A kutatás összefoglalója, célkitűzései szakemberek számára
Itt írja le a kutatás fő célkitűzéseit a témában jártas szakember számára.

Biológiai aktivitással rendelkező makromolekulák és ionok (fehérjék, peptidek, fehérje fragmentumok, karbonsavak, komplexek stb.) biológiai és környezeti mintákból történő analízise folyamatosan jelen levő probléma köszönhetően a mintaoldatok nagyfokú komplexitásának. A biológiai és környezeti analitikában a legfontosabb és gyakran a legbonyolultabb feladat a vizsgált komponensek elválasztása. A kromatográfiás hatékonyságot és szelektivitást jelentősen befolyásolják az oszlopban lejátszódó egyensúlyi és kinetikai folyamatok. Átfogó analitikai módszerek fejlesztéséhez szükséges ezen folyamatok mély és alapos ismerete. Elméleti modellekkel a hatékony kromatográfiás módszerek és új típusú állófázisok fejlesztése eredményesen támogatható.

Mindezek alapján a benyújtott kutatási projekt az alábbi három, összefüggő részből áll:
(1) bioaktív makromolekulák (fehérjék, peptidek) retenciós viselkedésének tanulmányozása ultranagy-hatékonyságú folyadékkromatográfiában,
(2) adatfeldolgozó eljárások fejlesztése kromatográfiás felbontás és detektálhatóság növelése érdekében,
(3) ionkromatográfiás módszerek fejlesztése bioaktív ionok meghatározására.

Mi a kutatás alapkérdése?
Ebben a részben írja le röviden, hogy mi a kutatás segítségével megválaszolni kívánt probléma, mi a kutatás kiinduló hipotézise, milyen kérdéseket válaszolnak meg a kísérletek.

A kutatás célja bioanalitikai célú folyadékkromatográfiás fejlesztések támogatása és elősegítése alapvető elméleti problémák vizsgálata és adatfeldolgozó algoritmusok fejlesztése révén. A tervezett kutatás az alábbi kérdéseket válaszolja meg, ill. az alábbi eredményekkel szolgál:
- bioaktív makromolekulák analitikai elválasztását befolyásoló legfontosabb egyensúlyi és kinetikai folyamatok feltárása,
- hatékony módszerek és eljárások fejlesztése retenciós folyamatok vizsgálatára a nagyhatékonyságú folyadékkromatográfiában,
- szerves és szervetlen anionok retenciós viselkedése közti különbségek feltárása izotermák vizsgálata alapján,
- szelektív és hatékony módszer fejlesztése kapacitás gradiens alkalmazásával karbonsavak analitikai elválasztására,
- hatékony adatfeldolgozó eljárások fejlesztése kromatográfiás csúcsok detektálásának és felbontásának javítására.

Mi a kutatás jelentősége?
Röviden írja le, milyen új perspektívát nyitnak az alapkutatásban az elért eredmények, milyen társadalmi hasznosíthatóságnak teremtik meg a tudományos alapját. Mutassa be, hogy a megpályázott kutatási területen lévő hazai és a nemzetközi versenytársaihoz képest melyek az egyediségei és erősségei a pályázatának!

A modern környezeti és biokémiai kutatások (pl. proteomika, metabolomika, lipodomika) során egyre bonyolultabb összetételű minták analízisére van szükség. Ezen minták teljes körű analízise a jelnleg alkalmazott folyadékkromatográfiás technikák továbbfejlesztését igényli. A biokémiai, környezeti és gyógyszeripari kutatások számára nélkülözhetetlen olyan nagyhatékonyságú folyadékkromatogárifás módszerek fejlesztése, mellyel lehetőség nyílik komplex összetételű biológia és környezeti minták analízisére. A tervezett kutatás a bioanalitikai célú folyadékkromatográfiás fejlesztéseket támogatja alapvető elméleti problémák vizsgálata és adatfeldolgozó algoritmusok fejlesztése révén. A biológia makromolekulák retenciós viselkedésének tanulmányozása során kapott eredmények az elméleti jelentőségükön túl közvetlenül felhasználhatók ezen komponensek analitikájának fejlesztésében és optimálásában. Az eredmények emellett praktikus javaslatokkal is szolgálnak az analitikus számára az optimális elválasztási körülmények és módszerek tekintetében. Az így kapott állófázisok alkalmazása növelheti a fehérjék és peptidek analitikai meghatározásának megbízhatóságát, hozzájárulva biológiai, biokémiai folyamatok mélyebb megértéséhez, biológiai gyógyszerek fejlesztéséhez és gyártásához. A projekt során fejlesztendő adatfeldolgozó algoritmusok nagymértékben javíthatják a nagyhatékonyságú folyadékkromatográfiás elválasztások megbízhatóságát.

A benyújtott pályázat fő erőssége, hogy többféle kutatási részterületre és megközelítésre támaszkodik (elválasztástudomány, termodinamika, matematikai modellezés, kemometria, tudományos programozás) és a projekt megvalósítása során ezek megfelelő arányban kerülnek alkalmazásra.

A kutatás összefoglalója, célkitűzései laikusok számára
Ebben a fejezetben írja le a kutatás fő célkitűzéseit alapműveltséggel rendelkező laikusok számára. Ez az összefoglaló a döntéshozók, a média, illetve az érdeklődők tájékoztatása szempontjából különösen fontos az NKFI Hivatal számára.

A modern biológia és környezeti analitikában az analitikusnak gyakran nagy számú komponenst tartalmazó minták összetételét kell megvizsgálnia. Mindez komoly kihívást jelent. A nagyhatékonyságú folyadékkromatográfiás módszerek megfelelő felbontást biztosíthatnak bonyolult összetételű minták analíziséhez. A hatékony analitikai módszerek további fejlesztéséhez azonban szükség van a analízis során lejátszódó fizikai és kémiai folyamatok alapos megismerésére. A kutatás célja biológiailag aktív makromolekulák és ionok (fehérjék, peptidek, fehérje fragmentumok, karbonsavak, komplexek stb.) nagyhatékonyságú analitikai elválasztását befolyásoló alapvető elméleti és gyakorlati szempontok vizsgálata. Jelen kutatás három fő részből áll:
(1) bioaktív makromolekulák (fehérjék, peptidek) retenciós viselkedésének tanulmányozása ultranagy-hatékonyságú folyadékkromatográfiában,
(2) adatfeldolgozó eljárások fejlesztése kromatográfiás felbontás és detektálhatóság növelése érdekében,

Analysis of biochemically active macromolecules and ionizable components (proteins, peptides, protein fragments, carboxylic acids, complexes, etc.) in biological and environmental fluids has long been an analytical problem, mostly because of the complex nature of the sample solutions. In biological and environmental analysis, the most important and sometimes most difficult task is the separation of the complex mixture of molecules. The efficiency and selectivity of a chromatographic analysis depends on the equilibrium and kinetic processes take place in the column during elution. Development of comprehensive analytical solutions requires the understanding of these processes. Suitable theoretical models can support the development of efficient chromatographic separation methods and stationary phases.

Accordingly, the goals of the proposed research project consists of three interconnected parts:
(1) study of retention behavior of bioactive macromolecules (proteins, peptides) in ultrahigh performance liquid chromatography,
(2) development of data management algorithms for the improving chromatographic resolution and detectability of compounds,
(3) development of ion chromatographic methods for the separation of anionic biomolecules.

The aim of the work is the support of further development of bioanalytical liquid chromatography by investigating important fundamental aspects and developing data management algorithms. The work of the proposed research project will:
- reveal and discuss the most important equilibrium and kinetic processes influencing the separation of bioactive macromolecules,
- serve with high performance methods and algorithm for the study of retention behavior in high performance liquid chromatography,
- give an insight to the retention behavior of small organic and inorganic anions on anion exchangers through the study of adsorption isotherm,
- deliver a selective and efficient capacity gradient method for the analysis of carboxylic acids,
- produce efficient data management algorithms for peak recognition and resolution of overlapping peaks.

What is the significance of the research?

Modern environmental and biochemical research (e.g. proteomics, metabolomics, lipodomics) is inherently complicated by the tremendous complexity of the samples involved in these studies. Current HPLC technologies need further development to analyze all the compounds in these samples. Development of high performance liquid chromatographic methods that allows the separation of complex biological and environmental samples are indispensable for future biochemical, pharmaceutical and environmental research. The results of the proposed research project will support development of bioanalytical liquid chromatography by investigating important fundamental aspects and developing data management algorithms. Besides the theoretical importances, the results of investigation of retention behavior of bioactive macromolecules can be used directly for the development and optimization of analysis of these compounds. The results will give practical suggestions for the analyst regarding the optimal separation conditions and methods. Successful application of these phases can improve the quality of analysis of peptides and proteins, providing additional information on biological/biochemical processes and/or quality assurance of development and production of biopharmaceuticals. The data management algorithms to be developed in the work will significantly improve the quality of analytical results obtained by high performance liquid chromatography.

The main strength of the proposed project is that is involves different approaches and fields of scientific research (separation science, thermodynamics, mathematical modeling, chemometrics and scientific computation) for the successful evaluation of the project.

In modern biochemical and environmental anylsis, the analysts must determine the composition of samples consisting numerous components. This is a challenging task. High performance liquid chromatographic methods can offer a separation power for the analysis of complex mixture. Further development of efficient analytical methods, however, requires the understanding of the processes take place in these systems. The goals of the proposed research project is to investigate fundamental and practical aspects of high performance separation of biochemically active macromolecules and ionizable components (proteins, peptides, protein fragments, carboxylic acids, complexes, etc.). The proposed research project consists of three interconnected parts:
(1) study of retention behavior of bioactive macromolecules (proteins, peptides) in ultrahigh performance liquid chromatography,
(2) development of data management algorithms for the improving chromatographic resolution and detectability of compounds,
(3) development of ion chromatographic methods for the separation of anionic biomolecules.

A projekt megvalósítása során kapott eredményeinkkel bioanalitikai célú folyadékkromatográfiás fejlesztéseket támogatunk alapvető folyadékkromatográfiás elméleti problémák vizsgálata, újszerű állófázis struktúrák tervezése és optimálása, valamint adatfeldolgozó algoritmusok fejlesztése révén. Vizsgáltuk monoklonális antitestek kationcserés elválasztási lehetőségeit szimultán pH és sógradiens alkalmazásával, különböző kationcserélő állófázisok és tömegskeptrometriás detektálás mellett is alkalmazható pufferrendszerek hatékonyságát, valamint az ioncserélő oszlopban kialakuló pH profilokat. Elméleti megközelítések segítségével oszlopkapcsolási stratégiák, gradiens állófázisok (szemcseméret, ioncsre-kapacitás) és többrétegű állófázis struktúrák folyadékkromatográfiás alkalmazhatóságtóságát vizsgáltuk. Megállapítottuk az optimális alkalmazhatóság körülményeit. Módszert fejlesztettünk komponensek izotermájának, a mozgófázis oszlopbeli állapotjellemzőinek meghatározására, valamint átlapoló csúcsok numerikus (in silico) felbontására. Viszgáltuk az oszlopban kialakuló nyomságradiens elválasztás hatékonyságára gyakorolt hatását. Analtikai módszereket fejleszettünk karbonsavak, gyógyszerhatóanyagok (cilostazol, apixaban) és szennyezőik (nátrium-azid, N-brómszukcinimid) meghatározására.
Results in English
Through the results obtained during the implementation of the project, we support the development of liquid chromatography methods for bioanalytical purposes by investigating fundamental theoretical problems of liquid chromatography, designing and optimizing novel stationary phases, and developing data processing algorithms. We studied the cation-exchange separation of monoclonal antibodies using simultaneous pH and salt gradient, the efficiency of MS compatible buffer systems with different cation-exchange stationary phases, and the developed pH profiles in ion-exchange columns. Theoretical approaches were used to investigate the applicability of column coupling strategies, gradient stationary phases (particle size, ion-exchange capacity) and multilayer stationary phase structures for liquid chromatography. The conditions for optimal applicability were established. Methods have been developed for the determination of component isotherms, state properties of mobile phase in the column, and numerical (in silico) resolution of overlapping peaks. The effect of the pressure gradient in the column on the separation efficiency was studied. Analytical methods have been developed for the determination of carboxylic acids, pharmacologically active substances (cilostazol, apixaban) and impurities (sodium azide, N-bromosuccinimide).
