Biotechnológiai kutatások során elengedhetetlen a sejttenyésztési módszerek állandó fejlesztése. Projectünk során egy eljárást dolgoztunk ki arra, hogy a tenyészetben a sejtek ne monolayer formában (2D), hanem hasonló módon, mint a szervezetben, 3 dimenzióban helyezkedjenek el. Ehhez magas víztartalmú, szilikon-jellegű géleket állítottunk elő. A géleket „építőkocka-jelleggel” többféle alkotórészből állítottuk elő, ezáltal a szerkezete könnyen variálható. Megállapítottuk, hogy a tenyészetbeli sejtek nemcsak „belakták” a gélt, de a tumoros sejtek több szempontból is jobban hasonlítottak a szervezetben található daganatokhoz, mint a 2D tenyészetek esetén (gyógyszerrezisztencia, aggregátumok képzése). A gélek használata nagyon egyszerű, a korábbi 2D eljárásokba gyakorlatilag módosítás nélkül beépíthető. A gélek fejlesztése érdekében aggregációra (gélképzésre) hajlamos peptideket is készítettünk. Ezekkel az oldhatatlan anyagokkal nehéz dolgozni, ezért kidolgoztunk egy praktikus módszert ilyen -rosszul oldódó- peptidek előállítására. A módszer a peptidkémia más területein is hasznos lehet. Az aggregálódó peptidek segítségével a szilikon-hidrogélek tulajdonságai jól finomhangolhatóak. A 3D sejttenyészetekről készült mikroszkópi képek kiértékelésére fejlesztettünk egy önálló tanulásra képes (AI-alapú) programot.

In biotechnological research, continuous development of cell culture methods are essential. In our project, we have developed a procedure to arrange cells in the culture not in a monolayer (2D), but in a manner similar to “real-life”, in three dimensions. To achieve this, we produced high-water-content, silicone-like gels. These gels were created from multiple building blocks, allowing for easy variation in their structure. We observed that the cells in the culture not only "inhabited" the gel, but the tumor cells, in several aspects, more closely resembled tumors found in the organism compared to 2D cultures (drug resistance, formation of aggregates). The use of these gels is very straightforward and can be incorporated into previous 2D procedures with virtually no modification. For the development of the gels, we also prepared peptides prone to aggregation (gel formation). Working with these insoluble materials can be challenging, thus we developed a practical method for producing such poorly soluble peptides. This method may also be useful in other areas of peptide chemistry. The properties of the silicone hydrogels can be finely tuned with the help of aggregating peptides. We have developed a self-learning (AI-based) program for the evaluation of microscopy images of the 3D cell cultures.
