A növények számos védekezési rendszert fejlesztettek ki a mikroorganizmusok elleni védekezés számára. Ezek közül az általános védekezés (BR) alapvető szerepét csak az utóbbi időszakban ismerték fel. E fontos rezisztencia mechanizmus funkciója minden olyan behatoló felismerése és gátlása, amely a növény tápanyagokban gazdag sejtközötti járataiba jut. Kutatócsoportunk az elsők között foglalkozott a baktériumok ellen kialakuló növényi BR folyamatok leírásával. Korábbi OTKA pályázataink keretében többek között „chip” technológia használatával több száz olyan gént azonosítottunk, melyek aktivitásváltozást mutattak a BR kialakulása alatt. Jelen pályázatunkban ezek közül a proteinek lebontásában vagy hasításában szerepet játszó gének részletesebb vizsgálatát szeretnénk elvégezni. A védekezésben a proteinázoknak a szignál létrehozásától kezdve, a szabályozásig és a patogének egyes virulencia faktorainak módosításán keresztüli közvetlen hatásán át, számos funkciója lehet. Tervezett kutatásaink első lépcsőjében a proteináz inhibitorok hatását vizsgáljuk a BR-el kapcsolatba hozható folyamatokra. Majd a korábban azonosított megváltozott aktivitást mutatott gének transzkripcióját vizsgáljuk meg részletesebben. Ezután egyes gének szerepét szeretnénk meghatározni a növényben történő túltermeltetésével, illetve a gén kifejeződésének csökkentésével illetve Arabidopsis mutánsok bevonásával. Azokat a proteinázokat, amelyekről a fentebb felsorolt kísérletek alapján azt találjuk, hogy szerepük lehet a BR kialakulásában, részletesebb vizsgálatoknak vetnénk alá (pl. szubsztrátspecifitás vizsgálat, lokalizáció). Ezen kívül irányított mutációval meg szeretnénk állapítani a proteináz aktivitás szükségességét a növényben kialakuló hatás szempontjából; Nagyszámú gén, vizsgálatokba történő bevonásával feltehetőleg találunk majd olyat vagy olyanokat, melyek részt vesznek valamilyen formában a BR kialakításában. A pályázatot három-négy kutató bevonásával [posztdoktorok, illetve doktorandusz(ok)] szeretnénk elvégezni. A kutatás elsősorban alapkutatás jellegű, de a védekezési folyamat részletesebb megismerése a későbbiekben felhasználható lehet a termesztett növények védekezési reakciójának fokozásához.
Summary
Plants developed several defense systems for resistance against microorganisms. The crucial role of basal resistance (BR) has only recently been recognized. The function of this important resistance mechanism is to inhibit all foreign organisms that enter into the intercellular spaces of plants. Our research group was one of the first who investigated the development of BR against bacteria. In our former OTKA projects we have identified hundreds of genes that showed activity changes during BR using chip methods. In our proposal we would like investigate in detail the role of the genes that are involved in protein degradation and cleavage in plants. Protease and protein degradation systems may have various functions in plant defense. They may be involved in the initial signal production, in the regulation of transcription and they may affect pathogens by modifying their virulence determinants. In our proposal, we will study the effect of different proteinase inhibitors and their combinations on BR related processes. We will also investigate in detail the transcription of previously identified protease genes. Then we would like to determine the role of the particular genes with the methods of over expressions and silencing. We also plan to study Arabidopsis plants that are mutant in protease genes. With those protease genes that show functions based on the above experiments we will investigate further (e.g. substrate specificity, localization). In addition, with directed mutagenesis we would like to determine the necessity of proteinase activity for the effect of the gene on BR. Investigating a high number of genes will increase the possibility to find those proteases that are involved in BR development. The research will be carried out with the involvement of thee or four persons (two post docs and one or two Ph.D. students). The research in the proposal is mainly basic research but the study of BR in detail may help to increase the resistance of crops in the future.

A növények kórokozók elleni védekezési rendszerének egyik első eleme az ún. általános rezisztencia (BR), melyet a mikroorganizmusok általánosan előforduló molekulái indukálnak. A BR kialakulásáról és a védekezés mechanizmusáról még kevés ismeret áll rendelkezésre. A pályázatunkban a protein anyagcsere és főleg a protein lebontás szerepét vizsgáltuk a BR kialakulása során, mely egyaránt részt vehet a növényi szignál folyamatok kialakulásában és a védekezési válaszreakciók létrejöttében is. Kísérleteinkben többek között a protein anyagcserében résztvevő gének kifejeződését és enzimaktivitás változását követtük. Továbbá egyes proteináz típusokra specifikus inhibitorok segítségével tanulmányoztuk a protein lebontás gátlásának következményét a BR, illetve annak marker folyamataira (pl. védekezési gének átíródása, kallóz beépülés stb.). Ezeken a kísérleteknek eredményei a cisztein és a szerin típusú proteinázok részvételét mutatták a BR kialakulása során. Inhibitorokkal kombinált, microarray módszerrel elvégzett transzkriptoma vizsgálatokkal kapcsolatokat sikerült kimutatni egyes jelátviteli utak és a proteoszómás lebontás között a BR létrejötte alatt. Proteináz génekben mutáns és túltermelő Arabidopsis növények, és protein lebontás és szintézis génekben csendesített Nicotiana benthamiana növények segítségével BR kialakulásában részt vevő szubtiláz, karboxipepetidáz, cisztein proteináz és proteaszóma géneket azonosítottunk.

Basal resistance (BR) is one of the first lines of plant defense against pathogens including bacteria. The elicitors of the BR are common microorganism derived molecules. The development and the mechanism of defense of BR are not well understood yet. In our project we focused on protein degradation processes during BR because it is an important element of defense, since it can be involved both in signal transduction and resistance response of plant cells. Pharmacological and functional genomic methods were used to find protease activity and protease genes that are important for BR. Among others, we measured the expression of protein metabolism genes and the protease activity changes during BR. In addition, using specific protease inhibitors we investigated the consequence of protease blocking on BR markers (e.g. resistance gene expressions and callose deposition). The results of these experiments strongly suggested the important role of serine and cysteine type poteases in BR. Transcriptomic microarray study using different inhibitors found connection between some signal transduction pathways and proteasome type protein degradation. Screening protease mutant and protease overexpressing Arabidopsis thaliana plants moreover protease gene silenced Nicotiana benthamiana resulted in identification of subtilase, carboxypeptidase and cysteine protease genes as well as genes of proteasome degradation system as elements of BR.
