A vas az élő szervezetek számára nélkülözhetetlen, ezzel egyidejűleg azonban toxikus is. A kelátterápiát akut fémmérgezések kezelésére alkalmazzák elsősorban, azonban napjainkban bebizonyították, hogy a vaskeláló desferrioxamine B megakadályozza atheroszklerotikus léziók kialakulását magas koleszterintartalmú diétán tartott nyulakban, valamint lecsökkenti a léziók vastartalmát. A vaskelálás javítja a koronária artéria betegségben szenvedők endotél funkcióját. Továbbá a vas-elvonás kelátképzők alkalmazásával a daganatok terápiájának új lehetősége. Előzetes eredményeink alapján - miszerint a hidrofób gomba eredetű sziderofórok gátolják az LDL hem mediálta oxidatív modifikációját (mely az érelmeszesedés kulcslépése) valamint orálisan adagolva patkányok emésztőrendszerből felszívódnak – feltételezzük, hogy a vasmentes sziderofórok meggátolják az oxidált LDL által előidézett endotélsejt toxicitást, valamint a simaizomsejtek kalcifikációját, továbbá megakadályozzák ateroszklerotikus léziók kialakulását ApoE deficiens egerekben. Tervezzük a vasmentes sziderofórok anti-proliferatív illetve tumor ellenes hatásának tesztelését hepatocelluláris karcinóma kísérletes modelljeiben. Végül előzetes eredményeink alapján megvizsgálnánk a desferricoprogén alkalmazásának hatékonyságát akut ólommérgezésben.
Summary
Although iron is crucial for life, an excess of iron is toxic. Chelation therapy is used as a treatment for acute toxic metal poisoning. More recently it has been demonstrated that the iron chelating agent desferrioxamine B inhibits atherosclerotic lesion formation in rabbits on cholesterol rich diet, and reduces iron content of plaques. Importantly, iron chelation improves endothelial function in patients with coronary artery disease. Also, depriving cancer cells of the essential nutrient iron (Fe) is a novel approach for cancer treatment. Drawing upon our previous observations that hydrophobic fungal siderophores inhibit heme mediated oxidative modification of LDL – a key event in atherogenesis – and that desferrisiderophores are absorbed in the gastrointestinal tract of rat we propose that desferrisiderophores provide protection against oxidized LDL mediated endothelial cytotoxicity and osteobalstic transformation of vascular smooth muscle cells in vitro and might provide protection against atherosclerotic lesion formation in ApoE mice in vivo. Also we plan to test anti-proliferative and antitumor activity of iron free hydrophobic fungal siderophores in experimental models of hepatocellular carcinoma. Finally based on our preliminary results we propose and plan to test that desferricoprogen is a potential drug for the treatment of acute lead poisoning.

Az érbetegségeken a hem eredetű vas fontos szerepet játszik. Korábbi munkáink a vasculáris betegségek újabb pathomechanizmusát írták le, ami a hem-hemoxigenáz-ferritin út. Mivel a gomba eredetű siderophorok jónéhány táplálékban megtalálhatók, kérdés, a velük történő vasmegkötés befolyásolja-e az érbetegségek progresszióját. Kiválasztottuk a forrásul szolgáló gomba specieseket, optimalizáltuk a tenyésztési körülményeket. Igazoltuk, hogy az akut hem-vas toxikus hatásai endotheliális sejtkultúra rendszerekben e természetes siderophorokkal megelőzhetők. Kutatásunk fő céljainak egyike annak tanulmányozása, hogy vasculáris betegségek esetében a gomba sideroforokkal az egész szervezet számára tudunk-e védelmet biztosítani. A vas-hipotézisnek megfelelően, a vas káros cardiovasculáris rendszer számára. A sclerotikus plakkon belüli vérzést a szabaddá vált hemoglobin oxidációja követi, methemoglobin, ferrylhemoglobin, szabad-hem képződés követi, következményes pro-oxidáns és pro-inflammációs hatásokkal. ApoE deficiens egerekben a hemoglobin stressz, a hemolysis az érelmeszesedés kialakulását felgyorsítja. Ezen egerek természetes siderophorrokkal, mint pl. desferricoprogénnel történő kezelése, szignifikánsan csökkenti a plakk méretet. A desferricoprogen a Camambert- és a Roquefort sajtokban megtalálható. Az érbetegségekben szerepet játszó vas forrásának és lokalizációjának további kutatását munkánk alapján is szükségesnek véljük.
Results in English
Heme derived iron plays an important role in vascular disorders. Our earlier studies raised this new pathomechanism of vascular diseases, which is the heme-heme oxygenase-ferritin pathway. Since fungal siderophores can be found in several natural foods, chelation if iron by these siderophores may have influence on the development of vascular disorders. Fungal organisms are selected as the sources for our studies. Using the methodologies of biotechnology, we describe the optimal culture conditions. Our studies demonstrate that toxic effects of acute heme iron can be prevented by natural siderophores, especially in vascular endothelial cell culture assays. One of the main goals of this research is whether these fungal siderophores provide protection against vascular disorders in whole organisms. According to the iron hypothesis, iron is believed to be detrimental for the cardiovascular system. Following intraplaque hemorrhage free hemoglobin is prone to oxidation forming methemoglobin, ferrylhemoglobin, free heme, and these metabolites exert pro-oxidant, pro-inflammatory effects. In apoE deficient mice hemoglobin stress or hemolysis accelerates atherosclerosis. Treatment of apoE deficient mice with natural siderophores, i.e. desferricoprogen found in Camabert- and Roquefort-type cheeses, significantly reduces plaque size. There is a need for more investigations in which issues like the source and the compartmentalization of iron in vascular disorders would be addressed.
