Célunk az FcgRIIb szerint tartalmazó, sejtmembrán-közeli intracelluláris motívuma és a MAPK kölcsönhatásának, valamint az FcgRIIb szerinen történő foszforilációja funkcionális következményeinek felderítése volt. Igazoltuk, hogy aktív MAP kinázok kötődnek az FcγRIIb-n levő dokkoló helyhez, amelyek az aktivált B sejtekben foszforilálhatják magát a receptort és/vagy más, ahhoz kötődő fehérjét. Azonosítottuk azt a legrövidebb IgG Fc szekvenciát, amely FcgRII által kiváltott jelek közvetítésére alkalmas. Megállapítottuk, hogy a szerin/alanin mutácót tartalmazó receptorok fokozott tirozin kináz (lyn) és Erk aktivációt tesznek lehetővé B sejtekben, ami arra utal, hogy a vad típusú receptort hordozó sejtekben ezek a kinázok kisebb mértékben aktiválódnak. Igazoltuk, hogy a sejtbejuttatható FcgRIIb foszfopeptidek alkalmasak a B sejt receptoron keresztül kiváltott jelek pozitív és negatív szabályozására a foszforilált aminosavtól (ser vagy tyr), a dózistól és a kezelési időtől függő módon. Kimutattuk, hogy a MAPK gátlása B sejtekben fokozza az FcgRIIb expressziót, feltehetőleg a receptor leválás megakadályozásán keresztül. Ez arra a enged következtetni, hogy aktivált B sejtekben a MAPK ezzel ellentétes folyamatot, a negatívan szabályozó FcgRIIb leválását vezérli. Eredményeink azt sugallják, hogy mind a szerinen, mind a tirozinon foszforilált FcgRIIb motívum negatív szabályozó szerepet játszik a B sejt stimuláció során.

Our aim was to explore the interaction between the membrane proximal intracellular motif of FcgRIIb and the MAPK in B lymphocytes and to reveal the functional consequences of the phosphorylation of FcgRIIb on serine. We first proved that active MAP kinases bind to a docking site on FcgRIIb, then may phosphorylate the receptor itself and/or any protein that may bind to it. We identified the shortest peptide corresponding to a linear sequence in IgG Fc that binds and induces signals via FcgRII. We described that ser/ala mutant FcgRIIb admit an enhanced protein tyrosine kinase (lyn) and Erk activation in B cells, suggesting that these kinases become less activated by BCR signals in cells expressing the wild type FcgRIIb . We have shown that cellmembrane permeable FcgRIIb phosphopeptides may positively and negatively regulate signals in B cells, depending on the kind of amino acid residue (ser or tyr), the dose and time of interaction. We also pointed out that the inhibition of MAPK in B cells enhances FcgRIIb expression, most probably by inhibiting receptor release. These data indicate that activated MAPK may induce an opposite process, namely the shedding of the negatively regulating FcgRIIb from activated B cells. Our results suggest that both ser and tyr phosphorylated motifs of FcgRIIb may downregulate antigen induced signals mediated via BCR.
