A következő kérdésekre kívánunk választ kapni:
1. Éhezés hűvös vagy semleges környezetben módosítja-e a napszaki ritmusokat (homeotermia megőrzésének kényszere vs hipo- vagy heterotermia egy nokturnális speciesben) ?
2. Az éhezést követő újratáplálás során gyors testhőmérséklet-emelkedés létrejön-e egérben és ha igen, mi a mechanizmusa ?
3. A szabadon mozgó körülmények között együtt vizsgálható hőszabályozási, aktivitási és energetikai paraméterek mennyiben korrelálnak normális (normothermiás) és lázas/hyperthermiás körülmények között ?
4. A cirkadián hőmérsékleti hullámzás kialakulásának anesztézia/műtét után megfigyelhető többnapos késése milyen tényezőkre vezethető vissza (a stressz reakció lehetséges szerepe) ?
5. Capsaicin- vagy resiniferatoxin-kezelt (deszenzibilizált) egerek hőregulációs válaszai különböznek-e a TRPV1-gén hiányos állatok hasonló válaszaitól ?
6. A fiziológiás hipertermia-tartományban működő TRPV3 gén hiánya esetén miképpen károsodik a hőreguláció ?
7. Van-e a fizikai aktivitásnak specifikus hőregulációs (hőtermelő) hatása egérben és ha igen, mi ennek a mechanizmusa ?
8. A hőregulációt és/vagy fizikai aktivitást módosító tényezők miképpen hatnak az energetikai hatásfokra, azaz milyen a testtömeg viselkedése ezekben a helyzetekben ?
9. Pirogének és feltételezett endogén mediátorok többnapos infúziója mennyiben módosítja az energetika vizsgálni kívánt paramétereit, illetve a napszaki, vagy egyéb periódusú ritmusait ?
Summary
The following questions should be answered:
1. Can daily rhythms be modified by fasting carried out in cool or neutral environment (i.e. is there a need to maintain homeothermia or rather hypo-/heterothermia may occur as a compromise) ?
2. Is there a rapid normothermia in mice during refeeding following fasting, if yes, what is its mechanism ?
3. Is there a correlation among parameters of thermoregulation, activity and heat production during normothermia or under conditions of hyperthermia or fever ?
4. What factors may play a role in the delay in the development of body temperature rhythm following anesthesia/laparatomy (a possible role for stress) ?
5. Are there any differences in thermoregulatory responses of capsaicin-/resiniferatoxin-desensitized mice as compared to those lacking TRPV1 gene ?
6. In case of lack of the TRPV3 gene (operating in the physiological hypertermic range) is there any deficiency of thermoregulation ?
7. Has physical activity any specific thermogenetic role in maintaing homeothermia during states of substrate deficiency such as fasting ?
8. What effects are exerted on energy efficiency by interventions modifying heat regulation and/or physical activity, that is, how complex energetics is reflected by changes in body mass ?
9. In what way parameters of energetics are influenced by infusions of pyrogens or by their putative mediators ? Are daily and other physiological rhythms modified by these interventions ?

C57/BL egerek maghőmérsékletét és lokomotor aktivitását vizsgáltuk biotelemetriás módszerrel különböző környezeti hőmérsékleten. Normális és zsírdús táplálás hatásait követtük a napszaki hullámzásokra táplálás, több napos teljes éheztetés és újratáplálás ideje alatt, valamint ismételt laparotomiás beavatkozást követően is. Mindezen változások energetikai hátterének megismerése céljából releváns mediátorok, gátló anyagok centrális vagy perifériás infúzióját végeztük ALZET minipumpa segítségével. Szabadon mozgó egerek napszaki hőmérsékleti és aktivitási hullámzásának nagysága fokozódik teljes éhezés során és a maghőmérséklet 30 °C-ra való csökkenése jelzi a túlélhető éhezés végét az aktivitás fokozódása mellett. Újratápláláskor, hidegben, elhízásban, CNTF infúzió alatt és laparotomia után csökken a hullámzások amplitúdója. CNTF és CCK-8 icv infúziója lázat vált ki. Eredményeink több új információval szolgáltak a szabadon mozgó egerek normális és kóros energetikájának mechanizmusáról.
Results in English
Changes of core temperature and locomotor activity were monitored in C57/BL mice under different thermal conditions by using a biotelemetric method. Effects of normal or fat-rich diet were observed on daily oscillatons during feeding, total fasting, re-feeding or on repeated laparotomy. To learn the energetic background of all these stimuli relevant meditors or blocker were infused either centrally or peripherally by using ALZET minipumps. Excursions of daily changes in core temperature and activity were found to be increased during total fasting with core temperature falling to about 30 °C as a sign of threshold of survivability with locomotor activity increasing progressively. Re-feeding, cold exposure, obesity, CNTF infusion or laparotomy led to reversible decreases of daily oscillations. Icv infusion of CNTF or CCK-8 induced fever. Our results furnished new information on the mechanisms of changes in energetics observed under normal and various types of pathological conditions in freely moving mice.
