes in the joint in the presence of TRPA1 receptors but they could trigger the inflammation cascade in the absence of the receptor.
