A kutatás összefoglalója, célkitűzései szakemberek számára

A diarilheptanoidok növényi eredetű fenolos komponensek, amelyeknek számos előnyös élettani hatását (pl. gyulladáscsökkentő és tumorellenes hatás) igazolták. A diarilheptanoidok jelenlétét igazolták néhány, hazánkban is honos növényben. Korábbi munkánk során jellemeztük a Corylus nemzetség három fajának diarilheptanoid-összetételét. Jelen pályázatunkban célul tűztük ki a diarilheptanoidok új, lehetséges növényi forrásainak feltárását. A Kárpát-medencében előforduló cserjék és fák (Alnus-, Betula-, Carpinus-, Juglans-, Acer-, Morus-fajok) különböző növényi részeiből szelektív extrakciós módszerrel állítunk elő diarilheptanoidokban dús kivonatokat. A kivonatok fitokémiai jellemzésére modern műszeres analitikai módszereket (HPLC-ESI-MS/MS, CE) alkalmazunk. A kivonatokban fellelhető diarilheptanoidokat kromatográfiás módszerekkel (oszlopkromatográfia, preparatív HPLC, CPC) izoláljuk, a komponensek szerkezetét NMR és MS/MS technikákkal azonosítjuk. A pályázat következő célja az izolált diarilheptanoidok citotoxicitásának meghatározása in vitro humán daganatos sejtvonalakon (melanoma, colon carcinoma, emlődaganat, tüdődaganat), illetve in vivo egér melanoma-modellben. A citotoxicitási adatok alapján vizsgáljuk a vegyületek szerkezet-hatás összefüggéseit.
A citotoxicitással rendelkező diarilheptanoidok farmakokinetikai jellemzőit egér szövetmintákból HPLC-MS, CE és NMR módszerekkel vizsgáljuk. Irodalmi adatok alapján a diarilheptanoidok alacsony biohasznosíthatósággal, valamint rossz vízoldhatósággal rendelkeznek, ezeket ciklodextrin zárványkomplexek formulálásával tervezzük javítani. A komplexek strukturális jellemzőit szerkezetvizsgáló módszerekkel jellemezzük.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A tervezett alapkutatás segítségével arra keressük a választ, hogy milyen új, a melanoma terápiájában alkalmazható citotoxikus növényi szekunder anyagcseretermékeket nyerhetünk ki a Kárpát-medencében honos fákból és cserjékből. A kutatás kiinduló hipotézise az, hogy a legelsőként leírt diarilheptán, a kurkumin esetében számos pozitív biológiai (döntően cititoxikus) hatást publikáltak in vivo tesztekben. A növényi fenoloidok ezen csoportjának fitokémiai és biológiai feltérképezése napjainkig nem történt meg, ugyanis a diarilheptán szerkezetekhez rendelhető biológiai hatásokról csak az elmúlt években jelentek meg az első adatok. Előzetes eredményeink alapján további ígéretes (és minden bizonnyal új diarilheptánokat tartalmazó) növényi forrásokat sikerülhet elsőként jellemeznünk. Keressük továbbá a szerkezet-hatás összefüggéseket melanoma sejtvonalon történő teszteléssel és vizsgáljuk a ciklodextrinek hatását a diarilheptanoidok biohasznosíthatóságának növelésére. Kísérleteink alapján elsőként tudjuk megválaszolni, hogy lineáris és ciklikus diarilheptán-származékok milyen metabolikus útvonalakon keresztül alakulnak át in vivo állatkísérletes modellekben.

Mi a kutatás jelentősége?

A tervezett farmakognóziai kutatás jelentősége olyan új, citotoxikus természetes vegyületek (és forrásaik) azonosítása, melyek alapját jelenthetik daganatos elváltozások (elsősorban melanoma) kezelésének. Az alapkutatás eredményeinek köszönhetően megtörténik a megjelölt növénycsaládokba tartozó, a Kárpát-medencében honos fák és cserjék farmakobotanikai feltérképezése. A modern gyógyszerkutatás vezérmolekula-keresésének egyik kitörési pontja a diverz szerkezeti elemekkel rendelkező természetes vegyületek bevonása a nagyhatékonyságú szűrővizsgálatokba. A tervezett alapkutatás során számos olyan bioaktív diarilheptán-származékot tervezünk izolálni és in vitro, illetve in vivo biológiai aktivitását feltérképezni, melyek ígéretes gyógyszerjelölt vegyületekké válhatnak citotoxikus hatásuknak köszönhetően. A legígéretesebb vegyületek metabolitjait is azonosítani kívánjuk, elősegítve ezzel szerkezet-hatás összefüggések és a gyógyszer-szerű viselkedésük molekuláris szintű megértését. A kedvezőtlen biofarmáciai tulajdonsággal rendelkező származékok biohasznosíthatóságát ciklodextrinek segítségével tervezzük növelni, az előállított komplexek fizikokémiai és analitikai karakterizálását követően vizsgáljuk biohasznosíhatóságukat is. Eredményeink ezáltal megteremtik a természetes eredetű citotoxikus diarilheptanoidok hasznosíthatóságának lehetőségét a humán daganatos megbetegedésekben, melynek a hazai gyógyszeripar potenciális erősödését és az egészséges társadalom célkitűzéseket egyaránt megvalósítja.
A megpályázott területen versenytársak mindazok az egyetemi/kutatóintézeti csoportok, akik a bioaktív természetes vegyületek kutatásával foglalkoznak. Tekintettel arra, hogy a diarilheptanoidok biológiai hatásának feltérképezése az utóbbi 5 évben intenzíven kutatott területté vált, így egyre több nemzetközi kutatócsoport kezdett foglalkozni e vegyületekkel. A Semmelweis Egyetem Farmakognóziai Intézetében évtizedek óta folyó fenoloid kutatás kiegészülve az I.sz. Patológiai és Kísérleti Rákkutató Intézetben folyó biológiai szűrökkel, a citotoxikus természetes vegyületek jellemzésének egy vezető csoportjává válhat.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

A diarilheptanoidok természetes eredetű vegyületek, legismertebb képviselőjük a kurkuma gyökerében nagy mennyiségben megtalálható kurkumin. A diarilheptanoidok gyógyászati jelentőségét és terápiás felhasználásának lehetőségét tudományosan is igazolt gyulladáscsökkentő és tumorellenes hatásaik támasztják alá.
Jelen pályázatunkban célunk a diarilheptanoidok új, lehetséges növényi forrásainak feltárása. A Kárpát-medencében előforduló cserjék és fák – éger-, nyír-, gyertyán-, dió-, eperfa- és juhar-fajok – különböző növényi részeiből (pl. kéreg-, gyökérkéreg-, levél-, termős és porzós virágzat-, valamint termés-mintákból) célzott kivonási módszerrel állítunk elő diarilheptanoidokban dús extraktumokat. A kivonatok minőségi és mennyiségi összetételének jellemzésére modern műszeres analitikai módszereket: nagyhatékonyságú folyadékkromatográfiát és kapilláris elektroforézist alkalmazunk. A kivonatokban fellelhető diarilheptanoidokat hagyományos és műszeres kromatográfiás módszerekkel izoláljuk, szerkezetüket mágneses magrezonancia spektroszkópiás és tandem tömegspektrometriás technikákkal azonosítjuk.
A pályázat következő fontos célja az izolált diarilheptanoidok tumorellenes hatásának meghatározása humán daganatos sejtvonalakon (melanoma, vastagbél-, emlő- és tüdődaganat), illetve egér melanoma-modellben. A daganatellenes hatással rendelkező diarilheptanoidok szervezetbeli sorsát jellemző farmakokinetikai tulajdonságokat egér szövetmintákban vizsgáljuk. A diarilheptanoidok terápiás alkalmazását korlátozó alacsony biohasznosíthatóságot ciklodextrin-alapú komplexek formulálásával tervezzük javítani.

Summary of the research and its aims for experts

Diarylheptanoids represent phenolic compounds of plant origin. Numerous effects have been proved for them including anti-inflammatory and anti-tumor activity. Occurence of diarylheptanoids have been confirmed in several plant sepcies native to Hungary. In our previous work the diarylheptanoid composition of species of the Corylus genus has been characterized. In the proposed study we plan to reveal potential new plant sources of diarylheptanoids. Extracts rich in diarylheptanoids will be prepared from different plant parts of trees and shrubs distributed in Central Europe (Alnus, Betula, Carpinus, Juglans, Acer, Morus species) using a selective extraction technique. For the phytochemical characterization, instrumental methods of analysis (HPLC-ESI-MS/MS, CE) will be applied. Diarylheptanoids present in the samples are proposed to be isolated by chromatographic methods (CC, preparative HPLC, CPC), structure elucidation of the constituents will be performed using NMR and MS/MS.
Further specific aim is the evaluation of cytotoxic activity of the isolated compounds in vitro against human cancer cell lines (melanoma, colon cancer, breast cancer, lung cancer), as well as in vivo in a murine melanoma model.
According to cytotoxicity of the compounds, structure-activity relationship will be evaluated. Pharmacokinetic properties of the diarylheptanoids are planned to be investigated in murine tissue samples by HPLC-MS, CE and NMR. According to the literature, bioavailability and aqueous solubility of diarylheptanoids is poor, these are proposed to be increased by formulation of cyclodextrin inclusion complexes. Structural characterization of the complexes will be performed.

What is the major research question?

The proposed research aimed at identifying new cytotoxic secondary plant metabolites from trees and shrubs native to the Carpathian Basin for the treatment of melanoma. The starting hypothesis resides in the numerous beneficial bioactivity of curcumin the first characterized diarylheptanoid. The complete phytochemical and biological characterization of this promising group of plant phenolics is still incomplete, as the first structure-activity data on these compounds appeared in the last few years. Based on our preliminary data, we plan to report new sources of diarylheptanoids with diverse structures in specified plant genera for the first time. Structure-activity relationships will be established based on screening experiments using melanoma cell lines. The effect of cyclodextrins on the bioavailability of diarylheptanoids is also planned to be addressed. Using in vivo animal models, the metabolic pathways of linear and cyclic diarylheptanoids will be addressed for the first time as well.

What is the significance of the research?

Identification of novel, cytotoxic compounds of natural origin (and that of their sources) represents the significance of the proposed pharmacognostical investigation. These compounds may provide a basis for the therapy of tumorous diseases (principally melanoma). The results of the basic research afford the pharmacobotanical revealing of the plants, which are member of the selected families and are native to Central Europe. Involving natural products with diverse structural characteristics into the high-throughput screening represents the key issue in the search for lead molecules in the course of modern drug discovery. During the proposed basic research we plan the isolation and the investigation of in vitro and in vivo biological activities of diarylheptane derivatives, which may be promising therapeutic candidate molecules due to their cytotoxic effect. We propose to identify the structure of the most promising compounds as well, thus promoting the molecular-level understanding of structure-activity relationships and the drug-like behavior of the compounds. We plan to enhance the bioavailability of the diarylheptanoid derivatives possessing unfavorable biopharmaceutical properties by the help of cyclodextrins. Besides the phisicochemical and analytical characterization of the formulated complexes, their bioavailability will be evaluated as well. Our results will provide a possibility for the use of natural, cytotoxic diarylheptanoids in the therapy of human tumor diseases. This will realize the potential recruitment of the national pharmaceutical industry, as well as the objective of the healthy society.
In regard to, that revealing the biological activities of diarylheptanoids has come to an intensively studied research field in the past five years, the number of international research teams dealing with these compounds is increasing. Due to the phenolic research running for decades in the Department, complemented with biological screening performed in the 1st Department of Pathology and Experimental Cancer Research, the Department of Pharmacognosy may turn into one of the leading research teams being concerned with the characterization of cytotoxic natural products.

Diaryheptanoids are natural compounds. A well-known representative, occuring in the rhizomes of turmeric, is curcumin. Medicinal significance of diarylheptanoids and perspectives of their therapeutic use are supported by their scientifically proven anti-inflammatory and anti-tumor activities.
In the proposed work we aim to reveal potential new plant sources of diarylheptanoids. Extracts rich in diarylheptanoids will be prepared from different plant parts (e.g. bark, rootbark, leaf, female and male flower and fruit samples) of trees and shrubs distributed in Central Europe (alder, birch, hornbeam, walnut, mulberry and maple species) using a selective extraction method. The qualitative and quantitative composition of the extracts will be characterized using modern instrumental techniques of analysis: high-performance liquid chromatography and capillary electrophoresis. Diarylheptanoids will be isolated from the extracts using classical and instrumental chromatographic techniques, their structures will be determined by nuclear magnetic resonance spectroscopy and tandem mass spectrometry.
The second specific aim of the work is the evaluation of the anti-tumor activity of diarylheptanoids against human cancer cell lines (melanoma, colon cancer, breast cancer, lung cancer), as well as in a murine melanoma model.
According to cytotoxicity of the compounds, structure-activity relationship will be evaluated. Pharmacokinetic properties characterizing the fate of diarylheptanoids in the organism will be studied in mouse tissue samples. The poor bioavailability of diarylheptanoids, limiting their therapeutic use, is planned to be increased by formulation of cyclodextrin complexes.

A kutatás során a Betulaceae, Moraceae, Juglandaceae, Sapindaceae, Cannabaceae és Fabaceae családba tartozó növényfajok diarilheptanoidjainak teljeskörű jellemzését végeztük. Optimalizált extrakciós eljárást fejlesztettünk a diarilheptanoid-típusú vegyületek kinyerésére növényi mintákból. Elsőként vizsgáltuk az Alnus × spaethii, Celtis occidentalis, Acer campestre és Acer pseudoplatanus fenoloidprofilját és leírtuk ciklusos és lineáris diarilheptanoidok jelenlétét Alnus cordata mintákban. Elsőként szolgáltattunk kvalitatív és kvantitatív adatokat a Carpinus betulusban előforduló diarilheptanoidokról. Szintén elsőként írtunk le ciklusos diarilheptanoidokat Corylus maxima kivonatokban. Az alábbi vegyületeket izoláltuk: hét diarilheptanoidot Alnus glutinosaból, tizenkét fenoloidot (nyolc diarilheptanoidot) C. betulusból, hét diarilheptanoidot C. maximaból, hat alkilamidot C. occidentalisból és öt fenoloidot Juglans nigraból. Az izolált vegyületek közül a karpinontriol A in vitro antiproliferatív hatást mutatott A2058 humán melanóma sejtvonallal szemben. Elsőként szolgáltattunk adatokat néhány diarilheptanoid-típusú vegyület kémiai stabilitásáról és membránon keresztül történő passzív diffúziójáról. Ciklodextrin-zárványkomplexeket állítottunk elő a diarilheptanoidok stabilitásának és oldhatóságának javítása céljából.
Results in English
We accomplished the comprehensive characterization of diarylheptanoids from species belonging to the Betulaceae, Moraceae, Juglandaceae, Sapindaceae, Cannabaceae, and Fabaceae families. We optimized the extraction procedure of diarylheptanoids from plant samples. We analyzed the phenolic profile of Alnus × spaethii, Celtis occidentalis, Acer campestre, and Acer pseudoplatanus, and described several cyclic and linear diarylheptanoids in Alnus cordata for the first time. We provided new data on the presence and quantities of diarylheptanoids in Carpinus betulus. We reported the occurrence of cyclic diarylheptanoids in Corylus maxima for the first time as well. We isolated seven diarylheptanoids from Alnus glutinosa, twelve phenolic compounds (eight diarylheptanoids) from C. betulus, seven diarylheptanoids from C. maxima, six alkylamides from C. occidentalis, and five phenolic compounds from Juglans nigra. Among the isolated compounds, carpinontriol A showed in vitro antiproliferative activity against A2058 human melanoma cells. We have also assessed the chemical stability and membrane permeability of several Corylus and Carpinus diarylheptanoids. We have formulated cyclodextrin inclusion complexes with some of the isolated diarylheptanoids, in order to improve their stability and aqueous solubility.
