Korábbi kísérleteinkben elsőként mutattuk ki, hogy a hidratálódott pollenszemek NAD(P)H oxidázaik révén reaktív oxigéngyökök képzésére képesek. Ha gátoljuk a pollen eredetű reaktív oxigéngyökök termelődését, drámai módon csökken az allergiás gyulladás a pollen kivonatokkal kezelt egerek légútjaiban. Ez arra utal, hogy a pollen eredetű NAD(P)H oxidázoknak alapvető szerepe lehet az allergiás reakciók kialakulásában. Eredményeink alapján a pollen eredetű allergiás gyulladások kialakulásához a pollen eredetű antigének és a pollen NAD(P)H oxidázok által a hámsejtekben kiváltott oxidatív stressz egyidejű jelenléte szükséges. Eddigi kísérleteinket, amelyekben a pollen NAD(P)H oxidázok hatását vizsgáltuk az allergiás reakciók kialakulásában, előzetesen szenzibilizált egereken végeztük. Az OTKA pályázat támogatásával azt szeretnénk tanulmányozni, hogy a hidratálódott pollenszemek által termelt reaktív oxigéngyököknek van-e szerepe az allergiás reakciók kialakulásának legelső szakaszában, azaz szenzibilizálódás folyamatában. A tervezett kísérleteinkben tehát arra a kérdésre keressük a választ, hogy a pollenszem expozíció képes e aktiválni a dendritikus sejteket, illetve az atópiás és nem atópiás egyénekből származó dendritikus sejtek között van-e fenotípusos, illetve funkcionális különbség.
Summary
We have recently discovered that hydrated pollen grains are able to generate reactive oxygen species (ROS) due to NAD(P)H oxidase activity in them. Removal of ROS producing activity from the pollen extracts dramatically reduces the allergic airway inflammation in a mouse model of allergic asthma. This phenomenon indicates that pollen grains’ NAD(P)H oxidases have essential role in the development of allergic responses. We propose that oxidative stress in the epithelial cells generated by pollen NAD(P)H oxidases augments allergic airway inflammation induced by pollen antigen. All our experiments related to this novel discovery were performed in sensitized mice. In these new series of experiments we will investigate the role of ROS generated by hydrated pollen grains in the initial phase of the development of allergic reactions (it is called sensitization). We try to answer the question whether pollen-generated ROS can activate dendritic cells and whether there are any phenotypic or functional differentiations between dendritic cells derived from atopic or non-atopic individuals.

Eredményeink szerint a hidratálódott pollenszemek által termelt reaktív oxigéngyököknek fontos szerepe van az allergiás reakciók kialakulásának legelső szakaszában (azaz a szenzibilizálódásban). Ezek a rendkívül reakcióképes gyökök gyulladást elősegítő citokinek termelődését váltják ki a myeloid dendritikus sejtekből, ezzel hozzájárulnak a lokális veleszületett immunválaszok kialakulásához, valamint adjuváns faktorként segítik a pollen antigének elleni szerzett immunitás kialakulását is. Ezzel szemben az oxidative stressznek kitett plazmacitoid dendritikus sejteknek inkább gyulladást gátló, vagy tolerogén hatása van az adaptív immunválaszok szabályozásában. Eredményeink arra is utalnak, hogy a pollenek által kiváltott oxidative stress megelőzésének, vagy csökkentésének a légutakban jótékony hatása lehet az allergiás tünetek kezelésében.
Results in English
Based on our results reactive oxygen species generated by hydrated pollen grains have an important role in the initial phase of the development of allergic reactions (it is called sensitization). These highly reactive radicals trigger pro-inflammatory cytokine production from myeloid DCs contributing to local innate immunity and also act as an adjuvant factor in the initiation of adaptive immune responses against pollen antigens. In contrast, plasmacytoid dendritic cells exposed to oxidative stress have an anti-inflammatory or tolerogenic role in regulating adaptive immune responses. Our data also imply that prevention or reduction of pollen-induced oxidative damage in the airways may have a beneficial role in therapy of allergic symptoms.
