Elsőként bizonyítottuk, hogy a Fru-2,6-P2 szint csökkentésével és növelésével is lehet agronómiailag értékelhető és reprodukálható fenotípusos eltéréseket létrehozni. Demonstráltuk, hogy a megnövelt Fru-2,6-P2 szint a növényeknél is adaptív előnyt jelent szárazság- és hidegstressz esetén az állati rendszerhez hasonlóan és ez a foszfoenolpiruvát karboxiláz (PEPC) CO2 refixációs aktivitásán, a Pi-függő fruktóz-6-foszfát kináz (PFP) energiamobilizációban való – növényekben eddig nem ismert – szerepvállalásán és a sztómák vízvesztést minimalizáló működésén alapszik. Igazoltuk, hogy a kontroll szignifikánsan alacsonyabb Fru-2,6-P2 szintje nem váltja ki a fenti folyamatokat. A Fru-2,6-P2 gumóspecifikus megváltoztatása a csirázási paraméterek megváltozását eredményezte hagyományos, sötétben való tárolás esetén, lényegi metabolikus átrendeződés nélkül. A cukormetabolit profilok szacharózt és keményítőt is érintő megváltozását abban az esetben figyeltünk meg, ha a gumókat 3 hétig erős fényben tároltuk. Míg a bezöldült gumók felszín-közeli szövetrészeiben a fotoszintetizáló levelekhez hasonló cukormetabolit profilokat kaptunk, közeledve a gumó belsejéhez ez a hasonlóság fokozatosan eltünt és a metabolitprofilok a kontroll sötétben tartott gumókra jellemző értékeket mutatták. További új eredményeink a PFP növényi stresszválaszban betöltött szerepét világították meg. Adataink szerint a PFP részt vesz a tárolt keményítő stressz-függő remobilizációjában olyan raktározószervekben, mint a burgonyagumó, vagy a sárgarépa répateste.

We were the first to prove that distinct phenotypic differences can be achieved by the up regulation and down regulation of the endogenous Fru-2,6-P2 levels. It has been demonstrated that the increased Fru-2,6-P2 level can lead to an increased adaptive ability in plants to drought and cold stress similarly to animal systems. This enhanced tolerance is based on the capability of phosphoenol pyruvate carboxylase (PEPC) to refix CO2, the involvement of Pi-dependent fructose-6-phosphate kinase (PFP) in the stress-dependent remobilisation of the energy reserves and the waterloss minimazing behavior of the stomatal appertures. It has been proved that the wild type level Fru-2,6-P2 content is unable to initiate the above stress responses. The tuber-specific manipulation of the Fru-2,6-P2 metabolism significantly influenced the sprouting of the tubers when they were stored in darkness, without causing major rearrangements in the metabolite profiles. Such major metabolic rearrangements, involving significant changes in the sucrose and starch contents, were observed when the tubers were stored under strong, continuous illumination for three weeks. Metabolite profiles of the green tubers were nearly identical to that of the leaves when tissue extracts were collected from the subepidermal region. At the same time, tissues from deeper cell layers were more similar to the control tubers. Finally, experimental proofs were provided to show that PFP has an important role in plant stress responses by participating in the remobilisation of the starch reserves in storage organs like potato tuber or carrot taproot.
