A transzláció befejező lépése a termináció. Ilyenkor az eukarióta Releasing Factor 1 (eRF1) terminációs faktor az eRF3-mal komplexet képezve kapcsolódik a stop kodonhoz, ami a fehérje felszabadulását és a riboszóma szétszerelődését eredményezi. Amennyiben az mRNS-en speciális szignálok vannak és/vagy a terminációs faktorok szintje alacsony, normál termináció helyett alternatív események, mint readthrough (RT) vagy Nonsense-mediated mRNA Decay (NMD) következik be.
A program célja a növényi termináció szabályozásának megismerése. Feltételezésünk szerint ennek legfontosabb eleme az eRF1 szint szabályozása, ugyanis növényekben, hasonlóan az élesztőhöz, mind a megemelkedett, mind a lecsökkent eRF1 szint fenotípust eredményez. Előzetes eredményeinkre alapozva kidolgoztunk egy modellt, mely szerint növényekben egy speciális autoregulációs mechanizmus felel az eRF1 fehérje szintjének szabályozásáért. A program során különböző riporter vonalak létrehozásával, elemzésével igazolni akarjuk a feltételezett eRF1 autoregulációs rendszer működését, a különböző eRF1 mutánsok összehasonlításával tisztázni kívánjuk az eRF1 regulációs ciklus biológia szerepét, és a zöldalga, Lycophyta, nyitva- és zárvatermő eRF1-ek szabályozások összevetésével meg akarjuk érteni a rendszer evolúcióját. Azonosítani szeretnénk a növényi terminációs komplex másik elemét, az eRF3-at, és fel kívánjuk tárni az eRF3 szabályozásának alapjait is.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Ha a riboszómóma egy stop kodonhoz ér a transzláció terminálódik. Bizonyos esetekben a termináció helyett alternatív események következnek be. Ilyen stop kodonhoz kapcsolódó alternatív esemény a frameshift, a readthrough vagy a Nonsense-mediated decay (NMD) is. Mivel eukariótákban a termináció, a readthrough és az NMD is létfontosságúak, feltételezzük, hogy ezek a folyamatok koordináltan szabályozottak. Az ismert, hogy az eRF1 terminációs faktor koncentrációja döntően befolyásolja a termináció, a readthrough, és a frameshift gyakoriságát is. Mindezek alapján feltételezzük, hogy az eRF1 fehérje szintje szigorúan szabályozott, és azt is, hogy az eRF1 szint alapvető szerepet játszik az alternatív terminációs események közti egyensúly biztosításában. Az eRF1 szabályozásról alig valamit tudunk. Az eRF1 szabályozás különösen bonyolult lehet növényekben, mivel növényekben -szemben a legtöbb eukariótával- az eRF1 mindig több kópiás.
Előzetes eredményeink alapján valószínűsítjük, hogy egy komplex és specifikus szabályozási folyamat stabilizálja az eRF1 szintet növényekben. Feltételezzük, hogy a szabályozott elem az eRF1-1 mRNS egyedülálló szerkezete. Feltételezzük azt is, hogy az eRF1 szint regulációjában mind a readthrough, mind az NMD (egy a transzláció terminációjához kapcsolódó minőségbiztosítási rendszer) alapvető szerepet játszik. Változatos tranziens és transzgénikus növény kísérletek révén kívánjuk feltárni, hogy hogyan szabályozódik a növényi eRF1, mi a biológiai jelentősége ennek a szabályozásnak, illetve tisztázni kívánjuk az eRF1 reguláció evolúcióját is. Munkánk során meg szeretnénk ismerni a másik alapvető terminációs faktor, a növényi eRF3 szabályozását is

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. A növényi transzláció terminációjáról nagyon keveset tudunk. Bár a legfontosabb terminációs faktorokat, az eRF1-eket leírták, a növényi terminációs komplex eRF3 komponensei eddig ismeretlenek. A növényi termináció mechanizmusáról, szabályozásáról semmit sem tudunk.
A program siker esetén alapvetően járulna hozzá a növényi génexpresszió egyik alapvető lépésének, a transzláció terminációs lépésének a megismeréséhez. Eredményeink révén jobban megérthetjük a növényi termináció működését, szabályozását, illetve transzláció terminációjának evolúcióját. Számos növényi vírus replikációjához nélkülözhetetlen a normál termináció átolvasáson, leolvasási keret váltáson (readthrough, frameshift) alapuló elkerülése. Ezen alternatív eseményeknek a gyakorisága jelentős részben az eRF1 terminációs faktor koncentrációján múlik, így a program központi elemének a sikere, az eRF1 szint szabályozásának megértése, új antivirális stratégiák kidolgozását eredményezheti.

A kutatás összefoglalója, célkitűzései laikusok számára

A DNS-ben kódolt genetikai információt az mRNS-ek továbbítják a sejt alapvető működéseiért felelős fehérjék felé. A DNS-mRNS információ átadást transzkripciónak, a mRNS-ről a fehérjére történő információ továbbítást transzlációnak nevezzük. A transzláció három fő lépése az iniciáció, az elongáció és a termináció. Korábban azt gondolták, hogy a transzláció szabályozása a legbonyolultabb iniciációs lépésnél történik. Az elmúlt évek eredményei alapján azonban világos, hogy (az elongáció és) a termináció is hatékony szabályozás alatt áll. A termináció szabályozásának a kulcsa, a transzláció befejezésében döntő szerepet játszó fehérje, az eRF1 terminációs faktor mennyiségének szigorú szabályozása. Azt régen tudjuk, hogy baktériumokban a terminációs faktor szintjét egy bonyolult önszabályozás tartja stabilan, de arról, hogy az eukarióták miként szabályozzák az eRF1 terminációs faktor szintjét szinte semmit sem tudunk.
Programunk során fel kívánjuk tárni, hogy hogyan regulálják a növények a transzláció befejező, terminációs lépését. Korábbi eredményeink alapján feltételezzük, hogy növényekben egy nagyon speciális és bonyolult szabályozási kör regulálja az eRF1 terminációs faktor mennyiségét. A program célja ennek a szabályozási körnek a megismerése, kísérletes elemzése. Mivel a növényi vírusok jelentős részének szaporodása a termináció részleges kijátszásán alapul, a termináció szabályozásának megismerése -a növényi génszabályozás jobb megértése mellett - új, hatékony antivirális rendszerek kialakítását is elősegítheti.

Termination is the final step of translation. Binding of the eukaryotic Release Factor 1 (eRF1) in a complex with the eRF3 protein to the stop codon stimulates the peptide release and the disassembling of the ribosome. Infrequently, if a transcript contains a specific signal and/or conditions do not favor efficient termination, alternative events such as translational readthrough (RT) or Nonsense-mediated decay (NMD) can occur at the stop codon.
The aim of this program is to unravel the regulation of translation termination in plants. As in plants, like in yeast, both over- and underexpression of eRF1 lead to altered phenotype, we hypothesize that the key element of termination regulation is the strict control of eRF1 level. Based on our preliminary results, we elaborated a model, in which we propose that eRF1 expression is stabilized by a complex and specific autoregulatory circuit in plants. During this program, various reporter lines will be created and analyzed to test the predictions of our autoregulatory model. Different eRF1 mutants will be studied to clarify the biological function of the autoregulatory system. We try to comparatively analyze the eRF1 regulation in algae, lycophytes, gymnosperms and angiosperms to understand the evolution of the eRF1 autoregulation. Finally, we try to identify the eRF3 component of the plant termination complex and study the regulation of eRF3 expression.

What is the major research question?

When a translating ribosome reaches a stop codon, different events can occur: normal termination, frameshift, translational readthrough or Nonsense-mediated mRNA decay (NMD). As termination, readthrough and NMD are all required for viability in certain eukaryotes, we assume that these stop codon related events are coordinately regulated. The frequency of readthrough, frameshift and normal termination strongly depends on the concentration of eRF1 termination factor. Therefore we hypothesized that the eRF1 protein level plays a critical role in keeping the balance between these alternative stop codon related events and that eRF1 protein level is strictly controlled in eukaryotes. Little is known about the regulation of eRF1 expression in eukaryotes. In addition, as in most eukaryotes eRF1 is a single copy gene, while in higher plants eRF1 is always present in multiple copies, we postulate that the regulation of eRF1 might be especially complex in plants.
Our preliminary data suggest that in plants a very special and highly complex autoregulatory circuit controls the expression of eRF1. We hypothesize that the unique structure of the eRF1-1 mRNA is the regulated component and that both translational readthrough and NMD, which is a translation termination coupled quality control system, play an important role in the control of eRF1. Using various transient expression and transgenic approaches, we try to clarify how eRF1 level is regulated in plants, what is the biological relevance of this regulation and that how this control system has evolved. We also want to identify and describe the regulation of eRF3 termination factor.

Little is known about translation termination in plants. Although the most important termination factors, the eRF1 proteins have been described, the eRF3 components of the plant termination complexes have not been identified yet. Nothing is known about the mechanism and the regulation of plant translation termination.
The success of this project could significantly contribute to our understanding of the translation termination step of plant gene expression. Our results could substantially improve our knowledge about the mechanism, regulation and evolution of translation termination of plants.
Bypassing normal termination is essential for the replication of several economically important plant viruses. The frequency of these alternative events such as translational frameshift and readthrough strongly depends on the concentration of eRF1 termination factor. Thus unraveling the regulation of plant eRF1 might be used to elaborate novel, efficient antiviral strategies.

mRNAs transfer the genetic information from the DNA towards proteins, which are responsible to carry out the most functions in the cell. The generation of mRNA from DNA is called transcription, while the process in which the information of mRNAs is transformed into proteins called translation. Translation has three main steps, initiation, elongation and termination. Previously it was believed that the initiation is the only regulated step of translation. However, recent results have shown that elongation and termination are also accurately controlled. The most important element of the termination regulation is the strict control of the expression of the key termination protein called eRF1 (Release Factor). Although it is known for while that in bacteria the Release Factor level is stabilized by a complex autoregulatory mechanism, it is still not understood how eukaryotes regulate the expression of eRF1.
During this program we want to unravel how the termination step of translation is regulated in plants. Based on our previous results we hypothesize that in plants a special and very complex regulatory circuit controls the level of eRF1 protein. During this program we try to experimentally analyze the mechanism and function of this regulatory circuit. As the replication of several plant viruses is based on the bypassing of normal termination, the success of this project –in addition to that it significantly contribute to our understanding of gene regulation of plant- might be useful to elaborate novel and efficient antiviral strategies.
