A gyakori előfordulás és nagy jelentőség ellenére a depresszió gyógyítása nem megoldott. A stressz szabályozásában központi szerepet játszó hypothalamo-hypophysis-mellékvese tengely (HHM) megváltozott működése az egyik kulcsfontosságú biológiai abnormalitás, mely a depressziósok 30-50%-ában megfigyelhető. Mind az érzelmek, mind a HHM tengely szabályozásában a vazopresszin (AVP) jelentősége alapvető. Az emlős agyban az AVP elsősorban a nucleus paraventricularis hypothalami (PVN) és nucleus supropticus (SON) magnocelluláris sejtjeiben termelődik. Az axonterminálisokból a vérbe ürülő neuropeptid antidiuretikus hormonként viselkedik. Ezen perifériás szerepén túl a magnocelluláris sejtek szómájából/dendritjeiből ürülő neuropeptid autokrin/parakrin módon vesz részt a HHM tengely szabályozásában. Egyre elfogadottabbá válik az is, hogy a hypothalamusból túlzott mértékben felszabaduló AVP elérhet limbikus területeket is, ahol az érzelmek szabályozásában is szerepet játszhat. Nemzetközi együttműködésünk során fel szeretnénk tárni az AVP neuromodulátor szerepét az endokrin stresszválasz és az érzelmi folyamatok összekapcsolásában. Ehhez spontán mutációval AVP hiányossá vált Brattleboro patkányok alap és AVP agonisták különböző agyterületre történő injekcióját követő magatartási és endokrin változásait vizsgálnánk. Eredményeink hozzájárulhatnak a depresszió kifejlődésének mélyebb megértéséhez és új hatásmechanizmusú, az eddigieknél esetleg kedvezőbb hatékonysággal rendelkező gyógyszerek kipróbálásához a depresszió kezelésében.
Summary
The pathological emotional state of patients suffering from mood disorders is frequently paralleled by an altered activity of the hypothalamo-pituitary-adrenal (HPA) axis. Arginine vasopressin (AVP) plays an outstanding role in the control of both emotions and neuroendocrine responses. In the mammalian brain, AVP is predominantly synthesized in magnocellular neurons located in the paraventricular (PVN) and supraoptic nuclei (SON) of the hypothalamus. AVP is released from axon terminals into the blood stream and acts as antidiuretic hormone. In addition to, but independently of this peripheral release, the neuropeptide is also released from somata/dendrites of magnocellular neurons in the extracellular fluid where it acts as an autocrine/paracrine signal that affects the HPA axis activity. There is growing evidence suggesting that the AVP released from the hypothalamus in an excessive manner reaches remote limbic brain areas where it interacts with non-synaptically located receptors to modulate emotional states. Here we propose to study the action of AVP as a volume transmitted neuromodulator that links the endocrine stress response with the emotional state of the individual. These experiments involve the use of AVP deficient Brattleboro rats. Administration of AVP agonists into distinct brain areas (including the PVN) by microdialysis will enable us to investigate the behavioral and endocrine consequences of the locally restored neuropeptide signalling. We expect that our study would provide deeper insights into the etiology of mood disorders and may result in the design of new treatments for these diseases.

Napjainkban a vazopresszinnek, mint fontos neurotranszmitter, neuromodulátor és stressz szabályozó molekulának a depresszióban betöltött szerepe intenzív kutatás tárgya. Munkánk során vazopresszin hiányos Brattleboro állatokat használtunk és megállapítottuk, hogy ezen állatok csökkent depresszió-szerű tüneteket mutattak számos magatartási tesztben. Ezzel párhuzamosan azonban a stressztengely (hypothalamo-hipofízis-mellékvese tengely) szabályozásában csak csekély jelentőséget találtunk. Az oxitocin részleges kompenzáló szerepét igazoltuk. Kimutattuk továbbá, hogy a nucleus paraventriculari hypothalami-ból felszabaduló vazopresszin parakrin úton segíti a stressz-stimulálta oxitocin és kortikoszteron plazma-szintek normalizálódását. A felnőttekkel ellentétben perinatálisan a vazopresszin tekinthető az ACTH elválasztás fő szabályozójának. Eredményeink arra utalnak, hogy a vazopresszin a depresszió kialakulásában fontos tényező, de valószínűleg nem a stressz-tengely befolyásolása révén hat, hanem egyéb agyi mechanizmusokat befolyásol.
Results in English
Vasopressin, an ubiquiter hormone is considered to be one of the factors related to the development of depression. In our studies the congenital deficit of vasopressin in Brattleboro rats leads to attenuated depression-like behavior. Because the strong connection between depression and the hypothalamo-pituitary-adrenal axis regulation we examined the role of vasopressin in stress. The vasopressin deficiency had no influence on basal levels of ACTH and corticosterone and had only mild impact on hormonal activation in response to different stressors. The partial compesation by oxytocin elevation was confirmed. Endogenous vasopressin released within the nucleus paraventricularis hypothalami acts as a paracrine signal to facilitate the return of plasma oxytocin and corticosterone to basal levels after acute stressor exposure. In contrast to adult, vasopressin seems to be the predominant ACTH secretagogue during the perinatal period. We suggest that brain vasopressinergic circuits distinct from those regulating the HPA axis are involved in generating depression-like behavior.
