A kutatás összefoglalója, célkitűzései szakemberek számára
Itt írja le a kutatás fő célkitűzéseit a témában jártas szakember számára.

Eddigi kutatásaink során a Rhodobacter (Rb.) sphaeroides bíborbaktériumból tisztított reakciócentrumot (RC) szén (szén nanocsövek (CNT) és CNT kötegek) és szilícium (porózus szilícium) alapú hordozók, valamint átmeneti fém oxidok (ITO) felületén kémiai és fizikai kötések által rögzítettük, különféle kötési technikák alkalmazásával. Az így létrejött minták szerkezetét és aktivitását többek között mikroszkópos technikákkal (AFM, EM) és flash fotolízises mérések segítségével jellemeztük.
A legfontosabb kérdések bio-hibrid anyagok előállítása során: 1) melyik a legjobb rendszer a hatékony energia átalakításhoz; 2) mennyire hatékony az energia átalakítás; 3) mennyire stabil a rendszerünk? Szén alapú anyagokat használunk (egyfalú és többfalú, funkcionalizált (karboxil- és amin-funkcionált) és nem-funkcionalizált szén nanocsöveket, szén nanocső kötegeket és grafén lemezeket). A RC-ot elsősorban Rb. sphaeroides bíborbaktériumból fogjuk izolálni, viszont adott esetben más fajok is hozzáférhetőek. A RC-ot az alapvető kötési technikák alkalmazásával fogjuk rögzíteni különböző orientációban (specifikus és nem specifikus) és vizsgálni fogjuk az orientáció struktúrára és aktivitásra kifejtett hatását.
A partner laboratóriumainkban elérhetőek számunkra különböző mikroszkópos technikák (AFM, EM, esetlegesen SRLM), amelyek segítségével elvégezhetjük a kompozitok strukturális jellemzését. A megkötött RC fotokémiai/fizikai aktivitását a kofaktorok redox állapotainak időállandójával jellemezhetjük, a specifikus hullámhosszokon. A minták alapvető elektronikai tulajdonságait pl. fényindukált vezetőképesség méréssel határozhatjuk meg, lépéseket tettünk spektroelektrokémiai mérésekre is.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Számos példa van arra, hogy a biológiai anyagok (a fehérjék is) szervetlen hordozón történő rögzítésüket követően is megőrzik aktivitásukat. A fotoszintetikus reakció centrum (RC) néhány olyan előnyös tulajdonsággal rendelkezik, ami gyakorlati hasznosíthatóság lehetőségét is felveti. A másik általunk használt igen ígéretes anyag a szén nanocső, kívételes mechanikai és vezetési tulajdonságokkal. (Említést érdemel, hogy mindkét anyag felfedezéséhez illetve jellemzéséhez Nobel díj fűződik.) Nagy kihívást jelent tehát olyan újgenerációs fotoaktív bio-nanokompozit létrehozása, amely ezeket a figyelemreméltó anyagokat társíthatja.
Eddigi laboratóriumi munkánk során képesek voltunk a RC-ot eltérő szénalapú anyagokon rögzíteni, különböző technikák segítségével. Lehetséges volt újtípusú hibrid anyagok létrhozása és jelelmzése, valamint esetleges gyakorlati alkalmazások is kilátásba kerültek. Két fő kérdés merült fel a kísérletek során. Egyrészt, hogyan változik a fényindukált elektrontranszport a RC fehérje komplexen belül, annak rögzítését követően. Másrészt, pontosan hogyan megy végbe az elektrontranszport a fehérje és a hordozó közötti felületen. (Korábbi eredményeinkkel már bizonyítottuk, hogy van redox kapcsolat a két anyag között.)
A kompozitok fotokémiai/fizikai tulajdonságainak leírásához elengedhetetlen ennek a két kérdésnek a megválaszolása. Másrészről, a kérdések megválaszolásához meg kell határozni a kompozit optimális működéséhez szükséges feltételeket.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

A természet által kifejlesztett biológiai anyagok igen specifikusak, érzékenyek, működésük nagyon hatékony és összetett. Felhasználásuk bio-(nano)hibrid anyagok létrehozásához mindig is kihívást jelentett a tudomány számára, hiszen nem csak a meglévő igen előnyös tulajdonságaikat hasznosíthatjuk, de akár új előnyös tulajdonságok is megjelenhetnek. Ily módon lehetőség nyílik környezetbarát, biológiailag lebontható anyagok létrehozására, pl. a bioszenzor vagy gyógyszeripari alkalmazás területén. A fotoaktív biológiai anyagok fontos felhasználási lehetősége nyílik pl. fotoelektronikus alkalmazásokban, energia átalakító rendszerekben , mint a (bio)szenzorok, fotovoltaikus cellák, integrált optikai berendezések, különféle képalkotó rendszerek, stb. Természetes környezetükön kívül a biológiai anyagok igen sérülékenyek és érzékenyek. Néhány esetben (mint az általunk korábban bemutatt rendszerek esetében) a létrejött mesterséges környezet képes az in vivo környezet valamilyen mértékű helyettesítésére, és hasonló stabilitást biztosítani.
Ezeknek a kompozitoknak számtalan hasznosítási lehetősége képzelhető el. A fő cél a fehérjén belüli elektron és proton transzport illetve energia hasznosítás kvalitatív és kvantitativ jellemzése lehet a a rögzítést követően. Ezenkívül további célunk a hordozó és a biomolekula közötti határfelület szerepének tisztázása is. A kutatás egyetemi tanszéken zajlik, tehát lehetőség nyílik hallgatók bevonására.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

Korábbi méréseink során már sikerrel kötöttük a reakciócentrumot szén nanocsövekhez. Az ilyen típusú anyagokat bio-nanokompozitoknak nevezzük, amelyeket a különleges tulajdonságaik miatt a „jövő anyagainak” is szoktak nevezni. Méréseink során már bizonyítottuk, hogy a fehérje a nano-rendszerekben nagy mértékben megőrzi fotoaktivitását és létrejöhet redox kapcsolat a kétféle anyag között. A projekt célja tökéletesíteni a kompozit felépítését,javítani annak stabilitását és fotoelektromos energiaátalakítási tulajdonságait.
Különböző típusú reakciócentrumot (karotinoidot tartalmazó és karotinoid mentes) kötünk különféle (egyfalú és többfalú, funkcionalizált és nem funkcionalizált) szén nanocsőhöz.
A környezeti hatások (hőmérséklet, páratartalom, oxidáló környezet) nagyban befolyásolhatják a kompozitok stabilitását, ezért ezeknek a tényezőket a hatását is jellemezni fogjuk. A szükséges tapasztalatokkal és laboratóriumi eszközökkel is rendelkezünk a minták előállításához. Bár a minták fotoaktivitásának alapvető jellemzéséhez az eszközeink rendelkezésre állnak, mivel a mintákat magukat is folyamatosan fejlesztjük, a mérési módszereket is állandóan tökéletesíteni szükséges.
A tervezett mérések új alapkutatási eredményhez vezethetnek, a fehérjén belüli és a két anyag között végbemenő töltésstabilizálódási folyamtokról, jövőbeli újgenerációs optoelektronikai alkalmazások lehetőségét vetítheti elő.

Previously photosynthetic reaction center protein (RC) purified from the purple bacterium Rhodobacter (Rb.) sphaeroides was bound to several carbon (different carbon nanotubes (CNTs), CNT bundles) and silicon (porous silicon) based materials and to transitional metal oxides (ITO) chemically and physically, by various binding procedures. The basic structural and functional characterization was also performed by standard (e.g. AFM, EM, flash photolysis) methods.
The main questions when bio-hybrid materials are investigated are: 1) what is the best system for the efficient energy conversion; 2) how efficient is the energy conversion; 3) what is the stability of the system? Here we are focusing on carbon based materials (single and multi walled, functionalized (carboxyl- and amine-functionalized) and non-functionalized CNTs, CNT bundles and graphene sheets). RCs will be prepared from Rb. sphaeroides, however, depending on the conditions other species are also available. By using the basic immobilization techniques, we will immobilize the RCs with different (specific and non-specific) orientations and the orientation effect on the structure and function will be investigated.
Different microscopy techniques are available in the partner laboratories where the structural characterization can be done (AFM, EM, perhaps SRLM). The photochemical/-physical activity of the immobilized RC can be well characterized by the life times of the redox states of the cofactors at specific wavelengths. The basic electric properties (e.g. light induced conductivity) of the samples can be measured, and we made a step forward to do spectroelectrochemical characterizations as well.

What is the major research question?
Describe here briefly the problem to be solved by the research, the starting hypothesis, and the questions addressed by the experiments.

There are many examples that biological materials (proteins as well) can be bound to inorganic carrier matrices largely retaining their activity. Photosynthetic reaction center proteins (RC) are very promising because these have many advantageous properties, which can be useful for practical applications. The other very promising materials are carbon nanotubes because of their exceptional mechanical and electrical properties. (It is worth to mention that the research connected to both materials was awarded by Nobel prize.) The combination of these interesting substances to a new type of bio-nanocomposite material is extremely challenging and the new type of characteristics can be envisaged.
Based on our earlier experiences in our laboratory we are able to bind RC to different carbon materials by various methods. This procedure results in an absolutely new type of hybrid material with new characteristics with potential practical application. There are two main questions. The main question is how the light induced electron transport within the RC is modified after the binding to the inorganic carrier? The other question is, what is the electron transport mechanism in the interface between the RC and CNT. (We already have shown that there is redox transient between the two components.)
It is essential to answer these two questions if one would like to describe the photochemical/-physical activity of the composites. On the other hand, in order to answer these two questions the optimal structure of the materials and the environmental conditions should be worked out.

What is the significance of the research?

Biological materials are developed by nature for extremely specific, sensitive and efficient functions with their complexity. It is a real challenge to use them in bio-(nano)hybrid composites and harness all their advantageous properties, in addition new type of characteristics can appear. This way it is possible to build environmentally friendly biodegradable items like drug delivery systems, biosensors. In addition, it is a challenge to use the photoelectric energy conversion phenomenon in special (opto)electronic devices, e.g. in (bio)sensing elements, photovoltaic cells, integrated optical devices, different imaging systems, etc. Biomaterials are sensitive when they are isolated out of their natural environment, so these applications usually require their integration into biocompatible abiotic matrices. In some cases (like in our system, as we already have shown it) this artificial environment can mimic the in vivo conditions providing a relative stability of the system.
The expected benefits and utilization can be envisaged in several aspects. The fundamental aspect is better understanding and quantification of coupling of the electron and proton transport, energy utilization processes within the protein (the biological component) after attaching it to carrier matrices. In addition, results will be gained about the role of the interface between the two types of the materials in the hybrid system. Besides this effect there are „technological”, „applied research” and „educational” benefits as well.

Summary and aims of the research for the public
Describe here the major aims of the research for an audience with average background information. This summary is especially important for NRDI Office in order to inform decision-makers, media, and others.

In our earlier experiments we already bound successfully photosynthetic reaction centers to carbon nanotubes. These type of hybrid materials are called bio-nanocomposites, which are also termed as the materials for future. We have already shown, as pilot experiments, that the protein retains its photoactivity and there is a redox interaction between the two parts of the complex. The aim of the project is to work out a better combination of the composite material for improved photoelectric energy conversion.
For this reason we will bind different type of reaction centers (carotenoid containing and non-containing) to different carbon nanotubes (single walled and multiwalled, functionalized and non-functionalized). The effect of the environment (temperature, humidity, oxygenic/non-oxygenic) on the long term stability will be investigated. We have experience and fully equipped laboratory facilities for the sample preparation. Although we also have basic equipments and instrumentation for characterizing the photoactivity of the sample the measurements require continuous development of the system. There are only few and expensive (if any) commercial instruments for measuring e.g. action spectra, quantum yield of the photochemical/-physical reactions.
Our results will be a better understanding of the nature of the electron transfer reaction in the interface between two components, which can be a basis of the design of new generation optoelectronic devices.

A projekt elsődleges célja fotoaktív bionankompozitok előállítása volt, elsősorban szénalapú vezető és félvezető anyagok, valamint bakteriális fotoszintetikus reakciócentrum felhasználásával. Az így létrehozott hibrid kompozitokat munkaelektródként alkalmaztunk egy elektrokémiai cellában, ahol az elnyelt fény hatására fotoáram keletkezett. Redox-aktív anyagok vagy herbicid hozzáadását követően a fotoáram szenzitív változást mutatott, így az elektródok bioszenzorként működtek. A biológiai komponens érzékenysége miatt fontosnak tartottunk minél stabilabb biokompozitok létrehozását és ennek érdekében számos kiegészítő mérést elvégeztünk. Nagy hangsúlyt fektettünk a kompozitokban fénygerjesztést követően keletkező szinglet oxigén mennyiségi meghatározására, illetve a szén nanocsövek ebben betöltött szerepének definiálására. Ezenkívül, izotópok mérésére alkalmas analitikai eszközökkel sikeresen meghatároztunk a szén nanocsövekhez kötött reakciócentrum mennyiségét, így pontosabb értéket kaphatunk a reakciócentrum immobilizálásának hatékonyságáról a hordozó felületen.
Results in English
The main concept of the project was the preparation of photoactive bionanocomposites based on two components: one was a conductive/semiconductive, principally carbon based material, the other was bacterial photosynthetic reaction center. The absorbed light induced photocurrent in the created hybrid electrodes where – depending on the occuring current - sensitive responses were given to any added redox active or herbicide molecules. Thus, these electrodes functioned as biosensors. In order to extend the possible life time of the biocomposites, several additional measurements were done regarding their working efficiency. Great emphasis was put on the light-induced singlet oxygen production inside the protein complex and also, on the carbon nanotubes as active participants in the ROS-related chemical and physical reactions. Also, isotope analytics was applied as a useful tool for determining the quantitative binding of the photosyntethic reaction center to the carbon-based carriers.
