A projekt fő célja a halak ivarát szabályozó genetikai és környezeti tényezők vizsgálata volt a genomika eszközeivel. Elsősorban a zebradániót használtuk modellként kutatásainkhoz, de más modell és haszon halfajokat is vizsgáltunk. A zebradánió ivarának vizsgálatához két mutáns vonalat használtunk, melyek segítették az ivarszerv kialakulásának jobb megértését. Kifejlesztettünk egy olyan transzgénikus jelzővonalat is, melyben a test csökkent pigmentációja miatt az ivari különbségeket jobban megfigyelhetjük. Egy speciális eljárást is kidolgoztunk, mellyel az ivarszervek kialakulását nagyszámú élő egyeden egyszerre tudtuk követni. Újra megvizsgáltuk a női egyedek hővel végzett átszexálását, hogy tovább optimalizáljuk az eljárást és hőérzékeny családokat azonosítsunk. A genomban kódolt és expresszált ivari markereket azonosítottunk két halfajból. Részt vettünk olyan kutatásokban is, melyek haszonhal fajok szaporodásáról ill. genomjáról bővítették ismereteinket. Sikerrel alkalmaztunk egy hőkezeléses eljárást kopoltyúféreg parazitával fertőzött lesőharcsa egyedek megmentésére. A kezelés jelentősen csökkentette a fertőzött kopoltyúk parazita terhelését a kezeletlen egyedekéhez képest. A hőkezelt és kezeletlen egyedek kopoltyújában és lépében kifejeződő génkészletek összehasonlítása révén több, mint 600 olyan gént azonosítottunk, amelyek expressziójának mértéke statisztikailag szignifikáns különbséget mutatott.

This project aimed to achieve better understanding of the genetic and environmental regulators of fish sex using the tools of genomics. We used the zebrafish as a model for our studies, but worked on several other model and food fish species as well. For the analysis of zebrafish sex, we have utilized two mutants, that helped us to gain better understanding of the gonad differentiation of the species. We have developed an improved transgenic reporter line with reduced pigmentation of the body to allow for better detection of differences between the two sexes. We have also worked out a special protocol that allowed us to follow gonad differentiation processes in vivo on an individual basis on a large number of specimens. We have revisited the formerly tested phenomenon of heat-induced masculinization to optimize the procedure and identify heat-sensitive families. We have discovered genomic and expressed sex markers in two fish species. We have taken part in the analysis of several food fish species to deepen our knowledge on the reproduction and/or genome. We have successfully used a heat-based treatment to rescue European catfish individuals infected by a gill monogenean exoparasite. The treatment has substantially reduced the parasite loads on the gills in comparison to untreated, infected controls. Comparative analysis of the heat-treated vs. untreated gills and spleens identified over 600 genes whose expression level changed significantly as a result of the treatment.
