Az élő sejtek szerkezeti és funkcionális integritásának fenntartásában és különböző folyamatok szabályozásában kiemelkedő szerepük van a membránfehérjéknek. Egy-egy membránfehérje szerkezetének, membránbeli szerveződésének és működésének megértése igen nagy, de egyben ígéretes kihívás mind az alapkutatás, mind a gyakorlati felhasználás tekintetében.
Jelen pályázat célja különböző béta-hordó (OMP fehérjék) és transzmembrán alfa-hélix fehérjék (különös tekintettel a V-ATPáz Vo domén c alegységeire) membránba való beépülési mechanizmusának vizsgálata. Az elterjedten használatos biofizikai technikák mellett méréstechnikailag új módszerek alkalmazását is szeretnénk megvalósitani. ''Statikus'' és kinetikai méréseket egyaránt tervezünk, melyek segítségével tanulmányozzuk a fehérjék szerkezetét, stabilitását, a c alegységek oligomerbe szerveződésének sajátságait és a fehérjehajtogatódás mechanizmusát.
A tématerületen szerzett korábbi tapasztalatainkra alapozva, és a különböző módszerek egymást kiegészítő kombinálásának köszönhetően a várható eredmények reményeink szerint lehővé teszik a hajtógatódással kapcsolatos általános törvényszerűségek meghatározását és ezen folyamatok jobb megértését, a vizsgált fehérjék (különösen a V-ATPáz) működési mechanizmusának és a lipid-fehérje kölcsönhatásoknak az alaposabb megismerését.
Summary
Membrane proteins are crucial components of living cells. They preserve the structural and functional integrity of cell organelles and regulate a lot of processes. The understanding of their structure, function and assembly in the membrane is a great and promising challenge regarding both basic science and practical aspects.
Our goal in the present application is the study of different beta-barrel (OMP proteins) and transmembrane alpha-helix (especially the Vo c subunits of V-ATPase) proteins reconstituted mostly in model membranes. Beyond widely used biophysical techniques, we would like to establish and use novel experimental methods. Performing both static and kinetic measurements we will study the structure, stability, folding of the proteins and the organization of the V-ATPase c subunits. Based on our previous experience on this field, and due to the combination of different techniques that complement each other, the expected results will allow us: to determine some general principles in the field of protein folding; to get further details regarding the studied proteins and the mechanism of their activity (e.g. the V-ATPase proton-pumping mechanism); and to understand specific lipid-protein interactions not only in the studied systems, but in general as well.

A pályázatban célul tűztük ki bakteriális béta-hordó és transzmembrán alfa-hélix fehérjék – különös tekintettel a V-ATPáz c alegységére – vizsgálatát. A natív c alegység hiányában a fehérje transzmembrán szegmenseivel analóg polipeptideket építettünk be lipidmembránokba, és spektroszkópiai módszerekkel vizsgáltuk szerkezetüket, a membránba épülésüket, valamint az egymással, lipidekkel és inhibitorokkal való kölcsönhatásukat. Kimutattuk, hogy a transzmembrán polipeptidek döntően alfa-hélix konformációt vesznek fel a membránban, és hogy oligomerizációra hajlamosak, továbbá azt is, hogy a c elegység negyedik hélixe (TM4) és az a alegység hetedik hélixe (TM7) között kölcsönhatás van. Megmutattuk azt is, hogy a negatívan töltött lipidek kölcsönhatása erőteljesebb a negyedik hélixszel, és hogy bizonyos V-ATPáz inhibitorok kölcsönhatnak a TM4 és a TM7 hélixekkel. A béta-hordó fehérjék közül a Fusobacterium nucleatum-ból izolált FomA fehérjét tanulmányoztuk. Meghatároztuk a fehérje termikus stabilitását, és a kitekeredéssel járó hőváltozás mértékét, továbbá azt, hogy denaturáló szerek milyen mértékben változtatják meg ezeket a paramétereket. Fluoreszcencia spektroszkópiával is követtük a hőmérséklet és a denaturáló szerek hatását. Inkubációs mérésekből információt nyertünk a fehérje kitekeredésének sebességével kapcsolatban, denaturáló szerek segítségével. Beállítottunk egy új mérési elrendezést, mellyel egy mintában egyidejűleg detektálhatók az ESR és fluoreszcencia jelek.
Results in English
In this project our goal was to study bacterial beta-barrel and transmembrane alpha-helix proteins (especially the c subunit of V-ATPase). Since the native c subunit was not available, we studied synthetic polipeptides corresponding to the transmembrane domains of the protein. Spectroscopic methods were used to characterize the structure, self-assembly and molecular interactions (lipid-peptide, peptide-peptide, peptide-inhibitor) of the peptides. We showed that the peptides are highly alpha-helical in the membranes, and that they tend to form oligomers. Moreover, an interaction between the fourth helix of the c subunit (TM4) and the seventh helix of the a subunit (TM7) was proven. Our data revealed stronger interaction of the TM4 peptide with negatively charged lipids, and also that certain V-ATPase inhibitors interact with the TM4 and TM7 peptides. Among from beta-barrel proteins FomA, isolated from Fusobacterium nucleatum, was studied. We determined the thermal stability of the protein, the heat of transition related to the unfolding process, and the effects of denaturing agents on these parameters. Temperature-induced changes and the effects of denaturants were followed by fluorescence spectroscopy as well. Incubation experiments were carried out with the help of denaturants to get information concerning the speed of the unfolding process. A novel experimental setup, capable of simultaneous detection of ESR and fluorescence signals in a sample, was also developed.
