A génexpresszió minőségbiztosítási (quality control) rendszerei biztosítják, hogy csak hibátlan mRNS-ek transzlálódhassanak, és megakadályozzák a hibás fehérjék felhalmozódását. Bár az állati minőségbiztosítási rendszerekről nagyon sokat tudunk, a növényi quality control rendszerekről kevés információnk van.
Programunk során a különböző növényi RNS- és fehérje szintű minőségbiztosítási rendszerek kapcsolatát vizsgáltuk. Kimutattuk, hogy növényekben legalább négy poszt-transzkripciós RNS quality control működik, a kétszálú, illetve cap vagy polyA nélküli hibás mRNS-ek elbontásáért felelős RNS silencing, a korai stop kodont tartalmazó hibás mRNS-ek degradáláért felelős NMD, a stop kodon nélküli aberráns transzkriptumok lebontását végző NSD, illetve a transzláció elongációját gátló hibákat hordozó mRNS-ek elimináláért felelős NGD rendszer. Munkánk során azonosítottuk a növényi NGD cis és transz faktorait, és feltártuk, hogy miként bontja el az NGD a hibás mRNS-eket. Bemutattuk, hogy a növényi NSD és NGD szorosan kapcsoltak, és együttműködnek az RNS silencing rendszerrel is, de nem kapcsolódnak az NMD-hez. Kimutattuk, hogy az NGD szorosan kapcsolódik egy növényekben eddig nem ismert fehérje minőségbiztosítási rendszerhez, az RQC (ribosome associated quality control) rendszerhez. Az RQC az elakadó riboszómán található nascens fehérjéket bontja le. Programunk során számos új ismeretet szereztünk a növényi minőségbiztosítási rendszerek működéséről, kapcsolatáról szerepéről.

Quality control systems ensure the fidelity of gene expression, they inhibit the translation of aberrant transcripts and prevent the accumulation of faulty proteins. Although the quality control systems have been intensively studied in animals and yeast, plant quality control systems are still poorly understood.
During the program we have studied the connection between various RNA and protein quality control systems. We demonstrated that at least four RNA quality control systems function in plants, the RNA silencing system, which eliminates double-stranded aberrant transcript or mRNAs lacking cap or polyA, the NMD system, which decays premature termination codon containing aberrant mRNAs, the NSD system, which degrades stop codon less faulty transcript and the NGD system, which eliminates mRNAs containing translation elongation blocking elements. We identified the cis and trans factors of NGD and unravelled the molecular mechanism of NGD. We have shown that plant NSD and NGD are strictly connected and they also cooperate with silencing but not with NMD quality control system. We have also revealed that the RQC (ribosome associated quality control) protein quality control system functions in plants, identified the components of it, and demonstrated that RQC is mechanistically connected to NGD. Taken together, our results significantly contributed to our knowledge about the mechanism, connections and functions of different quality control systems in plant.
