A pályázat célja a neuroszteroidoknak az idegrendszer öregedéssel kapcsolatos változásaiban játszott szerepének tanulmányozása és olyan terápiás lehetőségek kiderítése, melyek lehetőséget adnak az időskori kognitív problémák kezelésére. A munkaterv végrehajtása interdiciplináris megközelítést igényel, melynek során finomszerkezeti, elektrofiziológiai, viselkedési, valamint DNS chip módszereket alkalmazunk.
Munkahipotézisünk felállításakor az idegtudomány legújabb eredményeiből indultunk ki, amely szerint a központi idegrendszer korral kapcsolatos működési zavarai reverzibilisek és az idősödő agy regenerálódó képessége megőrizhető. Az idegi plaszticitás egyik fontos eleme az idegi kapcsolatok megváltozása, az un. szinapszis átrendeződés – ez a jelenség fontos tényezője a regenerációs kapacitásnak. Ugyanakkor irodalmi adatok és saját vizsgálataink azt is igazolták, hogy számos egyéb tényező mellett a nemi hormonok is képesek morfológiai szinaptikus plaszticitást indukálni és a közelmúltban azt is kimutattuk, hogy egy neuroszteroid, a dehidroepiandroszteron (DHEA) is hasonló hatású.
Kísérleteinkkel a következő feltételezéseinket kívánjuk igazolni:
i.) A hippocampális tüske szinapszisok átrendeződése és a kognitív funkciók összefüggenek, a szinapszisok számának változása (csökkenése) szerepet játszik az időskori memória-romlásban.
ii.) Nemi hormonok ill. DHEA képesek a korral összefüggő morfológiai (tüske szinapszisok számának csökkenése), kognitív (memóriaromlás) és elektrofiziológiai (LTP) változásokat visszafordítani.

A tervezett vizsgálatok közvetlen célja nem új hatóanyagok kifejlesztése, hanem a neuroszteroidok terápiás lehetőségeinek igazolása. Véleményünk szerint a DHEA hatásmechanizmusának ismerete közelebb vihet a központi idegrendszer regenerativ folyamatainak megértéséhez.
Summary
The objective of the proposed project is to investigate the role of neurosteroids in age-related changes of the nervous system and to explore their potential therapeutic value for reversing age-associated cognitive deficits. This program requires a multidisciplinary approach that spans from morphological, electrophysiological and behavioral studies to genomic analysis.
The proposed approach is original in a sense that it is based on recent observations of age-related dysfunctions in the nervous system being reversible and that the aging brain retains its capacity for plasticity and regeneration. An important aspect of neuronal plasticity is synaptic remodeling that could be a significant component in the regenerative capacity of the central nervous system (CNS). Research carried out in our laboratory demonstrates that among other factors, gonadal steroids are able to induce morphological synaptic plasticity in different parts of the CNS; and we have shown that dehydroepiandrosterone (DHEA) has similar morphogenic effects in both hypothalamus and hippocampus.
In the present project, we propose to test the following hypotheses:
i.) Remodeling of hippocampal spine synapses is associated with changes in cognitive function and hence, it may play a role in age-related memory loss.
ii.) Age-related alterations in cognitive functions, as well as in hippocampal spine synapses and electrophysiology could be reversed by sex steroid administration.

At this phase of research, our aim is not to directly develop a new drug, but to focus on the potential therapeutic value of neurosteroids. According to our opinion, the detailed knowledge of the way DHEA acts in the brain may provide new insights into the regenerative mechanisms of the central nervous system.

A 75954 sz. pályázat keretén belül a hippokampális tüskeszinapszisok szerepét vizsgáltuk az időskori memóriadeficit és a depresszió állatmodelljén. Legfontosabb megállapításaink: A tüskeszinapszisok térfogati sűrűsége nemi különbséget mutat és korfüggő módon csökken, ez a csökkenés 24 hónapos nőstények esetében szignifikáns. Gonadektomizált patkányok hippocampusában a korábban már kimutatott szinapszis számcsökkenést 17ß-ösztradiol ill. testosteron kezeléssel ellensúlyozni lehet és ugyanígy hat a dehidroepiandroszteron (DHEA) is. Érdekes – és további vizsgálatokat igénylő - eredmény, hogy az idős (24 hónap) patkányoknál szabályos LTP-t tudtunk kiváltani annak ellenére, hogy az agyterület glutamaterg transzmisszójának állapotát jelző mezőpotenciálok a fiatal állatokénál 30-35%-al alacsonyabb értéket mutattak. Előzetes vizsgálatok szerint ebben a korcsoportban a hormonoknak és a DHEA-nak nincs hatása a LTP-re. Kimutattuk, hogy 17ß-ösztradiol a hippocampus területén több olyan gén expresszióját fokozza, melyek szerepet játszhatnak a szinaptikus átrendeződésben. Morfometriai mérésekkel igazoltuk, hogy a ”learned helplessness” depressziós állatmodellben a tünetek súlyossága negativ korrelációt mutat a tüskeszinapszisok számával. A hagyományos antidepresszáns desipraminhoz hasonlóan 17ß-ösztradiol-lal történő kezelés is kivédte a depressziós állapotot kísérő szinapszisszám csökkenést és a viselkedési változásokat.
Results in English
In the present project (No.: 75954) we have studied the role of hippocampal spine synapses in animal models of cognitive ageing and depression, We were focusing on the effect of steroid hormones and (dehydroepiandosterone) DHEA in synaptic remodeling. We demonstrated age dependent decrease in the volume density of spine synapses in the stratum radiatum of CA1 region, this change is significant in case of 24 month old females. The decrease of spine synapse density observed after gonadectomy can be reversed by 17ß-estradiol, testosterone and DHEA. In spite of the decreased spine synapse density and a 30-35% decrease of field potentials (measure of glutamatergic synaptic transmission) we could induce normal LTP in aged animals. We plan to further study the cellular mechanisms of this unexpected observation. Preliminary results did not show any effects of hormones or DHEA on LTP induction in this group. Our gene expression studies have shown that 17ß-estradiol upregulates several genes in the hippocampus, some of them may play a role in the synaptic remodeling. In the ”learned helplessness” depression model we found significant loss of hippocampal spine synapses and the unbiased stereology measurements showed negative correlation between the synapse numbers and the severity of behavioral symptoms. Similarly to the antidepressant drug desipramine, 17ß-estradiol treatment has also reversed the stress induced loss of hippocampal spine synapses.
