Az elmúlt években a tuberculosis (tbc) megelőzésére és/vagy gyógyítására kifejlesztett peptid-alapú szintetikus oltóanyagok jelentős érdeklődésre tartanak számot. Jelen kutatás célja egy olyan multi-eptóp konjugátumot tartalmazó vakcina-jelölt kifejlesztése, mely jelentős mértékben képes kiváltani Th1 sejtes immunválaszt. A konjugátumok szintézise során a kiválasztott epitópokat kemoszelektív kapcsolási eljárások segítségével lipo-peptid típusú hordozóhoz kötök, majd jellemzem azok kémiai sajátságait. A szintetikus vakcinák hatékonysága és biohasznosulása fokozható a megfelelő formulázással. Kísérleteimben poli tejsav-glikolsav kopolimer (PLGA) nanorendszereket, illetve ciklodextrin-típusú formulázási technikákat alkalmazok. Az immunogenitás növelésére adjuváns (pl. bakteriális sejtfal alkotó glikolipid) beépítését tervezem. Az előállított készítmények immunstimuláló hatását in vivo vizsgálom egér modellen.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

Olyan betegségekben, ahol a kórokozó nagy antigén-diverzitást, illetve általános változékonyságot mutat (pl. gyorsan mutálódó vírusok, különböző életformát felvenni képes baktériumok) nagy jelentőséggel bírnak az olyan oltóanyagok, melyek egy preparátumban többféle immundetermináns részt is tartalmaznak. Peptid-alapú vakcinákban különböző eredetű epitópokat kombinálhatunk, így kiváltva egy általánosabb, hatékonyabb immunválaszt. A tbc kórokozója egy olyan baktérium, mely a fertőzés után ún. dormans állapotban akár évtizedekig is megbújhat az emberi szervezetben. Ez a látens fertőzés az emberiség egyharmadát érinti és komoly népegészségügyi problémákat okoz elsősorban azért, mert a dormans baktérium egyes kórképekben (pl. HIV fertőzés) újra aktiválódhat. A baktérium különböző életciklusaiban termelődött fehérjék T-sejt epitópjait egy lipo-peptid hordozóhoz konjugálok és az így előállított vakcina-jelöltek immunválasz-kiváltó hatását többféle formulációban vizsgálom.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

Az elmúlt években jelentősen megnőtt a klinikai tesztelés alatt álló peptid-alapú szintetikus vakcinák száma, melyek segítségével harcolni tudunk különböző tumoros megbetegedések, autoimmun betegségek és fertőző betegségek ellen. A szintetikus vakcinák azonban önmagukban általában nem elég immunogének, ezért immunstimuláló ágensek, ún. adjuvánsok hozzáadását igénylik. Az alkalmazott adjuváns és formuláció alapvetően meghatározza az immunválasz mértékét és minőségét. A tbc területén végzett kutatások bizonyították, hogy a vakcinák által kiváltott protektív immunitás eléréséhez feltétlenül szükség van az immunválasz Th1 polarizációjára. Ezért olyan technikákat alkalmazok, melyek növelhetik az epitóp peptidek által kiváltott Th1 citokinek termelését, hatékonyabb vakcina-formulázást eredményezve ezáltal. A kapott eredmények általánosan használhatóak majd a vakcina kutatásban, de főként a tbc prevenció és terápia területén nyithatnak új perspektívákat.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

A különböző megbetegedések megelőzésére illetve gyógyítására alkalmazott oltóanyagokkal szemben fontos elvárás, hogy ne okozzanak lokális és/vagy szisztémás mellékhatásokat. A szintetikus vakcinák – szemben az elölt vagy legyengített kórokozót tartalmazó oltóanyagokkal – jóval kisebb eséllyel okoznak ilyen mellékhatásokat, viszont nem is feltétlenül elég hatékonyak. Jelen kutatásban olyan technikákat kívánok kidolgozni, melyek segítségével növelhető a szintetikus oltóanyagok hatékonysága. Kutatásaimat elsősorban a tuberculosis (tbc) területén szeretném felhasználni, mivel a jelenleg alkalmazott BCG oltás felnőtt korban már nem nyújt megfelelő védettséget és újraoltás esetén mellékhatásokat okozhat. Mivel a vakcinák által okozott mellékhatások előfordulása jelentős mértékben leronthatja az átoltottság arányát, az oltóanyagok hatékonyságának növelése és a mellékhatások visszaszorítása komoly közegészségügyi jelentőséggel bírnak.

Over the last few years, prevention and/or treatment of tuberculosis (TB) with peptide-based synthetic vaccines attracted great attention. The main goal of the recent proposal is to develop a multi-epitope conjugate containing vaccine candidate, which can efficiently induce Th1 cellular immune response. Chemoselective coupling methods will be employed to conjugate selected epitopes to a lipo-peptide scaffold and the products will be chemically characterized. Vaccine efficacy and bioavailability of synthetic subunit vaccines can be improved with proper formulation. In this project poly(lactic-co-glycolic acid) (PLGA) copolymer and cyclodextrin-based formulation will be investigated. In order to increase the immunogenicity adjuvants, such as bacterial cell wall components, will be incorporated to nano-constructs. Preparations will be assayed in vivo for their immunstimulating activity in a murine model.

What is the major research question?

Research and development of vaccines against a variety of infectious diseases, where the causative agent shows high antigenic diversity due to fast mutation rate or metamorphosis across variation of stages, are still challenging. Therefore, immunization with combination of antigens derived from all stages of the parasite’s life cycle has crucial benefits. Multiple epitope containing polyvalent vaccines can induce a more effective and universal immune response. The causative agent of tuberculosis (TB) can render itself to a so-called dormant stage and remains in the human body for over decades after the infection. One third of the world’s population is affected with this latent infection which comprises a significant reservoir of future epidemics, particularly in countries with high HIV burdens. In this project, epitopes derived from immundominant proteins expressed during the different stages of the pathogen will be conjugated to a lipo-peptide carrier. The conjugates will be formulated in different ways and the provoked immune response will be measured.

What is the significance of the research?

The number of peptide-based subunit vaccines in clinical trials against diseases, like different types of cancer, autoimmune, allergic and infectious diseases, greatly increased in the past few years. However, subunit vaccines have generally proven inefficacy at inducing potent immunity. They rely on the delivery of immunstimulatory agents, so-called adjuvants. The used formulation and the type of adjuvants can significantly influence the immune activation and can regulate the polarity of the immune response. In the case of tuberculosis, it has been proven that potent Th1 immune response is needed to reach a long-lasting protective immunity. Therefore, the goal of this project is to develop new adjuvanted formulations which can optimize the amount of the released Th1 cytokines. Expected results of this project will have crucial benefits and opening new perspectives in vaccine research, particularly in the prevention of tuberculosis.

To minimize local and/or systemic adverse effects is crucial to new vaccine development. Synthetic vaccines are absent of potentially infectious or allergic material, which compromise many live-attenuated or inactivated pathogen containing vaccines. However, the protective efficacy of synthetic vaccines used alone is usually insufficient. In the present project, new techniques are being investigated with the aim to enhance the efficacy of synthetic vaccines. In the case of tuberculosis, recently applied vaccination (BCG) does not provide effective long-lasting immunity therefore adults are not protected sufficiently. As re-immunizing adults with BCG can cause serious side effects new type of TB vaccination is urgently needed. In general, new techniques aiming at lowering the incidence of adverse events and improving efficacy can greatly influence the rate of vaccination and has great benefits for public healthcare.

A tuberkulózis kórokozója nagy antigén-diverzitást, illetve általános változékonyságot mutat, ami megnehezíti a betegség elleni védekezést. A jelenleg alkalmazott oltás hatékonysága nem kielégítő, mert nem reflektál megfelelően a különböző baktérium populációkra. Peptid-alapú szintetikus vakcinákban különböző eredetű immundeterminánsokat kombinálhatunk, így váltva ki egy általánosabb immunválaszt, de a peptidek alacsony biohasznosíthatóságát és immunogenitását mindenképpen fokozni kell. Kutatásaim elsősorban arra irányultak, hogy kémiai módszerek segítségével növelni tudjuk a peptidek immunogenitását és in vivo stabilitását. A kísérletekhez 3 Mycobacterium tuberculosis eredetű T-sejt epitóp peptidet választottam és kemoszelektív ligációs technikák alkalmazásával elágazó láncú konstrukciótat állítottam elő. A gyors lebomlás kiküszöbölésére a konjugátumokat polimer (PLGA) nanokrészecskékbe csomagoltuk és módszert dolgoztunk ki a nanorészecskék felszínének peptid-módosítására is. A konjugálás és a nanokapszulázás nagyban növelte az epitóp peptidek internalizációját makrofág-típusú sejtekbe és jelentős T-sejt proliferáció és citokintermelés volt mérhető. Megállapítottuk továbbá, hogy a konstrukciók protektív immunitást váltottak ki M. tuberculosis fertőzött egér állatmodellen. Az eredményeket, melyek hozzájárulhatnak a peptid-alapú vakcinák továbbfejlesztéséhez, számos hazai és nemzetközi konferencián mutattuk be és publikáltuk rangos folyóiratokban.
Results in English
The most challenging factor in preventing tuberculosis is the ability to provide immunity against the multiple stages of the pathogen and provide cross-protection within the different subtypes. Immunization with a multivalent subunit vaccine, which combines multiple antigens derived from different stages of the pathogen’s life cycle, can reflect on the antigen diversity. The use of minimal epitope sequences, which can trigger the desired immune response, is a safe and reliable approach however, the limited immunogenicity and low bioavailability needs to be addressed. In this project, three promiscuous T-cell epitopes from different proteins expressed by Mycobacterium tuberculosis were selected based on previously reported antigenic properties. Epitope peptides were conjugated using different chemoselective ligation techniques to produce a branched-chain compound. To avoid the fast in vivo degradation, conjugates were encapsulated into polymeric (PLGA) nanoparticles (NP). Besides, a convenient synthetic method to cover the surface of the NPs with targeting peptides was also developed. Conjugation and nanocapsulation greatly improved the internalization to immune cells and a significant T-cell proliferation and cytokine response was measured. Furthermore, the constructs showed relevant vaccine efficacy against M. tuberculosis infection in a murine model of TB. Results were presented on several national and international conferences and published in relevant scientific papers.
