A Sirtuinok (Sirt enzimek) mennyisége és aktivitása meghatározza a sejtek osztódó képességét, ellenálló képességét, anyagcseréjét, a DNS javítását és öregedés folyamatát. A Sirt enzimek energia függőek s szorosan kapcsolódnak a NAD/NADH arányához. Már régóta ismert, hogy a sejtek, szervek illetve állatfajok metabolizmusa meghatározza a felhalmozódó oxidatív sérülés nagyságát, öregedés folyamatát és a maximális életkor hosszát.

Humán vázizomból a fittség (VO2max), életkor és SIRT1, SIRT3, SIRT4, SIRT6, NAMPT, PGC1alpha, UCP3, HIF fehérje és mRNA szintjét vizsgáljuk potenciális biomarkert keresve a vázizom öregedésére. A Sirtuinok számos kulcs fehérje postranszkripciós módosításával befolyásolják a kromatin hiszton ''feszességét'' s ezzel tarnszkripciót, szénhidrát és zsír metabolizmust, mitochondria biogenézisét és DNS sérülés javítását. Ezért potenciális öregedési biomarkerek lehetnek.

Wistar patkányokon végzendő munkáknál a testedzés és az IGF-1 bevitelének hatását vizsgálnánk Wistar patkányokon a sirtuinek által befolyásolt funkcionális és molekáris folyamtokban, de ezeken belül is elsősorban a mitochondria biogenézisében a SIRT1-PGC1-alpha-UCP3-HIF és redox tengelyt feltérképezve. Az IGF-1 alulszabályozás növeli az élettartamot, de az agyban paradox módon az IGF-1 neuroprotektív. Morris Maze és passzív avoidance funkció, neurogenézis és a fenti paraméterek vizsgálatát tervezzük. Az állatok vázizmából a hiszton SIRT1 függő deactylálását, is mérni fogjuk.

Meggyőződésünk szerint a vizsgálatunk eredményei segitenek az öregedés és a testedzés molekuláris folyamatainak jobb megértésében és a sirtuinek szerepének alaposabb megismerésében. Annak ellenére, hogy a fizikai inaktivitás valamennyi életmód-függő betegség rizikófaktora, a fizikai aktivitás vizsgálatát eddig hazánkban nem támogatták. Így marad most is?
Summary
Sirtuins play an important role in cell division, resistance to cope with stressors metabolism, DNA repair capacity and it is associated with aging. Sirtuins are dependent on NAD/NADH ratio and redox state. It is well known that the metabolism of the organs effects rate of free radical generation, oxidative damage and the activity of antioxidant system, which could modify the life-span.

In the current study we would like to measure the maximal oxygen uptake of young, middle aged and aged individuals. We will measure the content and mRNA level of SIRT1, SIRT3, SIRT4, SIRT6, NAMPT, PGC1alpha, UCP3, and HIF to find a biomarker for aging to human skeletal muscle. Sirtuins are effecting the transcription factors by the modification (deacetylation) of histones and the alteration of gene transcription would modify carbohydrate, fat metabolisms, biogenesis of mitochondria, as well as the repair of DNA. Therefore, we believe that some of the sirtuins can be used as a potential biomarker of aging.

We also plan animal study in which we will supplement IGF-1 to young and old Wistar rats since IGF-1 is implicated in aging process (down regulation of IGF-1 system increases the life-span in laboratory animals, while IGF-1 is a neuroptotector). We will use BrdU to see the neurogenesis (which reportedly induced by exercise), and we will focus on the mitochondria biogenesis by PGC-1 alpha, UCP3, HIF alpha system and on the DNA repair. The histone deacetylation will be measured from the skeletal muscle.
We will use some functional marker (VO2max for the whole body and Morris maze and passive avoidance tests for the brain function) since we are interested in the relationship between biochemical and physiological parameters.

We strongly believe that the results of the present study will help to understand better the molecular mechanism of aging process, especially the contribution of sirtuins and we will know whether exercise alter the activity of sirtuins.

A pályázat segítségével sikerült megismerni, hogy a sirtuin fehérjék milyen szerepet játszhatnak a vázizom és az agy öregedésében, valamint kimutattuk, hogy a testedzés öregedés-lassító hatása részben a sirtuin fehérjék szabályozásán keresztül valósul meg. Méréseink azt mutatják, hogy a mitokondriális biogenezis öregedés függő romlását mind az agyban mind a vázizomban lassítja a testedzés által aktivált SIRT1. Kimutattuk, hogy a 8-oxoG mennyiségi növekedését a testedzés lelassíthatja, mely részben az OGG1 nevű javító enzim aktiválásával történik. Mi mutattuk ki először, hogy az OGG1 acetilációja in vivo körülmények között is megjelenik. Kimutattuk, hogy a testedzés okozta neurogenesis független lehet a térbeli memória javulásától. Jelenleg is bírálat alatt álló munkáinkban a rezveratrol gén függő hatásairól számolunk be, ami felveti az "egyéni" medikáció fontosságát is. A pályázat segítségével 12 cikk és 62 impakt faktor született, emellett számos közlemény még bírálat alatt van.
Results in English
The present grant allow us to study the role of sirtuin proteins in aging process and we have shown that indeed, regular exercise activates sirtuins and this could be important in the process by which exercise attenuates aging. We have studied role of sirtuins and exercise in brain and skeletal muscle and found the sirtuins are involved in the biogenesis of mitochondria which decreases with aging, but can be attenuated by exercise training. We have shown the aging increases the levels of 8-oxoG, which can be decreased by exercise due to the fact that exercise activates OGG1, the enzyme which repairs 8-oxoG. Our novel observation revealed that OGG1 is acetylated in vivo. We have further shown that exercise-induced neurogenesis can be independent from the exercise-induced better spatial memory. We have also found that the effects of resveratrol, SIRT1 activating agent, is dependent on genetic, this finding could emphasize the importance of individual medication. By the help of this grant we have published 12 papers, with 62 impact factors, and some results are submitted to publication.
