Az extrecelluláris vezikulák (EV-k) a sejtek közötti kommunikáció egészen az elmúlt évekig kis jelentőségűnek tulajdonított szereplői. Számos fiziológiás és patológiás folyamatban vesznek részt, így napjainkban intenzíven kutatott terület az EV-k diagnosztikában történő alkalmazása. A területen megjelent számos publikáció ellenére mostanáig még nem történt áttörés a klinikumban az EV-k biomarkerként való alkalmazásában, aminek egyik oka, hogy nem állnak rendelkezésre standard nanorészecskék, melyek fiziko-kémiai tulajdonságaikban (méret és törésmutató) és összetételükben megközelítik az EV-ket.
Jelen kutatási projekt célkitűzése a biológiai referencia részecskék előállítása EV-k számára és azok részletes jellemzése. A projekt keretein belül liposzómák és organoszilika nanorészecskék előállítását és széles körű jellemzését tervezem. A liposzómák EV referencia részecskeként történő felhasználása az összetételükből eredő hasonlóság alapján egyértelműen adódik, azonban a nagyobb méretű (>200 nm) szűk méreteloszlású liposzómák előállítása ezidáig nehézségekbe ütközött. Az előállítani tervezett organoszilika nanorészecskék ötvözik a szilárd nanorészecskék kedvező tulajdonságait, ugyanakkor mezopórusos szerkezetükből és összetételükből adódóan a vezikulákéhoz hasonló törésmutatóval rendelkezhetnek.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Jelenleg nem érhetőek el standard nanorészecskék extracelluláris vezikulák méret-meghatározásához használt leggyakoribb módszerekhez, így nem lehetséges a különböző laboratóriumok és kórházak által elvégzett vizsgálatok egyértelmű összehasonlítása. Jelen pályázat célja EV-k biológiai referencia részecskéinek előállítása. Az EV-k biológiai referencia részecskéinek a vezikulákéhoz hasonló fiziko-kémiai tulajdonságokkal kell rendelkezniük, ami főként a méretet (50 – 1000 nm) és a törésmutatót, valamint az összetételt érinti.
A tervezett liposzómák magától értetődően vetődnek fel, mint az EV-k referencia részecskéi, ugyanakkor a nagyméretű liposzómák extrudálással történő előállítása túl széles méreteloszlást eredményez. Jelen pályázatban erre nyújt megoldást a szilícium mikorsziták alkalmazása, ami még nem ismert az irodalomban. Szintén új megközelítés a tervezett organoszilika részecskék előállítása és foszfolipidekkel történő felületmódosítása. Az organoszilika részecskék jól meghatározott mérete, és összetételükből adódó alacsony törésmutatója ideális referencia részecskékké teszi azokat.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. A tervezett biológiai nanorészecskék előállítása nagymértékben hozzájárul az extracelluláris vezikulák területén, a különböző detektálási és méretmeghatározási módszerek standardizációjához. Ez utóbbi lehetővé teheti a különböző laboratóriumok és kórházak mérési eredményeinek összehasonlítását, ami szükséges feltétele az EV-k diagnosztikában történő felhasználásának. A nemzetközi együttműködésben elvégzett kutatás eredménye, azaz EV-k biológiai referencia-jelölt részecskéinek előállítása folytatása lehet egy, számos kórházat érintő laboratóriumok közötti összehasonlító vizsgálatnak. Így az EV-k biológiai referencia részecskéinek előállítása hozzájárul azok klinikai diagnosztikában történő alkalmazásához, tehát a kóros elváltozások gyorsabb és pontosabb felismeréséhez.

A tervezett kutatást széles hazai és nemzetközi együttműködésben tervezzük megvalósítani. Jelen projektben Prof. Buzás Edit csoportjával tervezünk közös munkát, amely nemcsak a hazai, de a nemzetközi EV kutatásban is úttörő. A kifejleszteni tervezett referencia nanorészecskék tesztelésében és a laboratóriumok közötti összehasonlító vizsgálatok kivitelezésében Dr. Rienk Nieuwland (AMC University of Amsterdam) kutatócsoportjával szándékozunk együttműködni.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

Az extracelluláris vezikulák (EV-k) olyan biológiai eredetű nanorészecskék, melyek segítségével az elő szervezetek sejtjei egymással kommunikálnak. Az EV-k számos élettani folyamatban közreműködnek, így különböző betegségek esetén is szerephez jutnak. Napjainkban azonban még nem terjedt el az EV alapú diagnosztika, ami ezen részecskék kimutatásának valamint méretük és koncentrációjuk meghatározásának nehézségeivel magyarázható. Jelen pályázat célja olyan biológiai referencia részecskék kifejlesztése, amelyek tulajdonságaikban megközelítik a természetes EV-ket. Ilyen referencia részecskék kifejlesztésével lehetőség nyílik a különböző mérési módszerek eredményeinek elfogadott standardhoz történő viszonyítására, ami lehetővé teheti az EV-k felhasználását különböző megbetegedések gyors, és pontos diagnosztizálásához.
Az előállítani tervezett részecskék magukban foglalják a kozmetikai iparban és a gyógyászatban is használt liposzómákat, és a különböző élelmiszerekben is használt szilika nanorészecskékhez hasonló anyagokat.

Extracellular vesicles (EVs) are the players of intercellular communication, which attracted minor interest until recently. EVs are involved in many physiological and pathological conditions, so the use of EVs in diagnostics is the subject of active research in the field. Despite the numerous publications in the field, breakthrough in the use of EVs as biomarkers has not yet achieved due to the lack of standard nanoparticles, which mimics the physicochemical properties (size and refractive index) and composition of the EVs.
The goal of this research project is to develop biological reference nanoparticles for EVs and their detailed characterization. The preparation and broad characterization of liposomes and organosilica nanoparticles are planned in the proposed project. The use of liposomes as reference materials for EVs is straightforward owing the similarity in their composition, but the preparation of large sized (>200 nm) monodisperse liposomes is challenging. The planned organosilica nanoparticles combine the favorable properties of solid nanoparticles with the tunable refractive index of mesoporous materials that approaches the properties of EVs.

What is the major research question?

Standard nanoparticles for the size characterization methods of EVs are currently not available, so comparison of the results from different laboratories and hospitals cannot be performed. This project aims the development of biological reference materials for EVs. Those particles are expected to possess similar physicochemical properties to EVs, which mainly concerns their size (50 – 1000 nm), refractive indices, and composition.
The liposomes planned in this project are naturally comes into focus as reference materials for EVs, but the extruded liposomes in the size range above 200 nm possess broad size distribution. The use of silicon microsieves during the extrusion is proposed in this project, which was not reported in the literature before. Also the surface modification of organosilica nanoparticles is a new approach. The well determined size of organosilica particles and their low refractive index due to their structure and composition makes them ideal candidates for reference materials for EVs.

What is the significance of the research?

The planned development of biological nanoparticles for EV research contributes to the standardization of measurement techniques in the field. This enables the comparison of the results of different research groups and hospitals, what is a prerequisite for the use of EVs in diagnostics. The result of this work performed in an international cooperation, namely the development of candidate biological reference materials for EVs, might be continued in a worldwide interlaboratory comparison study. In this way, the development of EV reference materials contributes to their application in diagnostics, with other words, it helps the faster and more reliable diagnosis of diseases.

This research project will be carried out in a wide national and international collaboration. Within this research project we intend to work together with the working group of Prof. Edit Buzás, whose pioneering research in the EV field is renowned. The testing of the EV reference particles and the inter-laboratory comparison study will be performed in cooperation with the group of Dr. Rienk Nieuwland (AMC University of Amsterdam).

Extracellular vesicles (EVs) are biological nanoparticles that are used for the communication between the cells of living organisms. The EVs are involved in many physiological conditions, and they also play a role in diseases. However, the EV based diagnosis in hospitals is not yet reached, due to the difficulties associated to the detection of these particles and the characterization of their size and concentration. The goal of this project is to develop reference nanoparticles that resemble the properties of the naturally occurring EVs. The development of such particles contributes to relate the measurement results from different laboratories to international standards. The latter will enable the use of EVs in diagnosis of different diseases.
The reference particles to be developed covers liposomes, which are also used in cosmetics and health care, and silica nanoparticles similar to which are used in food additives.

A projekt keretében üreges organoszilika (HOB) és liposzóma minták előállítása, jellemzése és extracelluláris vezikulák méret és koncentráció-meghatározásához szükséges referencia mintaként történő alkalmazhatóságának tesztelése történ meg. A HOB minták tömör részecske templátok felhasználásával, szol-gél eljárással készültek, morfológiai jellemzésük, méreteloszlásuk meghatározása és kolloidstabilitásuk vizsgálata számos módszer felhasználásával történt meg. A HOB-k referencia-részecskékként való alkalmazhatóságát áramlási citometriával vizsgáltuk vérlemezke-eredetű EV-k felhasználásával. Mivel a HOB-k hasonlítanak az EV-k szerkezetére és fényszórási tulajdonságaira, azok alkalmasak az EV-koncentráció fényszóráson alapuló meghatározásának egy előre meghatározott mérettartományon belüli szabványosítására. A szintetikus liposzómák részecskeszám-koncentrációját röntgendiffrakció és mikrofluidikus ellenállás impulzus mérés segítségével határoztuk meg. A projekt eredményei hat kézirat és hét konferencia-előadásban kerültek bemutatásra. Három BSc, egy MSc és két PhD hallgató vett részt a kutatásban. A kutatási projekt fő eredménye, hogy a HOB-részecskékkel kapcsolatos eredmények egy nemzetközi projekthez vezettek (METVES II, www.metves.eu), amelyben a jelen pályázat vezető kutatója a „Referenciaanyagok kidolgozása” című munkacsomag felelőse.
Results in English
Hollow organosilica beads (HOBs) and liposomes were prepared, characterized and tested as reference particles for size and concentration measurements of extracellular vesicles (EVs). HOBs were prepared by a hard template sol-gel method and extensively characterized for morphology, size and colloidal stability. The applicability of HOBs as reference particles was investigated by flow cytometry using HOBs and platelet-derived EVs. Because HOBs resemble the structure and light scattering properties of EVs, HOBs with a given size will gate EVs of the same size. Therefore, HOBs are ideal reference beads to standardize optical measurements of EV concentration within a predefined size range. The particle number concentration of synthetic liposomes was determined using X-ray diffraction and microfluidic resistive pulse sensing. Results of the project were summarized in six manuscripts and seven conference presentations. Three BSc, one MSc and two PhD students were involved in the research. The main achievement of this research project is that the results on HOB particles led to an international project called METVES II (www.metves.eu) in which the PI will lead the work package on “Development of reference materials”.
