A Mycoplasma synoviae fertőzés nagy gazdasági veszteségekkel járó légzőszervi megbetegedést, ízületgyulladást és tojásvég elváltozást okoz a tyúk és pulyka állományokban. Világszerte elterjedt kórokozó, mely egyre gyakrabban okoz megbetegedéseket. A M. synoviae fertőzés elleni védekezésnek három lehetséges módja van; a mentesítést követő megelőzés, a vakcinázás és a gyógykezelés.
A pályázat célja a M. synoviae fertőzés elleni védekezés lehetőségeinek a fejlesztése:
1) Megbízható genotipizáló rendszert fejlesztünk ki, ami lehetővé teszi, hogy feltárjuk az egyes M. synoviae járványok forrását és terjedésének módját az egyes baromfi integrációk között és integrációkon belül, ezzel is növelve a megelőzés hatékonyságát.
2) Molekuláris rendszert fejlesztünk, ami lehetővé teszi a vakcina törzs és a klinikai izolátumok gyors elkülönítését.
3) Aktuális adatokat szolgáltatunk a M. synoviae törzsek antibiotikum érzékenységéről, amik nagyban segíteni fogják a fertőzések hatékony gyógykezelését. Gyors és olcsó molekuláris rendszereket fejlesztünk ki a M. synoviae törzsek egyes antibiotikumokkal szembeni érzékenységi profiljának a meghatározására. Ennek segítségével izolálás nélkül, közvetlenül a klinikai mintából (pl. légcső tampon), néhány órán belül meg lehet határozni a fertőzést okozó törzs antibiotikum érzékenységét. Így a gyakorló állatorvosok gyorsan célzott antibiotikum kezelést tudnak alkalmazni a fertőzött állományon olyan szerrel, aminek a legjobb hatékonyság mellett legkisebb a közegészségügyi értéke, megfelelő az élelmiszer egészségügyi várakozási ideje és a legkedvezőbb az ára is. Mindezek hatására a gazdasági veszteségek nagymértékben csökkenthetőek lesznek.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Milyen megbízható genotipizáló rendszert lehet kifejleszteni molekuláris epidemiológiai vizsgálatokhoz? Az élő, attenuált vakcina törzset el lehet-e különíteni molekuláris biológiai módszerrel a vad M. synoviae izolátumoktól? Milyen antibiotikum érzékenységi profillal rendelkeznek a hazai és közép-kelet európai M. synoviae törzsek? Milyen gének felelősek az egyes antibiotikumokkal szembeni rezisztencia kialakításában? Van-e lehetőség gyors, érzékeny és költséghatékony molekuláris biológiai módszereket kifejleszteni a M. synoviae törzsek antibiotikum érzékenységének meghatározására?

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Egy hazai és közép-kelet európai M. synoviae törzsgyűjteményt hozunk létre. Egy megbízható genotipizáló rendszert fejlesztünk ki, ami lehetővé teszi majd, hogy molekuláris epidemiológiai vizsgálatokat végezzünk, s ezáltal feltárjuk az egyes járványok forrását és terjedésének módját egy-egy baromfi integráción belül és az egyes integrációk között. Molekuláris rendszert fejlesztünk, ami lehetővé teszi a vakcina törzs és a vad törzsek gyors elkülönítését. Ezek a rendszerek növelni fogják a M. synoviae mentesítési programok hatékonyságát. Aktuális adatokat szolgáltatunk a magyarországi és közép-kelet európai M. synoviae izolátumok antibiotikum érzékenységéről, amik nagyban segíteni fogják a hazai fertőzések hatékony gyógykezelését. Meghatározzuk számos vad izolátumnak és in vitro szelektált rezisztens törzsnek a teljes genom szekvenciáját és elemezzük azt. Gyors és olcsó molekuláris rendszereket fejlesztünk ki a M. synoviae törzsek egyes antibiotikumokkal szembeni érzékenységi profiljának a meghatározására. Ennek segítségével izolálás nélkül, közvetlenül a klinikai mintából (pl. légcső tampon), néhány órán belül meg lehet határozni a fertőzést okozó M. synoviae törzs antibiotikum érzékenységét. Így a gyakorló állatorvosok gyorsan, célzott antibiotikum kezelést tudnak alkalmazni a fertőzött állományon olyan szerrel, aminek a legjobb hatékonyság mellett legkisebb a humán egészségügyi értéke, megfelelő az élelmiszer egészségügyi várakozási ideje és a legkedvezőbb az ára is. Mindezek hatására a gazdasági veszteségek nagymértékben csökkenthetőek lesznek.
A pályázott kutatás legjelentősebb eredménye az antibiotikum rezisztencia genetikai hátterének a feltárása lesz, illetve ezen genetikai markerek alapján kifejlesztett molekuláris biológiai módszerek az antibiotikum rezisztencia gyors kimutatására. Számításaink szerint ez egy nagy nemzetközi visszhangot kiváltó tudományos eredmény lesz. Továbbá ez lesz az első kutatás, ami aktuális adatokat fog szolgáltatni a közép-kelet európai M. synoviae törzsek antibiotikum érzékenységi profiljáról. A vakcina törzset elkülönítő és a járványtani nyomozásra kifejlesztett molekuláris biológiai rendszerek szintén világ szintű érdeklődésre tartanak majd számot.

A M. synoviae világszerte előforduló, egyre jelentősebb, baromfi állományokban nagy gazdasági károkkal járó megbetegedéseket okozó ágens. A pályázat célja a M. synoviae fertőzés elleni védekezés lehetőségeinek a fejlesztése:
1) Olyan módszert fejlesztünk ki, ami lehetővé teszi, hogy feltárjuk az egyes M. synoviae járványok forrását és terjedésének módját az egyes baromfi tenyésztő cégek között és cégeken belül, ezzel is növelve a megelőzés hatékonyságát.
2) Olyan eljárást dolgozunk ki, ami lehetővé teszi a vakcina törzs és a klinikai izolátumok gyors elkülönítését.
3) Aktuális adatokat szolgáltatunk a M. synoviae izolátumok antibiotikum érzékenységéről, amik nagyban segíteni fogják a fertőzések hatékony gyógykezelését. Gyors és olcsó módszereket fejlesztünk ki a M. synoviae törzsek egyes antibiotikumokkal szembeni érzékenységi profiljának a meghatározására. Mycoplasma synoviae is an economically important pathogen of poultry causing respiratory disease, infectious synovitis and eggshell apex abnormality in chickens and turkeys. This agent has a worldwide distribution and its occurrence is increasing. Control of this disease consists of three general aspects: eradication followed by prevention, vaccination or medication.
The main aim of the present project is to improve the control of M. synoviae infection:
1) A system for the genotyping of M. synoviae isolates will be developed. It will facilitate the discovery of possible M. synoviae reservoirs and infection routes between and within the different poultry farming companies, thus it will improve the disease prevention efforts.
2) An assay to differentiate the live attenuated vaccine strain from the clinical isolates will also be developed.
3) Essential data will be presented about the antibiotic susceptibility profiles of M. synoviae strains which will help the antibiotic therapy of infections. Methods allowing rapid and cost-effective molecular detection of the antibiotic susceptibility profile of M. synoviae isolates will be developed. This will allow us to determine the antibiotic susceptibility of the clinical isolates within few hours, without culture, directly from the clinical samples (e.g. trachea swabs). Veterinary practitioners will be able to rapidly start treating M. synoviae infected flocks and choose the best antibiotic which is the most effective, less important for human medicine, has appropriate withdrawal time and lowest price. Consequently the economic losses and spreading of antibiotic resistance will be markedly reduced.

Is it possible to differentiate the live attenuated vaccine strain from wild virulent M. synoviae strains by molecular methods? What are the antibiotic susceptibility profiles of the Hungarian and Central and Eastern European M. synoviae strains? What are the antibiotic resistance coding regions in the genome? Is it possible to develop rapid, sensitive and cost-effective molecular methods for determining antibiotic susceptibility of M. synoviae isolates?

What is the significance of the research?

A Hungarian and a Central and Eastern European M. synoviae strain collection will be established. A reliable and robust system for the genotyping of M. synoviae clinical isolates will be developed. This system will be used for molecular epidemiology purposes and will facilitate the discovery of possible M. synoviae reservoirs and infection routes between and within the different poultry integration companies. An assay to differentiate the live attenuated vaccine strain from the clinical isolates will also be developed. These methods will facilitate M. synoviae eradication programs. Essential data will be presented about the antibiotic susceptibility profiles of the Hungarian and other Central and Eastern European strains which will help the antibiotic therapy of M. synoviae infections. The genome of clinical M. synoviae isolates and in vitro selected antibiotic resistant clones will be sequenced and analyzed. Methods allowing rapid and cost-effective molecular detection of the antibiotic susceptibility profile of M. synoviae isolates will be developed. This will allow us to determine the antibiotic susceptibility of the clinical isolates within a few hours, without culture, directly from the clinical samples (e.g. trachea swabs). The veterinary practitioners will be able to rapidly start treating M. synoviae infected flocks and choose the best antibiotic which is the most effective, less important for human medicine, has appropriate withdrawal time and lowest price. Consequently the economic losses will be markedly reduced.
The most significant scientific result of this study will be the discovery of genetic markers of antibiotic resistance and the development of rapid molecular methods targeting the resistance mutations. It will be an internationally highly recognized result. Furthermore this study will be the first which provides data about the antibiotic susceptibility profile of the M. synoviae isolates of Central and Eastern Europe. The differentiation of the live attenuated vaccine strain from other isolates and the development of genotyping systems for molecular epidemiology purposes will also have a world-wide significance.

A pályázatban vállaltakat maradéktalanul elvégeztük, továbbá számos helyen, a pályázat témájához szorosan kapcsolódó vizsgálatokkal is kiegészítettük: - Felállítottunk egy hazai és egy nemzetközi törzsgyűjteményt. - Elsőként fejlesztettünk DIVA (vakcina-vad elkülönítő) rendszert az MS1 vakcinára. - Új DIVA rendszert fejlesztettünk az MS-H vakcinára. - Elsőként fejlesztettünk DIVA rendszereket a ts11, 6/85, F és K M. gallisepticum vakcinákra. - Egy multi-locus variable-number tandem repeat analysis (MLVA) fejlesztettünk a M. synoviae törzsek genotipizálására és elvégeztük a törzsek multi-locus sequence typing (MLST) vizsgálatát is. - Elsőként dolgoztunk ki egy MLST rendszert a M. gallisepticum törzsekre. - Meghatároztuk a hazai és egyéb Közép-Kelet európai M. synoviae törzsek antibiotikum érzékenységi profilját. - Törzseket gyűjtöttünk Ázsiából és meghatároztuk törzsek antibiotikum érzékenységi profilját. - Feltérképezzük a M. synoviae egyes antibiotikummal szembeni rezisztencia kialakulásáért felelős génjeit és mutációit. - Az azonosított genetikai markerek kimutatására gyors és költség-hatékony molekuláris biológiai teszteket fejlesztettünk. Kilenc angol és 2 magyar nyelvű cikket publikáltunk 27,794 IF értékben. Eredményeinkről 20 alkalommal számoltunk be előadás és 2 alkalommal poszter formájában. A kutatásokban 4 PhD hallgató vett részt, egy közvetlenül ezekből az eredményekből készítette a doktori értekezését. A kutatásokból 3 TDK dolgozat is született.

We have performed all experiments in accordance with the project plan and implemented several extra studies which are closely related to the project as well: - We established Hungarian and international strain collections. - We developed the first MS1 DIVA (differentiating infected from vaccinated animal) system. - We developed a new MS-H DIVA system. - We developed the first ts11, 6/85, F and K strain specific M. gallisepticum DIVA systems. - We developed the first multi-locus variable-number tandem repeat system (MLVA) for the genotyping of M. synoviae and we performed multi-locus sequence typing (MLST) as well. - We developed the first MLST system for M. gallisepticum. - We determined the antibiotic susceptibility profiles of Hungarian and other Central-Eastern European M. synoviae isolates. - We collected strains from Asia and determined their antibiotic susceptibility profile. - We determined the genes and mutations responsible for antibiotic resistance in M. synoviae. - We developed rapid molecular assays for the detection of these genetic markers. Nine international and 2 Hungarian papers were published with a cumulative IF: 27.794. We presented our results on conferences as an oral presentation (20) or as a poster (2). Four PhD student participated in the project. One of them wrote her dissertation from the results. The dissertation (TDK) of three undergraduate students were also prepared from the results.
