Harántcsíkolt izom nyújtása során passzív erő ébred aminek kialakításáért elsősorban a szarkomerekben található titin molekula felel. Bár az egyetlen titin molekulára ható in vivo erő értéke pontosan nem ismert, valószínűsíthető, hogy a szarkomerek nyújtásakor a titin nem csak rugalmasan nyúlik, de egyes doménjei ki is tekeredhetnek. A gombolyodási folyamat, mely során a kontrahát molekula natív szerkezetét, mindmáig pontosan nem ismert. Korábbi munkáinkban kimutattuk, hogy a molekula mechanikailag gyengébb doménjei élettanilag releváns erőtartományban kitekerednek és ugyancsak élettanilag releváns idő- és megnyúlásbeli skálán visszagombolyodnak. Lézercsipesszel végzett kísérletekben, a kitekert molekulát igen alacsony erő értékeknél tartva (<1 pN) erőfluktuációk tapasztalhatók, melyek feltehetően egy kitekert illetve egy kompaktabb molten globule állapot közötti gyors átmenetek következményei. Habár az egyedi molekulamanipulációs kísérletek arra engednek következtetni, hogy a titin gombolyodásában egy molten globule köztes állapot is szerepet játszik, az egy titinen belüli nagyszámú domének jelenléte miatt az egyedi állapotok időben és térben kiátlagolódnak, így azok nehezen azonosíthatók. Jelen pályázat célja a molten globule állapothoz kötött dinamika vizsgálata egyedi titin globuláris domének gombolyodási folyamatában. Ennek érdekében a kutatás során a teljes titinmolekula manipulációja helyett egy kettős szálú DNS fogantyú alapú rendszerben tervezem egyedi globuláris titin domének nanomanipulációját nagy felbontású erő-visszacsatolt lézercsipesszel. Ezen kísérletek várhatóan jobb megértését teszik lehetővé a domének gombolyodási folyamatának.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A kutatás célja a titinmolekula gombolyodási folyamatának részletes megismerése.
Az eddig elért eredményeink tükrében a kutatás kiinduló hipotézise: A titin domének gombolyodása egy molten globule intermedier állapoton keresztül zajlik. A hipotézis tesztelése csak egyedi domének vizsgálatával érhető el, mivel a titinben található több száz domén együttes gombolyodása megakadályozza diszkrét állapotok azonosítását. Terveimben szerepel egy immunoglobulin és egy fibronektin típusú domén illetve az ezekből képezett kiméra vizsgálata, mint a titin doméneket modellező egyszerű rendszerek. Megválaszolandó kérdések:
1. Milyen állapotokon keresztül történik a natív szerkezetbe gombolyodás mechanikailag denaturált titin domének esetén?
2. Milyen dinamikával történik az egyes állapotok közötti átmenet?
3. Milyen módon alakítják ki az egyedi domének gombolyodási folyamatai a teljes titinmolekulára jellemző kollektív gombolyodási viselkedést?

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

Bár már évek óta ismert, hogy az erőhatásra kitekert titinmolekula relaxációja során képes újra felvenni natív szerkezetét, a gombolyodási folyamat részleteiben nem ismert. Mindezidáig az erő vezérelt titin gombolyodási folyamatokat egy egyszerű két állapotú rendszer segítségével modellezték. Előzetes kísérleteink felvetik egy gombolyodási intermedier, feltehetően molten globule állapot jelenlétét is. A projekt révén választ kaphatunk a titin domének gombolyodási folyamatának részleteire, jobban megismerhetjük a molten globule állapot fehérjegombolyodásban betöltött szerepét. A molten globule állapot kimutatása amiatt jelentős, mert felvetődik egy újfajta molekulakontrakciós mechanizmus lehetősége, ami nem pusztán a kitekert lánc entrópikus rugalmasságán, hanem a molten globule konformáció dinamikáján is alapszik. Ez a kontrakciós mechanizmus fontos szerepet tölthet be olyan szarkomert ért stressz esetén ami domének kitekeredését okozza. Ilyen szarkomerikus stressz különösen fontos lehet olyan kardiológiai kórképekben, ahol a titin gént érintő deléciós mutációk révén rövidebb izoforma expresszált, ami révén növekszik a molekula feszítettsége, így a doménkitekeredés valószínűsége is.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

Az izomszövet rugalmasságának kialakításáért elsősorban a rugalmas titin izomfehérje felelős. A titin, az emberi szervezetben fellelhető legnagyobb fehérjemolekula, mikroszkopikus rugóként feszül ki az izomrostokat felépítő alapegységekben, a szarkomerekben. A fehérje rugalmasságának következtében képes megnyúlni és összehúzódni az izomműködés során. A titin erőhatásra először megnyúlik, majd a benne található domének szerkezete az erőhatásra megbomlik és a domének kitekerednek. Ha ezután hagyjuk a molekulát relaxálni, a domének visszaveszik szerkezetüket, felgombolyodnak. A felgombolyodás folyamata mindmáig nem pontosan ismert. Előzetes kísérleteink eredményei alapján, melyekben teljes hosszúságú egyedi titin molekulákat nyújtottunk lézercsipesszel, azt feltételezzük, hogy a gombolyodás egy molten globule közti állapoton keresztül zajlik. Ennek vizsgálatára egyedi titin doméneket tervezünk manipulálni molekuláris erőmérő lézercsipesszel, annak érdekében, hogy feltérképezzük a gombolyodásban szerepet játszó egyes állapotokat. Az eredmények segítenek pontosabban megérteni a titin gombolyodáson keresztüli rövidülését, így egyes szívelégtelenségek esetén a szöveti rugalmasság csökkenésének mechanizmusát.

During muscle stretch passive force develops that is mainly determined by the giant sarcomeric protein titin. Upon stretch, titin is thought to expand in a hierarchical order, according to the different elasticity and stability of its molecular regions. However, the exact force acting on a single titin molecule in vivo is not precisely known. Because force induces unfolding, it has been suggested that titin’s sarcomeric extension may also involve the unfolding of its domains. The sarcomeric unfolding of titin is especially relevant in the case of titin-truncation-linked pathologies, in which the dynamics between the unfolded and refolded states may play an additional role in the molecule’s mechanics. In the previous works my research group has demonstrated that mechanically weak globular domains of titin unfold at physiologically relevant forces and refold within physiologically relevant spatial and temporal scales. Although titin refolds if incubated in the contracted state, the exact trajectory of the folding process is unclear.
The aim of this proposal is to investigate the role of molten globule state in the refolding mechanism of individual titin domains. To reach this objective, in addition to manipulating whole titin molecules, I plan to develop a double stranded DNA handle based method to manipulate various individual globular domains from titin with high resolution force-feedback optical tweezers. I anticipate that this will allow identifying the exact states involved in the refolding process.

What is the major research question?

The aim of this project is to get a better understanding of the mechanism of titin folding.
Based on our previous results we hypothesize that refolding of titin domains occurs through an intermediate, most likely a molten globule state. This hypothesis can only be tested on single titin domains, because the large number of domains present in the full length molecule results in the ensemble behavior of titin folding, which restricts the characterization of individual states in the folding process. I plan to investigate the folding of an immunoglobulin and a fibronectin titin domain and also the chimera construct of the two domains. The individual domain constructs are simple experimental models of titin domains, while the domain chimera is a simple model on the collective folding of titin domains. Questions to be answered by experiments:
1. What structural states are involved in the folding process of mechanically denatured titin domains?
2. What is the transition dynamics between the states?
3. How are individual domain folding trajectories involved in the ensemble folding od full length titin?

Although it is known for years that denatured titin molecules refold when relaxed at contracted conformations, the exact mechanism of the folding process remain unknown. So far the force-drivein unfolding and refolding processes in titin have been successfully explained and modeled with a two-state model. Our preliminary experiments on native titin suggests that an intermediate, most likely a molten globule state is also involved in the folding process. With this research project we hope to get a better insight into the folding process of titin's globular domains, and to have a better understanding on the role of molten globule state in the folding of titin. Because the transition from the unfolded to the molten-globule state shortens the chain faster than a purely entropic collapse, a novel molecular contraction mechanism may arise, based on entropic elasticity and molten-globule dynamics. Such contraction mechanism may be especially relevant in case of cardiac pathologies involving truncated titin mutants, which leads to sarcomeric stressed conditions when titin's domains become unfolded.

Titin, the largest human polypeptide is responsible for the passive elastic properties of skeletal and cardiac muscle. It spans the structural units of muscle fibers as a molecular spring element. Due to its elasticity it is capable of extension and contraction during muscle movement. When stretch, first the molecule extends and above a certain force threshold its structural units, the so called domains loose their integrity and unfold. When this unfolded molecule is relaxed, its domains recover their structure, they refold. The exact mechanisms of refolding still remains unknown. Based on our preliminary experiments, full length titin stretched with optical tweezers are folded through intermediate, so called molten globule states. To test this hypothesis, we plan to manipulate individual domain units of titin with optical tweezers, in order to reveal the states involved in the folding process. Our results will provide a better understanding of the folding process of titin, thus a better understanding of alterations in tissue biomechanics in case of some cardiac pathologies.

A kutatás során a harántcsíkolt izom rugalmasságát kialakító titin molekula szerkezetváltozásait vizsgáltuk lézercsipesszel, különös tekintettel a kitekert szerkezetből történő gombolyodás folyamatára. Kísérleteinkben kimutattuk, hogy az előzetesen kitekert titinmolekula a gombolyodása kb. 3 pN nagyságú erőkifejtésre képes másodperces időskálán. Az erőkifejtés a feltekeredő domének különböző szerkezeti állapotok közötti dinamikus átmentéivel magyarázható́. Mivel a domén kitekeredés valószínűsége elenyésző ebben az erőtartományban, a fluktuációk arra utalnak, hogy a domének feltekeredése a natív szerkezetbe a kitekertből egy kompaktabb, molten-globule állapoton keresztül történik, aminek révén a molekula erőkifejtésre képes. Összességében kimutattuk, hogy a titinmolekula gombolyodásának szerves része egy intermedier molten-globule állapot, ami szerkezetét tekintve még nem koszolidált mint a natív, de hossz tekintetében közel ugyanakkora. A molten-globule állapot lehetővé teszi a kitekert fehérje lánc gyors rövidülését egy a natív állapot közeli hosszra, amiből láncfluktuációkon keresztül történik meg az egyes domének gombolyodása. Ez az extra kontrakciós képesség fontos szereppel bírhat az izomrostok rövidülési mechanizmusában.
Results in English
In this research project, we have investigated the structural changes of titin, the elastic filament of muscle sarcomeres. Particularly we explored the role of structural intermediates during the folding process of titin. In our findings, we have demonstrated that when the unfolded molecule is allowed to relax it is able to generate a force of about 3 pN on the second time scale during its folding. The force generation can be explained by the dynamic structural transitions of titin domains. Whereas net force generation is caused by the ensemble folding of the elastically-coupled domains, force fluctuations arise due to a dynamic equilibrium between unfolded and molten-globule states. In sum, we have demonstrated that the molten globule intermediate is an essential part of titin’s folding process. The structure of this intermediate is not as consolidated as the native, but its length is nearly the same. The ensemble molten-globule dynamics delivers significantly added contractility that may assist sarcomere mechanics, and it may reduce the dissipative energy loss associated with titin unfolding/refolding during muscle contraction/relaxation cycles.
