Az apoptózis a sejtek elhalásának egyik legintenzívebben tanulmányozott, legjobban ismert formája. A sejthalált indukáló receptorok, köztük az általunk vizsgált Fas receptor, illetve az ezek által kiváltott jelet továbbító molekulák az apoptózis kutatás központi elemei. Míg a sejthalálindukáló komplex kialakulását követő események jól ismertek, a komplex kialakulását előidéző jelátviteli folyamtokról nem rendelkezünk adatokkal. Munkánk során elsősorban az SHP-1 tirozin foszfatáz szerepét kivántuk tisztázni a Fas-indukált sejthalál folyamataiban. Eredményeink szerint B-sejteken az SHP-1 a sejthalál indukáló komplex kialakulását ,valamint a Fas internalizációját gátolja, és ezen folyamatokon keresztül a Fas-kiváltott sejthalál negatív regulátora. FasL stimulációt követően az SHP-1 a Vav defoszforilácójának, továbbá a Fas-ezrin kapcsolat létrejöttének szabályozója, miáltal kulcsfontosságú a Fas-indukciót követő citoszkeleton átrendeződés szempontjából. Az SHP-1 tehát a Fas-közvetített jelátvitel kezdeti lépéseinek szabályozója meghatározva a Fas-citoszkeleton kapcsolat létrejöttét, életidejét. A Fas-közvetített nem apoptotikus szignálok fontossága egyre inkább előtérbe kerül az utobbi években. Kimuttatuk, hogy a Fas recepotr aktivációja proliferációt okozhat B-sejteken, és bizonyítottuk, hogy különböző molekula komplexek kapcsolódnak a Fashoz apoptotikus és nem apoptotikus jelek indukciójakor.

Among apoptotic pathways, the one triggered by the death receptor Fas (CD95/Apo-1/TNFSFR6), a member of the tumor necrosis factor receptor (TNFR) superfamily, upon engagement by its ligand, Fas ligand (FasL), is perhaps one of the most widely studied. Despite the wealth of information on events downstream of death-inducing signaling complex (DISC) formation, little is known about the events necessary to generate and regulate the early formation of this complex. We specifically focused our interest on the role of SHP-1 tyrosine phosphatase in the regulation of Fas-mediated cell death. Our results strongly indicate that in B cells, the tyrosine phosphatase SHP-1 acts as a negative regulator of Fas internalization and subsequently of the DISC formation and cell death signaling. By regulating Vav dephosphorylation as well as Fas-ezrin association upon Fas stimulation, SHP-1 controls the cytoskeletal rearrangement and thereby regulating the earliest steps of Fas-mediated signaling. The Fas-mediated non-apoptotic signals became emphesized in th last years. We provided evidences about the Fas-induced proliferation of B-cells, showing the formation of different Fas-associated molecular complexes upon apoptotic and non apoptotic condition.
