Az emberi genomban tárolt információ kifejeződését transzkripciós faktorok biztosítják. A magreceptorok olyan transzkripciós faktorok, melyek aktivitását lipidoldékony molekulák szabályozzák. Az emberi genom megismerése lehetővé tette a fehérje kódoló gének kifejeződésének a vizsgálatát, de meglehetősen keveset tudunk a szabályozó régiók szerveződéséről és a cisz ható elemekről, különösen az egész genom kontextusában gondolkodva.
Egy olyan kísérletsorozatot tervezünk végrehajtani, amelynek során, elsősorban kromatin immunprecipitációs módszerekkel megvizsgáljuk, hogy hova kötődnek a genomban a PPARg, RARa, VDR es RXR receptorok egy normál primer emberi immunsejtben, a dendrititkus sejtekben. Ez a pályázat szerves folytatása az eddig elvégzett munkánknak, amelyben szisztematikusan feltérképeztük azokat az mRNS szintű változásokat, amelyeket a receptor aktiválása okoz ebben a sejttípusban. Az elvégzett vizsgálatok során bebizonyosodott, hogy a dendritikus sejtek kiválóan alkalmasak magreceptorok által szabályozott útvonalak térképezésére. Meg fogjuk határozni a hiszton módosításokat, DNS metilációt, DNáz hiperszenzitivást valamint az RNS pol II és receptor kötőhelyeket. Ezeket a kísérleteket először a genom egy kisebb szakaszán a CD1 klaszteren végezzük el majd kiterjesztjük a teljes genomra. A nyert adatok fontos új ismereteket adnak a receptorok működéséről és arról is, hogy hogyan történik a gének aktiválása illetve gátlása a teljes genom szintjén. Ezeket az adatokat kiegészítjük még olyan potenciálisan klinikailag is releváns informaciókkal mint SNPk megléte a szabályozó régiókban.
Summary
The expression of the information coded in the human genome is mediated via the activity of transcription factors. Nuclear hormone receptors are regulators of transcription in vertebrates in response to fatty ligand. The completion of the human genome has allowed the identification of expressed regions of protein coding genes, however, very little is known on the organization and distribution of their regulatory regions and cis-acting elements especially in the context of the entire genome.
We are proposing to carry out a series of unbiased sequence interrogation studies using principally chromatin immunprecipitation, of the genuine chromatin binding sites of nuclear hormone receptors PPARg, RARa, VDR and RXRa in primary human immune cells (dendritic cells). This effort is a logical continuation and extension of our previous work in which we have systematically analyzed the transcriptional changes at the mRNA level brought about by activation of these receptors. Those studies have established human primary dendritic cells as a cell type suitable for analyses of complex regulatory circuits coordinated by nuclear receptors and lipid signaling. The proposed mapping of histone modifications, DNA methylation, DNAse hypersensitivity along with RNA pol II binding and receptor occupancy will be carried out first on the cluster of CD1 genes on chromosome 1 as a model and later extended to the whole genome. This should provide us with a rich and unique source of data on the function of the receptors in a normal human cell type. In addition mechanistic insights into the regulatory logic of nuclear receptor circuits, transcriptional activation and repression will be gained along with clinically relevant information on SNPs localized to binding and regulatory regions.

A kutatómunka során szisztematikusan feltérképeztük az un RXR heterodimer típusú receptorok szerepét a humán monocita eredetű dendritikus sejtek differenciálódása és immunfunkciói során. Ezek a receptorok a PPARg, LXR, VDR és RAR receptorok volta, melyeket kiegészítettünk az RXR receptorral is. A specikus utvonalak azonosítása mellett általános megállapításként elmondhattuk, hogy ez a receptorcsalad olyan molekuláris szenzorként működik, mely képes átprogramozni a sejt genkifejeződését külső és belső lipid szintek változása során. Összefüggést talaltunk az IL4 STAT6-on kerestüli szignalizációja és a PPARg receptor között és legújabban feltérképeztük a az RXR receptor genomi kötőhelyeit ls cisztromikus kölcsönhatásait is.
Results in English
During our work we have systematically mapped the so called RXR heterodimeric receptors roles in monocyte derived dendritic cells during differentiation and immune function. These receptors included PPARg, LXR, VDR and RXR and also included their heterodimeric partner RXR. Based on the data obtained one can conclude that besides the specialized pathways identified, these receptors act as molecular sensors in detecting changing extra and intracellular lipid levels. We have also identified an interaction between IL4 mediated STAT6 signaling and PPARg and most recently we have detrained the RXR cistrome and its interactions.
